Pre-messenger-RNA maturation in nematodes and in several other lower eukaryotic phyla involves spliced leader (SL) addition trans-splicing. In this unusual RNA processing reaction, a short common 5' exon, the SL, is affixed to the 5'-most exon of multiple pre-mRNAs. The nematode SL is derived from a trans-splicing-specific approximately 100-nucleotide RNA (SL RNA) that bears striking similarities to the cis-spliceosomal U small nuclear RNAs U1, U2, U4 and U5 (refs 3, 4); for example, the SL RNA functions only if it is assembled into an Sm small nuclear ribonucleoprotein (snRNP). Here we have purified and characterized the SL RNP and show that it contains two proteins (relative molecular masses 175,000 and 30,000 (M(r) 175K and 30K)) in addition to core Sm proteins. Immunodepletion and reconstitution with recombinant proteins demonstrates that both proteins are essential for SL trans-splicing; however, neither protein is required either for conventional cis-splicing or for bimolecular (trans-) splicing of fragmented cis constructs. The M(r) 175K and 30K SL RNP proteins are the first factors identified that are involved uniquely in SL trans-splicing. Several lines of evidence indicate that the SL RNP proteins function by participating in a trans-splicing specific network of protein protein interactions analogous to the U1 snRNP SF1/BBP U2AF complex that comprises the cross-intron bridge in cis-splicing. 相似文献

A series of removal experiments were performed on Dipodomys merriami, D. microps, and Perognathus longimembris to test for the importance of competition for food and microhabitats in a heteromyid community in the Great Basin Desert. Each of these species was removed singly to determine the short-term effects on the microhabitat preferences of the remaining species. We correctly predicted, based on differences in diet, that the removal of D. microps (a foliovore) would have no effect on D. merriami or P. longimembris (granivores). Using the dominance hierarchy theory, we correctly predicted that removal of a larger heteromyid, D. merriami, would have an effect on the microhabitat use of the smaller P. longimembris, but not vice versa. While our results offer strong evidence of competition for food and microhabitats, the short-term reactions were weak compared to the long-term reactions found in other studies of heteromyids. 相似文献

The human pan-leukocyte antigen Leu-8 has attracted wide interest because its presence or absence identifies suppressor-inducer and helper-inducer CD4+ T-lymphocyte subsets respectively. We report here that Leu-8 is the human homologue of the mouse Mel-14 homing receptor, a molecule that promotes the initial adhesion of blood-borne lymphocytes to the specialized post-capillary endothelium of peripheral lymph nodes. We also show that Leu-8 can adopt both conventional and phospholipid anchored forms, a finding that may have relevance in the context of antigen shedding following activation or homing. The assignment of lymphocytes to different functional classes based on lymph node homing potential may represent a more general association between lymphocyte function and tissue distribution. 相似文献

Interaction with antigen-presenting accessory cells is thought to be an important step in B-cell activation, and the B-cell receptor CD22, which is coordinately expressed with surface immunoglobulin, has been proposed to participate in the antigen response. Here we show that CD22 has a structure closely related to myelin-associated glycoprotein (MAG, a neuronal adhesion protein), and mediates monocyte and erythrocyte adhesion. Like CD2, the T-cell erythrocyte receptor, CD22 may facilitate antigen recognition by promoting antigen-nonspecific contacts with accessory cells. 相似文献

Summary Treatment of radioactively labeled guinea-pig skin soluble collagen or calf skin collagen with the flavonoid (+)-catechin makes the collagen resistant to the action of mammalian collagenase but not to the action of bacterial collagenase. Complete resistance to the action of the mammalian enzyme may be achieved by incubating 0.6 mg of collagen (dry weight) with 0.1 mM (+)-catechin, followed by dialysis to remove the unbound flavonoid. Since incubation of the mammalian enzyme with (+)-catechin does not inhibit its activity, it is postulated that (+)-catechin binds tightly to collagen and modifies its structure sufficiently to make it resistant to enzyme degradation.Acknowledgments.
