The Wellcome Trust Case Control Consortium (WTCCC) primary genome-wide association (GWA) scan on seven diseases, including the multifactorial autoimmune disease type 1 diabetes (T1D), shows associations at P < 5 x 10(-7) between T1D and six chromosome regions: 12q24, 12q13, 16p13, 18p11, 12p13 and 4q27. Here, we attempted to validate these and six other top findings in 4,000 individuals with T1D, 5,000 controls and 2,997 family trios independent of the WTCCC study. Minimising the fabric loss during spreading can reduce the total production costs of garment manufacturing. Although marker planning is a major determinant of material utilisation, considerable savings on materials also will be made from the area of spreading loss. In the spreading process, it is general to have variance on the fabric yardage between each fabric roll, and the number of splice markings on each cutting lay is also varied. This paper aim to study how the fabric roll planning and the number of splice marks of a cutting lay will affect the material usage in the spreading process. By using a genetic algorithm approach, the amount of fabric to be saved by optimising the fabric roll sequence was derived by comparing the best arrangement and the worst arrangement of the fabric roll sequences of a cutting lay. The spliceosome is a dynamic macromolecular machine that assembles on pre-messenger RNA substrates and catalyses the excision of non-coding intervening sequences (introns). Four of the five major components of the spliceosome, U1, U2, U4 and U5 small nuclear ribonucleoproteins (snRNPs), contain seven Sm proteins (SmB/B', SmD1, SmD2, SmD3, SmE, SmF and SmG) in common. Following export of the U1, U2, U4 and U5 snRNAs to the cytoplasm, the seven Sm proteins, chaperoned by the survival of motor neurons (SMN) complex, assemble around a single-stranded, U-rich sequence called the Sm site in each small nuclear RNA (snRNA), to form the core domain of the respective snRNP particle. Core domain formation is a prerequisite for re-import into the nucleus, where these snRNPs mature via addition of their particle-specific proteins. Here we present a crystal structure of the U4 snRNP core domain at 3.6?? resolution, detailing how the Sm site heptad (AUUUUUG) binds inside the central hole of the heptameric ring of Sm proteins, interacting one-to-one with SmE-SmG-SmD3-SmB-SmD1-SmD2-SmF. An irregular backbone conformation of the Sm site sequence combined with the asymmetric structure of the heteromeric protein ring allows each base to interact in a distinct manner with four key residues at equivalent positions in the L3 and L5 loops of the Sm fold. A comparison of this structure with the U1 snRNP at 5.5?? resolution reveals snRNA-dependent structural changes outside the Sm fold, which may facilitate the binding of particle-specific proteins that are crucial to biogenesis of spliceosomal snRNPs. 相似文献

采用同样尺寸的钢梁和混凝土梁,通过中间绞结和底面粘贴FRP布而形成的组合梁,对其进行抗弯试验来研究钢筋混凝土梁弯曲或弯剪裂缝所导致的FRP剥离破坏.试验共设计了7组21根组合梁,通过2个对称的集中荷载进行抗弯试验,重点研究了组合梁破坏过程和形态、FRP布在加载过程中应变分布的规律、不同的裂缝开展程度对组合梁初裂荷载和极限承载能力的影响以及混凝土强度对FRP与混凝土粘结强度的影响.试验结果表明:组合梁的初裂荷载与FRP的长度没有关系,主要取决于裂缝的竖向位移;裂缝的竖向位移和FRP的长度对梁的极限荷载具有较大的影响,特别是在FRP锚固长度较小的情况下.针对梁中弯曲及弯剪裂缝引起的FRP剥离破坏进行了深入的试验研究,为工程结构加固提供了一定的参考依据. 相似文献

It is known that heterologous antiserum against rat kidney homogenate may induce congenital malformations when injected into pregnant rats during the period of organogenesis. Teratogenic rabbit antibodies against a purified rat renal tubular glycoprotein were isolated, labelled with 125I and injected into pregnant rats on the 10th day of gestation. Extracts of visceral yolk-sacs (VYS) and embryos were obtained 16 h later and chromatographed separately on a Sephacryl S-300 gel filtration column. The resultant chromatograms showed several radioactive peaks, one of which coincided with the eluate of intact rabbit immunoglobulins G (IgG). We interpret the result as an indication that some undigested intact teratogenic IgG were present in VYS and the embryo. 相似文献
