Summary This paper reviews the influence of the geochemical environment on the epidemiology of human dental caries. The best documented association is that between water borne fluoride and reduced caries prevalence. The influence of fluoride was first reported during the early decades of this century in Colorado, USA, and led to the fluoridation of some public water supplies in several countries. In all cases, fluoridation has been followed by significant improvements in dental health and no adverse effects in general health. Other trace elements in food and water have now been linked with dental caries. Molybdenum has been associated with reduced caries prevalence whereas selenium and lead appear to have adverse effects. Cavity formation in teeth probably involves a localised dissolution of the enamel surface by the products of bacterial activity. It is possible that the incorporation of trace metals into the apatite microcrystals of enamel may alter their physical properties, especially solubility, and hence their susceptibility to degradation.  相似文献

Although often regarded as a network-former in conventional silicate glasses, Al(2)O(3) alone cannot be obtained as a bulk glass. Until now, glasses comprising continuously linked [AlO(x)] polyhedra have been prepared in only a few systems under very fast cooling conditions, which limits their dimensions to a few millimetres. Yet it is desirable to prepare bulk, or monolithic, alumina-rich glasses, with the prospect of superior mechanical, chemical and optical properties. Here we report a novel process for preparing very-high-alumina glasses and nanoscale glass-ceramics. Fully dense bulk articles in net shape are obtained through viscous sintering of glass microbeads. Additional heat treatment of the consolidated glasses leads to fully crystallized transparent glass-converted nanoceramics with a hardness similar to that of alumina. This method avoids the impracticably high applied pressures (more than 1 GPa) that have been required in most cases to prepare nanocrystalline ceramics by sintering, owing to the concurrent nature of densification and grain growth under pressureless conditions. The reported techniques can be extended to form glasses and nanoceramics in other oxide systems that do not include a conventional glass-forming component.  相似文献

The concentrations and locations of elements that strongly partition into the fluid phase in rocks provide essential constraints on geochemical and geodynamical processes in Earth's interior. A fundamental question remains, however, as to where these incompatible elements reside before formation of the fluid phase. Here we show that partitioning of calcium between the grain interiors and grain boundaries of olivine in natural and synthetic olivine-rich aggregates follows a thermodynamic model for equilibrium grain-boundary segregation. The model predicts that grain boundaries can be the primary storage sites for elements with large ionic radius--that is, incompatible elements in the Earth's mantle. This observation provides a mechanism for the selective extraction of these elements and gives a framework for interpreting geochemical signatures in mantle rocks.  相似文献

The objective of the present study was to determine levels of DNA fragmentation in blood leukocytes from guinea pigs by single-cell gel electrophoresis (comet assay) after exposure to the chemical warfare nerve agent (CWNA), soman, at doses ranging from 0.1 LD50 to 0.4 LD50, once per day for either 5 or 10 days. Post-exposure recovery periods ranged from 0 to 17 days. Leukocytes were imaged from each animal, and the images analyzed by computer. Data obtained for exposure to soman demonstrated significant increases in DNA fragmentation in circulating leukocytes in CWNA-treated guinea pigs compared with saline-injected control animals at all doses and time points examined. Notably, significantly increased DNA fragmentation was observed in leukocytes 17 days after cessation of soman exposure. Our findings demonstrate that leukocyte DNA fragmentation assays may provide a sensitive biomarker for low-dose CWNA exposure.Received 29 July 2003; accepted 14 August 2003  相似文献

Intraflagellar transport (IFT) proteins were first identified as essential factors for the growth and maintenance of flagella in the single-celled alga Chlamydomonas reinhardtii. In a screen for embryonic patterning mutations induced by ethylnitrosourea, here we identify two mouse mutants, wimple (wim) and flexo (fxo), that lack ventral neural cell types and show other phenotypes characteristic of defects in Sonic hedgehog signalling. Both mutations disrupt IFT proteins: the wim mutation is an allele of the previously uncharacterized mouse homologue of IFT172; and fxo is a new hypomorphic allele of polaris, the mouse homologue of IFT88. Genetic analysis shows that Wim, Polaris and the IFT motor protein Kif3a are required for Hedgehog signalling at a step downstream of Patched1 (the Hedgehog receptor) and upstream of direct targets of Hedgehog signalling. Our data show that IFT machinery has an essential and vertebrate-specific role in Hedgehog signal transduction.  相似文献

Extracellular Ca(2+) (Ca(2+)(o)) is required for various physiological and developmental processes in animals and plants. In response to varied Ca(2+)(o) levels, plants maintain relatively constant internal Ca(2+) content, suggesting a precise regulatory mechanism for Ca(2+) homeostasis. However, little is known about how plants monitor Ca(2+)(o) status and whether Ca(2+)(o)-sensing receptors exist. The effects of Ca(2+)(o) on guard cells in promoting stomatal closure by inducing increases in the concentration of cytosolic Ca(2+) ([Ca(2+)](i)) provide a clue to Ca(2+)(o) sensing. Here we have used a functional screening assay in mammalian cells to isolate an Arabidopsis complementary DNA clone encoding a Ca(2+)-sensing receptor, CAS. CAS is localized to the plasma membrane, exhibits low-affinity/high-capacity Ca(2+) binding, and mediates Ca(2+)(o)-induced [Ca(2+)](i) increases. CAS is expressed predominantly in the shoot, including guard cells. Repression of CAS disrupts Ca(2+)(o) signalling in guard cells, and impairs bolting (swift upward growth at the transition to seed production) in response to Ca(2+) deficiency, so we conclude that CAS may be a primary transducer of Ca(2+)(o) in plants.  相似文献

The origin of the ratites, large flightless birds from the Southern Hemisphere, along with their flighted sister taxa, the South American tinamous, is central to understanding the role of plate tectonics in the distributions of modern birds and mammals. Defining the dates of ratite divergences is also critical for determining the age of modern avian orders. To resolve the ratite phylogeny and provide biogeographical data to examine these issues, we have here determined the first complete mitochondrial genome sequences of any extinct taxa--two New Zealand moa genera--along with a 1,000-base-pair sequence from an extinct Madagascan elephant-bird. For comparative data, we also generated 12 kilobases of contiguous sequence from the kiwi, cassowary, emu and two tinamou genera. This large dataset allows statistically precise estimates of molecular divergence dates and these support a Late Cretaceous vicariant speciation of ratite taxa, followed by the subsequent dispersal of the kiwi to New Zealand. This first molecular view of the break-up of Gondwana provides a new temporal framework for speciation events within other Gondwanan biota and can be used to evaluate competing biogeographical hypotheses.  相似文献

Commensurate with the importance of rapidly and efficiently evaluating motivationally significant stimuli, humans are probably endowed with distinct faculties and maintain specialized neural structures to enhance their detection. Here we consider that a critical function of the human amygdala is to enhance the perception of stimuli that have emotional significance. Under conditions of limited attention for normal perceptual awareness-that is, the attentional blink-we show that healthy observers demonstrate robust benefits for the perception of verbal stimuli of aversive content compared with stimuli of neutral content. In contrast, a patient with bilateral amygdala damage has no enhanced perception for such aversive stimulus events. Examination of patients with either left or right amygdala resections shows that the enhanced perception of aversive words depends specifically on the left amygdala. All patients comprehend normally the affective meaning of the stimulus events, despite the lack of evidence for enhanced perceptual encoding of these events in patients with left amygdala lesions. Our results reveal a neural substrate for affective influences on perception, indicating that similar neural mechanisms may underlie the affective modulation of both recollective and perceptual experience.  相似文献
