The endangered medicinal herb, Changium smyrnioides can only be found in deciduous forest gaps within the middle to northern subtropical broad-leaved evergreen forest zone of China. The considerable plasticity of its shoot and root structure helps it to capture light more effectively in winter and early spring, and to adapt to the soil moisture conditions in its narrow habitat. Another medicinal plant, Anthriscus sylvestris, is of similar economic importance but commonly distributed widely. In contrast to C. smyrnioides, it has low structural plasticity. It is also specialized to adapt to the moist and sunny environment, where habitat, such as the banks of creeks and rivers, is abundant.  相似文献

Faithful transmission of the genome requires that a protein complex called cohesin establishes and maintains the regulated linkage between replicated chromosomes before their segregation. Here we report the unforeseen participation of Caenorhabditis elegans TIM-1, a paralogue of the Drosophila clock protein TIMELESS, in the regulation of chromosome cohesion. Our biochemical experiments defined the C. elegans cohesin complex and revealed its physical association with TIM-1. Functional relevance of the interaction was demonstrated by aberrant mitotic chromosome behaviour, embryonic lethality and defective meiotic chromosome cohesion caused by the disruption of either TIM-1 or cohesin. TIM-1 depletion prevented the assembly of non-SMC (structural maintenance of chromosome) cohesin subunits onto meiotic chromosomes; however, unexpectedly, a partial cohesin complex composed of SMC components still loaded. Further disruption of cohesin activity in meiosis by the simultaneous depletion of TIM-1 and an SMC subunit decreased homologous chromosome pairing before synapsis, revealing a new role for cohesin in metazoans. On the basis of comparisons between TIMELESS homologues in worms, flies and mice, we propose that chromosome cohesion, rather than circadian clock regulation, is the ancient and conserved function for TIMELESS-like proteins.  相似文献

将人工全化学合成的尿激酶原(pro-urokinase)cDNA克隆到表达载体pET3d中,转化大肠杆菌BL21(DE3)LysS,经IPTG诱导,获得了占菌体总蛋白18%的高表达。经SDS-PAGE和Western-blot鉴定,表达产物主要为单链尿激酶,并且在菌体内基本以无活性的包涵体形式存在。经体外变复性,从IL培养基中可获得300000单位的活性尿激酶原。  相似文献

A new method based on mass fluxes and observed ozone profiles was developed to estimate crosstropopause ozone flux. Using this method, we estimated the cross-tropopause ozone flux in a stratospheric-tropospheric exchange event that occurred over East Asia in March 2001.The result revealed that the ozone flux across the tropopause in this event was an order of magnitude higher than the global and hemispheric average. Compared to the traditional method using a linear relationship between ozone mixing ratio and potential vorticity near the tropopause, the cross-tropopause ozone flux evaluated with ozonesonde data was somewhat higher, although the orders of the two values were the same.  相似文献

Summary Effects were examined of atropine, diazepam, pethidene, promethazine, scopolamine, omnopon and papaverine on basal and noradrenaline-stimulated lipolysis in rat isolated fat cells and on rat adipose tissue cyclic AMP phosphodiesterase activity. Papaverine at high concentration (1 mM) inhibited both basal and hormone-stimulated lipolysis, whereas diazepam enhanced basal lipolysis. At a clinical dose, omnopon increased both basal and noradrenaline-stimulated lipolysis. Adipose tissue cAMP phosphodiesterase activity was strongly inhibited by 1 mM diazepam, papaverine, promethazine and omnopon (280 g ml–1). Lack of enhancement of lipolysis by the established cAMP phosphodiesterase antagonist papaverine, is compartible with simultaneous inhibition also of adipose adenyl cyclase. Diazepam-stimulated lipolysis is compatible with its phosphodiesterase inhibitory activity. It is proposed that papaverine-containing omnopon may offer some survival advantages during surgical stress by facilitating a caloric supply.The authours are grateful to Dr D. C. 为研究有无对侧刺激声 (CAS)作用下 ,瞬态诱发耳声发射 (TEOAE)信号的变化特征 ,根据耳蜗的非线性特性 ,提出了将近似熵的分析方法引入到耳声发射信号的研究中 ,并将近似熵的分析方法从时间序列复杂性的度量中引入到频率序列复杂性的度量中。结果表明 :无 CAS作用下 ,TEOAE频域信号的近似熵在各频带处 ,蜗性病变耳的近似熵明显低于正常耳的近似熵 ,蜗后病变耳的近似熵与正常耳的近似熵无明显差别。在 CAS作用下 ,听力正常耳的近似熵下降 ,而感音神经性耳聋的近似熵上升 ,且近似熵上升的频带与耳蜗或蜗后神经患病处频带相关 .根据研究结果 ,提出了利用有无对侧刺激声作用下耳声发射信号近似熵的变化来诊断感音神经性耳聋的方法  相似文献
