A simple method for reduction of non-specific fluorescence

Résumé Cette technique pour la réduction de la fluorescence non-spécifique utilise diverses méthodes communes en une série spéciale. Premièrement, les anticorps fluorescents sont adsorbés avec la poudre du foie. Suivant la centrifugation, une colonne de Séphadex-25 est utilisée pour séparer la fluorescéine libre. La méthode contraire donne une fluorescence non-spécifique qui ressemble à celle obtenue avec une seule adsorption ou Séphadex filtration.

Effect of prostaglandin E1 on cholesterol biosynthesis in rat liver

Riassunto La prostaglandina E₁ (PGE₁) ha un duplice effetto sulla sintesi epatica del colesterolo: a basse concentrazioni determina un incremento della incorporazione di acetato in colesterolo, mentre a concentrazini superiori a 50 nmoli essa determina una riduzione di questo paramentro.     

Absence of effects of dibutyryl cyclic guanosine 3',5'-monophosphate on release of alpha-amylase, 45Ca efflux, and protein synthesis in rat pancreas in vitro.

Dibutyryl cyclic GMP did not affect basal, or carbachol stimulated secretion of alpha-amylase from rat pancreas. The nucleotide did not have a significant effect on 45Ca release from the pancreas nor did it alter the response to carbachol. The dibutyryl analogue of cyclic GMP did not duplicate or alter the inhibitory effect of carbachol on 3H-leucine incorporation into pancreatic trichloroacetic acid-precipitable protein.     

Effect of PGE1 on lipogenesis in perfused rat liver

Summary In perfused livers of 24 hour-fasted rats, PGE₁ (prostaglandin E₁) infused continuously into the perfusate, was found to cause a 45% increase in the incorporation of 1-¹⁴C acetate into liver fatty acids. PGE₁ was found to have no effect, however, on the activity of the key lipogenic enzymes.     

Uptake of glucose-C14 and fructose-C14 by buffalo spermatozoa in extenders during cold storage

Summary Buffalo spermatozoa exhibit insignificant uptake of glucose-C¹⁴ and fructose-C¹⁴ when semen samples were preserved in cold for 96 h in 2 extenders. Incorporation of C¹⁴ in spermatozoa, TCA precipitable proteins and soluble sugar phosphates was either equal or less in semen samples preserved in cold. CAW appeared to be a better extender than SKMEY as revealed by sugar uptake in cold.Acknowledgment. We are indebted to Dr D. Sundaresan, Director, for his kind encouragement in this study.     

High resolution of heterochromatin of Drosophila melanogaster by distamycin A

A DNA-binding AT-specific antibiotic, distamycin A, was used as inhibitor of the condensation process of the heterochromatic regions in Drosophila melanogaster embryonic cells. By this treatment the structural organization of heterochromatin at interphase is preserved until metaphase. The different patterns observed are interpreted as chronological steps in the condensation process.     

Difference of the inhibitory action of verapamil on the positive inotropic effect of Ca2+ between spontaneously hypertensive and normotensive rat myocardium

Verapamil, a calcium entry blocker, had a greater inhibitory effect on the positive inotropic effect of excess Ca2+ in SHR than in NWR, suggesting that the cardiac responsiveness to verapamil was enhanced in SHR.     

Calcitonin-like peptide in the shrimpPalaemon serratus (Crustacea,Decapoda) during the intermolt cycle

Summary A molecule immunologically related to salmon calcitonin has been detected in the hemolymph of the shrimpPalaemon serratus. Its concentration varies inversely with the calcium level during the molt cycle; a maximum (14 ng/ml) is found in the post-molt stage and a minimum (0.5 ng/ml) during the premolt stage.
