The I kappa B kinase (IKK), consisting of the IKK1 and IKK2 catalytic subunits and the NEMO (also known as IKK gamma) regulatory subunit, phosphorylates I kappa B proteins, targeting them for degradation and thus inducing activation of NF-kappa B (reviewed in refs 1, 2). IKK2 and NEMO are necessary for NF-kappa B activation through pro-inflammatory signals. IKK1 seems to be dispensable for this function but controls epidermal differentiation independently of NF-kappa B. Previous studies suggested that NF-kappa B has a function in the growth regulation of epidermal keratinocytes. Mice lacking RelB or I kappa B alpha, as well as both mice and humans with heterozygous NEMO mutations, develop skin lesions. However, the function of NF-kappa B in the epidermis remains unclear. Here we used Cre/loxP-mediated gene targeting to investigate the function of IKK2 specifically in epidermal keratinocytes. IKK2 deficiency inhibits NF-kappa B activation, but does not lead to cell-autonomous hyperproliferation or impaired differentiation of keratinocytes. Mice with epidermis-specific deletion of IKK2 develop a severe inflammatory skin disease, which is caused by a tumour necrosis factor-mediated, alpha beta T-cell-independent inflammatory response that develops in the skin shortly after birth. Our results suggest that the critical function of IKK2-mediated NF-kappa B activity in epidermal keratinocytes is to regulate mechanisms that maintain the immune homeostasis of the skin.  相似文献

The cytoplasmically inherited bacterium Wolbachia pipientis is a widespread parasite of arthropods that manipulates the reproductive biology of its hosts, often to their detriment, in order to foster its own transmission through egg cytoplasm. Here we report that infection by Wolbachia restores fertility to Drosophila melanogaster mutant females prevented from making eggs by protein-coding lesions in Sex-lethal (Sxl), the master regulator of sex determination. Suppression of sterility by Wolbachia discriminates markedly among similar germline-specific Sxl alleles, and is not observed for mutations in other genes that produce similar 'tumorous ovary' phenotypes, including one that blocks Sxl germline expression. This allele and gene specificity indicates that suppression probably results from a specific interaction with Sxl protein, rather than from a bypass of the normal germline requirement for this developmental regulator or from an effect on Sxl expression. The Sxl-Wolbachia interaction provides a rare opportunity to explore host-parasite relationships at the molecular level in a model insect. Furthermore, demonstration that a parasite infection can counteract the deleterious effects of mutations in host genes illustrates how hosts might become dependent on parasites.  相似文献

Reciprocity, whether direct or indirect, is thought to be the key to establishing cooperation among non-relatives. But Riolo et al. have presented a model in which cooperation is instead based on similarity: agents donate only when their partner's 'tag' lies within a 'tolerance' range around their own. Here we point out that their model requires individuals with identical tags to cooperate with each other, and show that cooperation tends to collapse when individuals bearing identical tags are given the option of not donating. A candidate prostate cancer susceptibility gene at chromosome 17p   总被引:23,自引:0,他引:23  
It is difficult to identify genes that predispose to prostate cancer due to late age at diagnosis, presence of phenocopies within high-risk pedigrees and genetic complexity. A genome-wide scan of large, high-risk pedigrees from Utah has provided evidence for linkage to a locus on chromosome 17p. We carried out positional cloning and mutation screening within the refined interval, identifying a gene, ELAC2, harboring mutations (including a frameshift and a nonconservative missense change) that segregate with prostate cancer in two pedigrees. In addition, two common missense variants in the gene are associated with the occurrence of prostate cancer. ELAC2 is a member of an uncharacterized gene family predicted to encode a metal-dependent hydrolase domain that is conserved among eukaryotes, archaebacteria and eubacteria. The gene product bears amino acid sequence similarity to two better understood protein families, namely the PSO2 (SNM1) DNA interstrand crosslink repair proteins and the 73-kD subunit of mRNA 3' end cleavage and polyadenylation specificity factor (CPSF73).  相似文献

The mammal fauna of western South Dakota is imperfectly known; in particular there is a notable paucity of data from Indian reservations in the region. We collected mammals from January 2003 through May 2006 using a variety of standard methodologies, including trapping with snap-traps and Macabee traps, shooting, and salvaging road- and hunter-killed animals. Herein we provide 23 specimen-based county records representing 17 species of mammals. In addition, a photo record constitutes the first report of Dasypus novemcinctus from South Dakota and the northernmost record in the United States. The possible local extirpation of Marmota flaviventris in the southern Black Hills is discussed.  相似文献

Germination studies of the pollen grains of a Scrophulariaceae plant, Torenia asiatica Linn. have revealed that the maximum percentage of germination and longer tubes were attained in 15 percent sucrose solution. Among the chemicals tried, calcium and boron enhanced germination. Less germination occurred among grains treated with potassium or magnesium salts. Abnormalities such as branched tubes with or without vesicles were found in chemically treated pollen grains.    相似文献

The reliability of monitoring visual obstruction and estimating standing herbage with a modified Robel pole was examined for high-elevation meadows in sedimentary soils on the Bighorn National Forest, Wyoming. Our objectives were to (1) test a modified pole graduated with 1.27-cm (0.5-inch) bands for estimating standing herbage based on linear regression of visual obstruction readings (VORs) on standing herbage, (2) validate the derived regression, (3) provide sample size estimates, and (4) develop guidelines for monitoring mountain grasslands. Each transect had 20 visual obstruction stations spaced 10 meters apart with 4 visual obstruction readings at each station. At 4 stations, vegetation within a 0.25-m 2 area was clipped at ground level. VORs and clipped standing herbage were averaged by transect for analysis. Visual obstruction reliably predicted average standing herbage (dry weights) for mountain meadows ( r 2 = 0.81, s x¯ = 382 kg ⋅ ha –1 ). Standing herbage ranged from 387 kg ⋅ ha –1 to 3930 kg ⋅ ha –1 , with a mean of 1742 kg ⋅ ha –1 . A validation data set of 13 transects sampled across the range of variation in standing crop showed that 85% of transects fell within the 90% prediction limits. We recommend a minimum of 4 transects for monitoring key areas or small areas up to 259 ha (640 acres) when managers need to consider differences in VOR bands and address multiple objectives. Cluster analyses (ISODATA) applied to the pole readings resulted in 4 visual obstruction categories: short, short-intermediate, tall-intermediate, and tall. This tool provides a simple, reliable, and cost-effective (time-saving) alternative to clipping vegetation and obtaining dry weights for monitoring. New invasions, better field data, and novel spatial-modeling techniques often drive the need to revisit previous maps and models of invasive species. Such is the case with the at least 10 species of Tamarix, which are invading riparian systems in the western United States and expanding their range throughout North America. In 2006, we developed a National Tamarisk Map by using a compilation of presence and absence locations with remotely sensed data and statistical modeling techniques. Since the publication of that work, our database of Tamarix distributions has grown significantly. Using the updated database of species occurrence, new predictor variables, and the maximum entropy (Maxent) model, we have revised our potential Tamarix distribution map for the western United States. Distance-to-water was the strongest predictor in the model (58.1%), while mean temperature of the warmest quarter was the second best predictor (18.4%). Model validation, averaged from 25 model iterations, indicated that our analysis had strong predictive performance (AUC = 0.93) and that the extent of Tamarix distributions is much greater than previously thought. The southwestern United States had the greatest suitable habitat, and this result differed from the 2006 model. Our work highlights the utility of iterative modeling for invasive species habitat modeling as new information becomes available. Late Holocene bison bones eroding from the badlands topography of Cathedral Gorge State Park in southeastern Nevada have been identified as the remains of Bison and date between approximately 400 and 850 years old. The bones were originally thought to be turn-of-the-century cattle and then early bison, so park personnel had need to solve this interpretive problem. A multidisciplinary team sought information on archaeology, geomorphology, paleontology, and paleoenvironment to document the 1st established occurrence of bison in this part of Nevada and to develop a hypothesis explaining their demise.  相似文献
