Hot-spot residue in small heat-shock protein 22 causes distal motor neuropathy   总被引:19,自引:0,他引:19  
Distal hereditary motor neuropathies are pure motor disorders of the peripheral nervous system resulting in severe atrophy and wasting of distal limb muscles. In two pedigrees with distal hereditary motor neuropathy type II linked to chromosome 12q24.3, we identified the same mutation (K141N) in small heat-shock 22-kDa protein 8 (encoded by HSPB8; also called HSP22). We found a second mutation (K141E) in two smaller families. Both mutations target the same amino acid, which is essential to the structural and functional integrity of the small heat-shock protein alphaA-crystallin. This positively charged residue, when mutated in other small heat-shock proteins, results in various human disorders. Coimmunoprecipitation experiments showed greater binding of both HSPB8 mutants to the interacting partner HSPB1. Expression of mutant HSPB8 in cultured cells promoted formation of intracellular aggregates. Our findings provide further evidence that mutations in heat-shock proteins have an important role in neurodegenerative disorders.  相似文献

Numerous phenological studies on spiders belonging to the families Oxyopidae, Thomisidae, Lycosidae, Selenopidae (e.g. Selenops cocheleti) and Salticidae (e.g. Psecas chapoda and Psecas viridipurpureus) have been conducted in the neotropical region. However, studies that simultaneously compare population dynamics and age structure in populations of sympatric species are limited, especially in Oxyopidae. The population dynamics and phenology of two congeneric lynx spider (Peucetia rubrolineata and Peucetia flava) were examined in southeastern Brazil. Several characteristics of the age structure were compared between the two spider species. The variation in the total abundance of individuals and age structure and their relationship with climatic variables were similar between the two spiders. Adults of these spiders were present mainly in spring, indicating an annual reproductive cycle and a ‘stenochronous spring’ phenological pattern. The recruitment of spiders occurred in summer for P. rubrolineata and P. flava, followed by successive phenological peaks at all stages of development. Rainfall and temperature had a positive correlation to population flux in the two spider species studied. Despite the significant climatic effects observed in the Serra do Japi, the phenological pattern of the population in this study was not always repeated in other species of spiders (e.g. Selenops cocheleti).  相似文献

In this paper we consider a novel procedure to forecasting the US zero coupon bond yields for a continuum of maturities by using the methodology of nonparametric functional data analysis (NP‐FDA). We interpret the US yields as curves since the term structure of interest rates defines a relation between the yield of a bond and its maturity. Within the NP‐FDA approach, each curve is viewed as a functional random variable and the dynamics present in the sample are modeled without imposing any parametric structure. In order to evaluate forecast the performance of the proposed estimator, we consider forecast horizons h = 1,3,6,12… months and the results are compared with widely known benchmark models. Our estimates with NP‐FDA present predictive performance superior to its competitors in many situations considered, especially for short‐term maturities. Bats (Chiroptera), one of the most diverse groups in terms of taxonomy, morphology and ecology, are known for their nocturnal behaviour of flight and feeding. Although there is no consensus on the evolution of nocturnality in bats, many authors mention risk of predation, overheating, competition and mobbing by non-competitor species as arguments to justify nocturnal instead of daytime flight in bats. Herein we describe the first records of three genera of phyllostomid bats flying, foraging and drinking water during daytime in the Brazilian Amazon. All taxa were recorded drinking water, and some Phyllostomus sp. individuals were recorded foraging on termites, alongside birds. Risk of dehydration and overheating in roosts, as well as low competition in daytime, may explain the emergence of phyllostomid bats before sunset.  相似文献

We examine trust at the theory level as an engine driving sustainable change. We propose a new measurable dimension of the trust construct: reciprocity. Cooperative reciprocal relationships are its building blocks, enabling stakeholders to ‘try out’ interactions with less risk, calibrating level of effort, time and emotional investment before continuing further. Reciprocity includes three measurable elements: common goals furthering equitable and healthier communities, mutual self-interests and gratitude/indebtedness. We utilize a process model showing how these building blocks interrelate within a health systems context to jump-start and sustain reciprocal relationships. Process models are valuable in addressing internal dynamics of human relationships in complex systems as a complement to factor- or variance-based models. We applied theory by longitudinally tracking community health volunteers, examining reciprocity in close-to-the-community health systems. These methods and application of theory have relevance to environments where institutional structures are open to multistakeholder-driven innovation.  相似文献

The persistent prevalence of childhood overweight and obesity raises significant concerns about the impact on health, society and the economy. Responding to a target announced in September 2015 by the New South Wales (Australia) Premier to reduce childhood overweight and obesity by five percentage points by 2025, a system dynamics model was developed to support Government and stakeholders responsible for meeting the target. A participatory model building process, drawing cross‐sectorial expertise, was undertaken to estimate the individual and combined impact of interventions on meeting the target. The model demonstrated that it is theoretically possible to meet the target by implementing a comprehensive combination of policies and programmes. When limited to existing and enhanced population health interventions, the modelled result did not reach the target. The project provides an example of how participatory simulation modelling can combine a broad range of interventions together into likely scenarios and usefully inform government decision‐making. © 2018 The authors.
