Microbes comprise the majority of extant organisms, yet much remains to be learned about the nature and driving forces of microbial diversification. Our understanding of how microorganisms adapt and evolve can be advanced by genome-wide documentation of the patterns of genetic exchange, particularly if analyses target coexisting members of natural communities. Here we use community genomic data sets to identify, with strain specificity, expressed proteins from the dominant member of a genomically uncharacterized, natural, acidophilic biofilm. Proteomics results reveal a genome shaped by recombination involving chromosomal regions of tens to hundreds of kilobases long that are derived from two closely related bacterial populations. Inter-population genetic exchange was confirmed by multilocus sequence typing of isolates and of uncultivated natural consortia. The findings suggest that exchange of large blocks of gene variants is crucial for the adaptation to specific ecological niches within the very acidic, metal-rich environment. Mass-spectrometry-based discrimination of expressed protein products that differ by as little as a single amino acid enables us to distinguish the behaviour of closely related coexisting organisms. This is important, given that microorganisms grouped together as a single species may have quite distinct roles in natural systems and their interactions might be key to ecosystem optimization. Because proteomic data simultaneously convey information about genome type and activity, strain-resolved community proteomics is an important complement to cultivation-independent genomic (metagenomic) analysis of microorganisms in the natural environment. 相似文献

Pitch size varies in official soccer matches and differently sized pitches are adopted for tactical purposes in small-sided training games. Since interactive team behaviour emerges under constraints, the authors evaluate the effect of pitch size (task) manipulations on interactive team behaviour in small-sided soccer games. Four 4-a-side (plus goalkeepers) small-sided games were played: a reference game (30×20 m), length manipulation (24×20 m), width manipulation (30×16 m), and a combination (24×16 m). Using position data (100Hz), three measures quantifying the teams’ interaction were calculated: longitudinal inter-team distance, lateral inter-team distance, and surface area difference. Means and standard deviations, correlations and coupling values were calculated. Running correlations were calculated over a 3-s window to evaluate interaction patterns. As expected, a shorter pitch results in smaller longitudinal inter-team distance, lateral inter-team distance decreased for narrow pitches, and smaller total playing area resulted in decreased surface area. Unanticipated, a crossover effect was present; length and width manipulations also triggered changes in lateral and longitudinal direction respectively. Inter-team distances and surface area difference differed significantly across conditions. Interaction patterns differed across conditions for all measures. So, highly tactically relevant, soccer teams seem to adapt their interactive behaviour according to pitch size in small-sided games. 相似文献

This article uses a recently proposed measure, the overall replacement ratio or ORR, to assess the extent to which migration alters intergenerational replacement within the United Kingdom. The UK as a whole can be seen to experience 'replacement migration' as immigration compensates for fertility below the replacement level. However, the article shows that the impact of migration differs radically in the different regions of the country. South East England experiences very substantial immigration from both the rest of the UK and overseas, far more than is needed for intergenerational replacement, whereas most of the rest of the UK sees little or no net immigration and the ORR remains below the replacement level. 相似文献

The 2000-based national population projections, carried out by the Government Actuary at the request of the Registrars General, show the population of the United Kingdom rising from 59.8 million in 2000 to nearly 65 million by 2025. Longer-term projections suggest the population will peak at nearly 66 million around 2040 and then gradually start to fall. The population will become gradually older with the median age expected to rise from 37.4 years in 2000 to 42.4 years by 2025. In 2000, there were 1.3 million (12 per cent) more children aged under 16, than people of state pensionable age. This paper describes work in progress which seeks to establish whether there are sufficient similarities between high-level teams operating in the business, government, and sporting environments to justify further research in these areas. It distinguishes strategic and tactical teams and identifies the characteristics of each within their separate disciplines. The paper concludes that there is indeed worthwhile research to be conducted in this field. Two systemic inquiries, based on soft systems methodology (SSM), into the potential for using community of practice (CoP) theory by an Australian-government created research and development corporation to change its knowledge management (KM) strategy, are reported. Key staff were engaged in the inquiry into how to build a third-order KM strategy based on CoPs; an exploration of key published work on CoPs yielded four SSM activity models—‘being a community practice system’; ‘doing the work of imagination sub-system’; ‘doing the work of alignment sub-system’ and ‘doing the work of engagement sub-system’. These models can be used as heuristics to aid the purposeful design of CoPs in other settings. SSM, enacted as a systemic inquiry, can be understood as a form of systemic action research, which was well equipped to deal with CoP theory and, when enacted participatively, can generate important systemic insights. The inquiry began the process of fostering an appreciation of third order KM but, on the evidence available, did not lead to on-going commitment to a CoPs-based KM strategy. Future research should acknowledge how the framing of research situations influences the research process, the importance of the design of practice change settings and the limited evidence for purposeful interventions leading to successful CoPs. 相似文献

The relationship between solar volatiles and those now in the Earth's atmosphere and mantle reservoirs provides insight into the processes controlling the acquisition of volatiles during planetary accretion and their subsequent evolution. Whereas the light noble gases (helium and neon) in the Earth's mantle preserve a solar-like isotopic composition, heavy noble gases (argon, krypton and xenon) have an isotopic composition very similar to that of the modern atmosphere, with radiogenic and (in the case of xenon) solar contributions. Mantle noble gases in a magmatic CO2 natural gas field have been previously corrected for shallow atmosphere/groundwater and crustal additions. Here we analyse new data from this field and show that the elemental composition of non-radiogenic heavy noble gases in the mantle is remarkably similar to that of sea water. We challenge the popular concept of a noble gas 'subduction barrier'--the convecting mantle noble gas isotopic and elemental composition is explained by subduction of sediment and seawater-dominated pore fluids. This accounts for approximately 100% of the non-radiogenic argon and krypton and 80% of the xenon. Approximately 50% of the convecting mantle water concentration can then be explained by this mechanism. Enhanced recycling of subducted material to the mantle plume source region then accounts for the lower ratio of radiogenic to non-radiogenic heavy noble gas isotopes and higher water content of plume-derived basalts. 相似文献

Advances in extreme-ultraviolet (EUV) and X-ray optics are providing powerful new capabilities in high-resolution imaging and trace-element analysis of microscopic specimens, and the potential for fabricating devices of smaller critical dimensions in next-generation integrated circuit lithography. However, achieving the highest resolution with such optics usually requires the illuminating EUV or X-ray beam to be highly monochromatic. It would therefore be highly desirable to have large-field-of-view, sub-100-nm resolution optics that are achromatic to a significant degree, allowing more light to be utilized from broader bandwidth sources such as laser-produced plasmas. Here we report an achromatic Fresnel optical system for EUV or X-ray radiation that combines a Fresnel zone plate with a refractive lens with opposite chromatic aberration. We use the large anomalous dispersion property of the refractive lens material near an absorption edge to make its fabrication practical. The resulting structure can deliver a resolution comparable to that of the Fresnel zone plates that have achieved the highest resolution (25 nm; ref. 3) in the entire electromagnetic spectrum, but with an improvement of two or more orders of magnitude in spectral bandwidth. 相似文献
