RNA interference (RNAi) regulates gene expression by the cleavage of messenger RNA, by mRNA degradation and by preventing protein synthesis. These effects are mediated by a ribonucleoprotein complex known as RISC (RNA-induced silencing complex). We have previously identified four Drosophila components (short interfering RNAs, Argonaute 2 (ref. 2), VIG and FXR) of a RISC enzyme that degrades specific mRNAs in response to a double-stranded-RNA trigger. Here we show that Tudor-SN (tudor staphylococcal nuclease)--a protein containing five staphylococcal/micrococcal nuclease domains and a tudor domain--is a component of the RISC enzyme in Caenorhabditis elegans, Drosophila and mammals. Although Tudor-SN contains non-canonical active-site sequences, we show that purified Tudor-SN exhibits nuclease activity similar to that of other staphylococcal nucleases. Notably, both purified Tudor-SN and RISC are inhibited by a specific competitive inhibitor of micrococcal nuclease. Tudor-SN is the first RISC subunit to be identified that contains a recognizable nuclease domain, and could therefore contribute to the RNA degradation observed in RNAi.  相似文献

Understanding how larvae from extant hydrothermal vent fields colonize neighbouring regions of the mid-ocean ridge system remains a major challenge in oceanic research. Among the factors considered important in the recruitment of deep-sea larvae are metabolic lifespan, the connectivity of the seafloor topography, and the characteristics of the currents. Here we use current velocity measurements from Endeavour ridge to examine the role of topographically constrained circulation on larval transport along-ridge. We show that the dominant tidal and wind-generated currents in the region are strongly attenuated within the rift valley that splits the ridge crest, and that hydrothermal plumes rising from vent fields in the valley drive a steady near-bottom inflow within the valley. Extrapolation of these findings suggests that the suppression of oscillatory currents within rift valleys of mid-ocean ridges shields larvae from cross-axis dispersal into the inhospitable deep ocean. This effect, augmented by plume-driven circulation within rift valleys having active hydrothermal venting, helps retain larvae near their source. Larvae are then exported preferentially down-ridge during regional flow events that intermittently over-ride the currents within the valley.  相似文献

There is huge potential for genetic exchange to occur within the dense, diverse anaerobic microbial population inhabiting the gastrointestinal tract (GIT) of humans and animals. However, the incidence of conjugative transposons (CTns) and the antibiotic resistance genes they carry has not been well studied among this population. Since any incoming bacteria, including pathogens, can access this reservoir of genes, this oversight would appear to be an important one. Recent evidence has shown that anaerobic bacteria native to the rumen or hindgut harbour both novel antibiotic resistance genes and novel conjugative transposons. These CTns, and previously characterized CTns, can be transferred to a wide range of commensal bacteria under laboratory and in vivo conditions. The main evidence that gene transfer occurs widely in vivo between GIT bacteria, and between GIT bacteria and pathogenic bacteria, is that identical resistance genes are present in diverse bacterial species from different hosts.  相似文献

One of the most notable trends in mammalian evolution is the massive increase in size of the cerebral cortex, especially in primates. Humans with autosomal recessive primary microcephaly (MCPH) show a small but otherwise grossly normal cerebral cortex associated with mild to moderate mental retardation. Genes linked to this condition offer potential insights into the development and evolution of the cerebral cortex. Here we show that the most common cause of MCPH is homozygous mutation of ASPM, the human ortholog of the Drosophila melanogaster abnormal spindle gene (asp), which is essential for normal mitotic spindle function in embryonic neuroblasts. The mouse gene Aspm is expressed specifically in the primary sites of prenatal cerebral cortical neurogenesis. Notably, the predicted ASPM proteins encode systematically larger numbers of repeated 'IQ' domains between flies, mice and humans, with the predominant difference between Aspm and ASPM being a single large insertion coding for IQ domains. Our results and evolutionary considerations suggest that brain size is controlled in part through modulation of mitotic spindle activity in neuronal progenitor cells.  相似文献

We found that PPM1D, encoding a serine/threonine protein phosphatase, lies within an epicenter of the region at 17q23 that is amplified in breast cancer. We show that overexpression of this gene confers two oncogenic phenotypes on cells in culture: attenuation of apoptosis induced by serum starvation and transformation of primary cells in cooperation with RAS.  相似文献

The interaction between an imposed shear flow and an order--disorder transition underlies a broad range of phenomena. Under the influence of shear flow, a variety of soft matter is observed to spontaneously form bands characterized by different local order---for example, thermotropic liquid crystals subjected to shear flow exhibit rich phase behaviour. The stability of order under the influence of shear flow is also fundamental to understanding frictional wear and lubrication. Although there exists a well developed theoretical approach to the influence of shear flow on continuous transitions in fluid mixtures, little is known about the underlying principles governing non-equilibrium coexistence between phases of different symmetry. Here we show, using non-equilibrium molecular dynamics simulations of a system of spherical particles, that a stationary coexistence exists between a strained crystal and the shearing liquid, and that this coexistence cannot be accounted for by invoking a non-equilibrium analogue of the chemical potential. Instead of such thermodynamic arguments, we argue that a balancing of the crystal growth rate with the rate of surface erosion by the shearing melt can account for the observed coexistence.  相似文献

Population dynamics of Canada lynx (Lynx canadensis) have been of interest to ecologists for nearly sixty years. Two competing hypotheses concerning lynx population dynamics and large-scale spatial synchrony are currently debated. The first suggests that dispersal is substantial among lynx populations, and the second proposes that lynx at the periphery of their range exist in small, isolated patches that maintain cycle synchrony via correlation with extrinsic environmental factors. Resolving the nature of lynx population dynamics and dispersal is important both to ecological theory and to the conservation of threatened lynx populations: the lack of knowledge about connectivity between populations at the southern periphery of the lynx's geographic range delayed their legal listing in the United States. We test these competing hypotheses using microsatellite DNA markers and lynx samples from 17 collection sites in the core and periphery of the lynx's geographic range. Here we show high gene flow despite separation by distances greater than 3,100 km, supporting the dispersal hypothesis. We therefore suggest that management actions in the contiguous United States should focus on maintaining connectivity with the core of the lynx's geographic range.  相似文献
