Spinocerebellar ataxia type 1 (SCA1) is a dominantly inherited neurodegenerative disease caused by expansion of a glutamine tract in ataxin-1 (ATXN1). SCA1 pathogenesis studies support a model in which the expanded glutamine tract causes toxicity by modulating the normal activities of ATXN1. To explore native interactions that modify the toxicity of ATXN1, we generated a targeted duplication of the mouse ataxin-1-like (Atxn1l, also known as Boat) locus, a highly conserved paralog of SCA1, and tested the role of this protein in SCA1 pathology. Using a knock-in mouse model of SCA1 that recapitulates the selective neurodegeneration seen in affected individuals, we found that elevated Atxn1l levels suppress neuropathology by displacing mutant Atxn1 from its native complex with Capicua (CIC). Our results provide genetic evidence that the selective neuropathology of SCA1 arises from modulation of a core functional activity of ATXN1, and they underscore the importance of studying the paralogs of genes mutated in neurodegenerative diseases to gain insight into mechanisms of pathogenesis.  相似文献

香港是一个市区人口相对集中的城市 ,公路和公共交通运输网的优化非常重要 ,随着经济的发展 ,人口的增加 ,对公共交通设施的需求越来越大 ,每年香港政府都要投资三亿港币建设新的公共交通设施 ,满足日益增长的需求 .这些新的交通设施要从许多公路和公共交通的规划项目中选择 ,这个问题数学上归结为一个 0 -1规划问题 .本文用遗传算法对这一 0 -1规划问题进行了求解 ,这是第一次将遗传算法应用于实际的网络设计问题 ,这一模型将有助于香港政府进行交通规划 ,香港 2 0 0 6年规划数据被用于进行实例研究.  相似文献

In this paper, we consider the price trend model in which it is assumed that the time series of a security's prices contain a stochastic trend component which remains constant on each of a sequence of time intervals, with each interval having random duration. A quasi‐maximum likelihood method is used to estimate the model parameters. Optimal one‐step‐ahead forecasts of returns are derived. The trading rule based on these forecasts is constructed and is found to bear similarity to a popular trading rule based on moving averages. When applying the methods to forecast the returns of the Hang Seng Index Futures in Hong Kong, we find that the performance of the newly developed trading rule is satisfactory. The most up‐to‐date annual average daily traffic (AADT) is always required for transport model development and calibration. However, the current‐year AADT data are not always available. The short‐term traffic flow forecasting models can be used to predict the traffic flows for the current year. In this paper, two non‐parametric models, non‐parametric regression (NPR) and Gaussian maximum likelihood (GML), are chosen for short‐term traffic forecasting based on historical data collected for the annual traffic census (ATC) in Hong Kong. These models are adapted as they are more flexible and efficient in forecasting the daily vehicular flows in the Hong Kong ATC core stations (in total of 87 stations). The daily vehicular flows predicted by these models are then used to calculate the AADT of the current year, 1999. The overall prediction and comparison results show that the NPR model produces better forecasts than the GML model using the ATC data in Hong Kong. Copyright © 2006 John Wiley _ Sons, Ltd.  相似文献

显著性检测在图像分割、物体检测、视觉效果评估等领域有非常广泛的用途。为了增强这种图像预处理技术的实用性,同时对人眼视觉系统的机理进行研究和模拟,该文提出了一种基于鲁棒主成分分析和多个色彩通道的显著性检测方法。将图像的多个色彩空间重构成每一列都线性相关的矩阵,然后将显著性区域看作是矩阵的稀疏成分,将背景信息看作是低秩成分。经过对新矩阵进行鲁棒主成分分析后就可以将显著性区域提取出来。最后加入人眼视觉系统中的显著性先验和中心先验机理,使得提出的模型更加合理。实验结果表明:该方法在量化指标和视觉效果方面超过了当前很多先进的方法,验证了这种假设的合理性。  相似文献

Copper nanorods have been synthe-sized in mesoporous SBA-15 by a low-temperature metal organic chemical vapor deposition (MOCVD) employing copper (II) acetylacetonate, Cu(acac)2, and hydrogen as a precursor and reactant gas, re- spectively. The hydrogen plays an important role in chemical reduction of oganometallic precursor which enhances mass transfer in the interior of the SBA-15 porous substrate. Such copper nanostructures are of great potentials in the semiconductor due to their unusual optical, magnetic and electronic properties. In addition, it has been found that chemically modi- fying the substrate surface by carbon deposition is crucial to such synthesis of copper nanostructures in the interior of the SBA-15, which is able to change the surface properties of SBA-15 from hydrophilic to hydrophobic to promote the adsorption of organic cupric precursor. It has also been found that the copper nanoparticles deposited on the external sur- face are almost eliminated and the copper nanorods are more distinct while the product was treated with ammonia. This approach could be achieved under a mild condition: a low temperature (400℃) and vac-uum (2 kPa) which is extremely milder than the con- ventional method. It actually sounds as a foundation which is the first time to synthesize a copper nanorod at a mild condition of a low reaction temperature and pressure.  相似文献
