Zusammenfassung Ergebnisse quantitativ histochemischer Untersuchungen am Gelenkknorpel von Meerschweinchen verschiedener Altersstufen werden diskutiert. Unmittelbar nach Abschluss des Längenwachstums sinkt die Enzymtätigkeit ab; im vorgeschrittenen Alter senkt sich die Tätigkeit einiger Enzyme weiter, während für andere wie UDPGLDH, 6-PGDH, MDH, -Glucosidase, Sulfatase und Kathepsin D eine Erhöhung der Aktivität beobachtet wurde.     

How probable is probable? A numerical translation of verbal probability expressions

The reported experiment took place in a professional forecasting organization accustomed to giving verbal probability assessments (‘likely’, ‘probable’, etc.). It attempts to highlight the communication problems caused by verbal probability expressions and to offer possible solutions that are compatible with the forecasters overall perspective on their jobs Experts in the organization were first asked to give a numerical translation to 30 different verbal probability expressions most of which were taken from the organization's own published political forecasts. In a second part of the experiment the experts were given 15 paragraphs selected from the organization's political publications each of which contained at least one verbal expression of probability. Subjects were again asked to give a numerical translation to each verbal probability expression The results indicate that (a) there is a high variability in the interpretation of verbal probability expressions and (b) the variability is even higher in context. Possible reasons for the context effect are discussed and practical implications are suggested.     

A change in the freshwater balance of the Atlantic Ocean over the past four decades

The oceans are a global reservoir and redistribution agent for several important constituents of the Earth's climate system, among them heat, fresh water and carbon dioxide. Whereas these constituents are actively exchanged with the atmosphere, salt is a component that is approximately conserved in the ocean. The distribution of salinity in the ocean is widely measured, and can therefore be used to diagnose rates of surface freshwater fluxes, freshwater transport and local ocean mixing--important components of climate dynamics. Here we present a comparison of salinities on a long transect (50 degrees S to 60 degrees N) through the western basins of the Atlantic Ocean between the 1950s and the 1990s. We find systematic freshening at both poleward ends contrasted with large increases of salinity pervading the upper water column at low latitudes. Our results extend a growing body of evidence indicating that shifts in the oceanic distribution of fresh and saline waters are occurring worldwide in ways that suggest links to global warming and possible changes in the hydrologic cycle of the Earth.     

The ELF4 gene controls circadian rhythms and flowering time in Arabidopsis thaliana

Many plants use day length as an environmental cue to ensure proper timing of the switch from vegetative to reproductive growth. Day-length sensing involves an interaction between the relative length of day and night, and endogenous rhythms that are controlled by the plant circadian clock. Thus, plants with defects in circadian regulation cannot properly regulate the timing of the floral transition. Here we describe the gene EARLY FLOWERING 4 (ELF4), which is involved in photoperiod perception and circadian regulation. ELF4 promotes clock accuracy and is required for sustained rhythms in the absence of daily light/dark cycles. elf4 mutants show attenuated expression of CIRCADIAN CLOCK ASSOCIATED 1 (CCA1), a gene that is thought to function as a central oscillator component. In addition, elf4 plants transiently show output rhythms with highly variable period lengths before becoming arrhythmic. Mutations in elf4 result in early flowering in non-inductive photoperiods, which is probably caused by elevated amounts of CONSTANS (CO), a gene that promotes floral induction.     

Chemical and photooxidation of thiothixene (Navane®): Structure of the thiothixene fluorophor

Summary The spontaneous photooxidation of thiothixene (Navane^®) to 2-(N,N-dimethyl-sulfonamido)-9-thioxanthone is described. The corresponding sulfoxide is demonstrated to be the fluorescent species obtained upon permanganate oxidation of thiothixene in the fluorometric assay procedure of Mjörndal and Oreland.Acknowledgments. Mutagenic insertion and chromosome engineering resource (MICER)

Embryonic stem cell technology revolutionized biology by providing a means to assess mammalian gene function in vivo. Although it is now routine to generate mice from embryonic stem cells, one of the principal methods used to create mutations, gene targeting, is a cumbersome process. Here we describe the indexing of 93,960 ready-made insertional targeting vectors from two libraries. 5,925 of these vectors can be used directly to inactivate genes with an average targeting efficiency of 28%. Combinations of vectors from the two libraries can be used to disrupt both alleles of a gene or engineer larger genomic changes such as deletions, duplications, translocations or inversions. These indexed vectors constitute a public resource (Mutagenic Insertion and Chromosome Engineering Resource; MICER) for high-throughput, targeted manipulation of the mouse genome.     

Vernalization requires epigenetic silencing of FLC by histone methylation

To ensure flowering in favourable conditions, many plants flower only after an extended period of cold, namely winter. In Arabidopsis, the acceleration of flowering by prolonged cold, a process called vernalization, involves downregulation of the protein FLC, which would otherwise prevent flowering. This lowered FLC expression is maintained through subsequent development by the activity of VERNALIZATION (VRN) genes. VRN1 encodes a DNA-binding protein whereas VRN2 encodes a homologue of one of the Polycomb group proteins, which maintain the silencing of genes during animal development. Here we show that vernalization causes changes in histone methylation in discrete domains within the FLC locus, increasing dimethylation of lysines 9 and 27 on histone H3. Such modifications identify silenced chromatin states in Drosophila and human cells. Dimethylation of H3 K27 was lost only in vrn2 mutants, but dimethylation of H3 K9 was absent from both vrn1 and vrn2, consistent with VRN1 functioning downstream of VRN2. The epigenetic memory of winter is thus mediated by a 'histone code' that specifies a silent chromatin state conserved between animals and plants.     

High-resolution record of Northern Hemisphere climate extending into the last interglacial period

Two deep ice cores from central Greenland, drilled in the 1990s, have played a key role in climate reconstructions of the Northern Hemisphere, but the oldest sections of the cores were disturbed in chronology owing to ice folding near the bedrock. Here we present an undisturbed climate record from a North Greenland ice core, which extends back to 123,000 years before the present, within the last interglacial period. The oxygen isotopes in the ice imply that climate was stable during the last interglacial period, with temperatures 5 degrees C warmer than today. We find unexpectedly large temperature differences between our new record from northern Greenland and the undisturbed sections of the cores from central Greenland, suggesting that the extent of ice in the Northern Hemisphere modulated the latitudinal temperature gradients in Greenland. This record shows a slow decline in temperatures that marked the initiation of the last glacial period. Our record reveals a hitherto unrecognized warm period initiated by an abrupt climate warming about 115,000 years ago, before glacial conditions were fully developed. This event does not appear to have an immediate Antarctic counterpart, suggesting that the climate see-saw between the hemispheres (which dominated the last glacial period) was not operating at this time.
