Many representations of sensory stimuli in the neocortex are arranged as topographic maps. These cortical maps are not fixed, but show experience-dependent plasticity. For instance, sensory deprivation causes the cortical area representing the deprived sensory input to shrink, and neighbouring spared representations to enlarge, in somatosensory, auditory or visual cortex. In adolescent and adult animals, changes in cortical maps are most noticeable in the supragranular layers at the junction of deprived and spared cortex. However, the cellular mechanisms of this experience-dependent plasticity are unclear. Long-term potentiation and depression have been implicated, but have not been proven to be necessary or sufficient for cortical map reorganization. Short-term synaptic dynamics have not been considered. We developed a brain slice preparation involving rat whisker barrel cortex in vitro. Here we report that sensory deprivation alters short-term synaptic dynamics in both vertical and horizontal excitatory pathways within the supragranular cortex. Moreover, modifications of horizontal pathways amplify changes in the vertical inputs. Our findings help to explain the functional cortical reorganization that follows persistent changes of sensory experience. 相似文献

The malaria parasite Plasmodium falciparum is one of the most successful human pathogens. Specific virulence factors remain poorly defined, although the adhesion of infected erythrocytes to the venular endothelium has been associated with some of the syndromes of severe disease. Immune responses cannot prevent the development of symptomatic infections throughout life, and clinical immunity to the disease develops only slowly during childhood. An understanding of the obstacles to the development of protective immunity is crucial for developing rational approaches to prevent the disease. Here we show that intact malaria-infected erythrocytes adhere to dendritic cells, inhibit the maturation of dendritic cells and subsequently reduce their capacity to stimulate T cells. These data demonstrate both a novel mechanism by which malaria parasites induce immune dysregulation and a functional role beyond endothelial adhesion for the adhesive phenotypes expressed at the surface of infected erythrocytes. 相似文献

Plasmodium falciparum infected erythrocytes containing mature trophozoites and schizonts sequester along venular endothelium and are not in the peripheral circulation of patients with malaria. Knobs appear on infected erythrocytes and are the points of attachment to endothelium. Sequestration may protect the parasite from splenic destruction and may play a role in the pathogenesis of cerebral malaria. Correlates of sequestration have been developed in vitro using cultured human endothelium and an amelanotic melanoma cell line. Knobless strains (K-) of P. falciparum fail to sequester in vivo and to bind to cells in vitro. We now present evidence that the receptor for cytoadherence is the glycoprotein, thrombospondin. Aotus monkey or human erythrocytes containing knobby (K+) but not Aotus erythrocytes containing knobless strains of P. falciparum bind to immobilized thrombospondin. Neither binds to the adhesive proteins laminin, fibronectin, factor VIII/von Willebrand factor or vitronectin. Both soluble thrombospondin and anti-thrombospondin antibodies inhibit binding of parasitized Aotus erythrocytes to immobilize thrombospondin and to melanoma cells which secrete thrombospondin. 相似文献

In 1994 ground fire ignited in forests of Douglas-fir, Pseudotsuga menziesii var. glauca (Mirb.) Franco, on Beaver Mountain, Utah. The Douglas-fir beetle, Dendroctonus pseudotsugae Hopkins, attacked a range of moderately fire-injured host conifers in 1995. Logistic regression models run for 1995 data illustrated that 1 year after the fire event the Douglas-fir beetle selected and attacked large-diameter Douglas-fir with 60%-80% bole char, 60%-80% crown volume scorch, and 50%-70% probability of mortality due to fire. In 1996 beetle preference shifted to smallerdiameter trees with lighter fire injury, because most large, fire-damaged conifers were colonized by beetles in 1995. Although beetle populations did not reach outbreak proportions outside the fire boundary, host selection shifted to green trees in 1997 along the burn perimeter. Log linear analysis indicated that increased brood production was conditioned by increased diameter and moderate fire damage to the trees. 相似文献

Accurate time- and cost-efficient mapping is central to successful rangeland invasive plant management. In this study sampling together with Inverse Distance Weighted (IDW) interpolation modeling was tested as a mapping alternative to expensive full-coverage delineation survey mapping methods. Our objective was to examine accuracies of presence/absence maps generated from 18 sampling strategies (3 sampling methods × 6 sample densities) using IDW. Invasive plant field survey maps with known accuracies were used to generate samples and to test interpolation results at 2 sites. Site 1 was approximately 6.0 km 2 , dominated by Russian knapweed ( Acroptilon repens L.). Site 2, an upland area of approximately 13.5 km 2 , was dominated by spotted knapweed ( Centaurea maculosa Lam). Sampling method × sample density combinations were gathered from field survey infestation maps using repeated computer-based sampling methods. IDW modeling was applied to each of the sample data sets. Accuracies of the IDW interpolation results were calculated by re-referencing field survey maps. We determined that sampling at density of 0.25% (about 1 point per ha) using a systematic sampling method was the preferred sampling strategy for both sites. This combination of sampling density and method yielded 85% accurate presence/absence maps. We conclude that sampling combined with IDW interpolation modeling can generate accurate invasive plant maps and is a potential alternative to current delineation survey methods. 相似文献
