Protein quality control is vital for all living cells and sophisticated molecular mechanisms have evolved to prevent the excessive accumulation of unfolded proteins. High-temperature requirement A (HtrA) proteases have been identified as important ATP-independent quality-control factors in most species. HtrA proteins harbor a serine-protease domain and at least one peptide-binding PDZ domain to ensure efficient removal of misfolded or damaged proteins. One distinctive property of HtrAs is their ability to assemble into complex oligomers. Whereas all examined HtrAs are capable of forming pyramidal 3-mers, higher-order complexes consisting of up to 24 molecules have been reported. Tight control of chaperone and protease function is of pivotal importance in preventing deleterious HtrA-protease activity. In recent years, structural biology provided detailed insights into the molecular basis of the regulatory mechanisms, which include unique intramolecular allosteric signaling cascades and the dynamic switching of oligomeric states of HtrA proteins. Based on these results, functional models for many family members have been developed. The HtrA protein family represents a remarkable example of how structural and functional diversity is attained from the assembly of simple molecular building blocks.  相似文献

The formation of low-mass stars like our Sun can be explained by the gravitational collapse of a molecular cloud fragment into a protostellar core and the subsequent accretion of gas and dust from the surrounding interstellar medium. Theoretical considerations suggest that the radiation pressure from the protostar on the in-falling material may prevent the formation of stars above ten solar masses through this mechanism, although some calculations have claimed that stars up to 40 solar masses can in principle be formed via accretion through a disk. Given this uncertainty and the fact that most massive stars are born in dense clusters, it was suggested that high-mass stars are the result of the runaway merging of intermediate-mass stars. Here we report observations that clearly show a massive star being born from a large rotating accretion disk. The protostar has already assembled about 20 solar masses, and the accretion process is still going on. The gas reservoir of the circumstellar disk contains at least 100 solar masses of additional gas, providing sufficient fuel for substantial further growth of the forming star.  相似文献

Continental-scale rivers with a sandy bed sequester a significant proportion of their sediment load in flood plains. The spatial extent and depths of such deposits have been described, and flood-plain accumulation has been determined at decadal timescales, but it has not been possible to identify discrete events or to resolve deposition on near-annual timescales. Here we analyse (210)Pb activity profiles from sediment cores taken in the pristine Beni and Mamore river basins, which together comprise 720,000 km2 of the Amazon basin, to investigate sediment accumulation patterns in the Andean-Amazonian foreland. We find that in most locations, sediment stratigraphy is dominated by discrete packages of sediments of uniform age, which are typically 20-80 cm thick, with system-wide recurrence intervals of about 8 yr, indicating relatively rare episodic deposition events. Ocean temperature and stream flow records link these episodic events to rapidly rising floods associated with La Ni?a events, which debouch extraordinary volumes of sediments from the Andes. We conclude that transient processes driven by the El Ni?o/Southern Oscillation cycle control the formation of the Bolivian flood plains and modulate downstream delivery of sediments as well as associated carbon, nutrients and pollutants to the Amazon main stem.  相似文献
