The International Human Genome Sequencing Consortium (IHGSC) recently completed a sequence of the human genome. As part of this project, we have focused on chromosome 8. Although some chromosomes exhibit extreme characteristics in terms of length, gene content, repeat content and fraction segmentally duplicated, chromosome 8 is distinctly typical in character, being very close to the genome median in each of these aspects. This work describes a finished sequence and gene catalogue for the chromosome, which represents just over 5% of the euchromatic human genome. A unique feature of the chromosome is a vast region of approximately 15 megabases on distal 8p that appears to have a strikingly high mutation rate, which has accelerated in the hominids relative to other sequenced mammals. This fast-evolving region contains a number of genes related to innate immunity and the nervous system, including loci that appear to be under positive selection--these include the major defensin (DEF) gene cluster and MCPH1, a gene that may have contributed to the evolution of expanded brain size in the great apes. The data from chromosome 8 should allow a better understanding of both normal and disease biology and genome evolution.  相似文献

The kinetic, morphological, crystallographic, and magnetic characteristics of thermally induced martensites in Fe-13.4wt% Mn-5.2wt% Mo alloy were investigated by scanning electron microscopy (SEM), transmission electron microscopy (TEM), and Mössbauer spectroscopy. The experimental results reveal that two types of thermal-induced martensites, ? (hcp) and α′ (bcc) martensites, are formed in the as-quenched condition, and these transformations have athermal characters. Mo addition to the Fe-Mn alloy does not change the coexistence of ? and α′ martensites with the Mn content between 10wt% and 15wt%. Besides, Mössbauer spectra reveal a paramagnetic character with a singlet for the γ (fcc) austenite and ? martensite phases and a ferromagnetic character with a broad sextet for the α′ martensite phase. The volume fraction of α′ martensite forming in the quenched alloy is much more than that of the ? martensite.  相似文献

Previous studies suggest that neuronal activity may guide the development of synaptic connections in the central nervous system through mechanisms involving glutamate receptors and GTPase-dependent modulation of the actin cytoskeleton. Here we demonstrate by in vivo time-lapse imaging of optic tectal cells in Xenopus laevis tadpoles that enhanced visual activity driven by a light stimulus promotes dendritic arbor growth. The stimulus-induced dendritic arbor growth requires glutamate-receptor-mediated synaptic transmission, decreased RhoA activity and increased Rac and Cdc42 activity. The results delineate a role for Rho GTPases in the structural plasticity driven by visual stimulation in vivo.  相似文献

Alpha-neurexins couple Ca2+ channels to synaptic vesicle exocytosis   总被引:1,自引:0,他引:1  
Synapses are specialized intercellular junctions in which cell adhesion molecules connect the presynaptic machinery for neurotransmitter release to the postsynaptic machinery for receptor signalling. Neurotransmitter release requires the presynaptic co-assembly of Ca2+ channels with the secretory apparatus, but little is known about how synaptic components are organized. Alpha-neurexins, a family of >1,000 presynaptic cell-surface proteins encoded by three genes, link the pre- and postsynaptic compartments of synapses by binding extracellularly to postsynaptic cell adhesion molecules and intracellularly to presynaptic PDZ domain proteins. Using triple-knockout mice, we show that alpha-neurexins are not required for synapse formation, but are essential for Ca2+-triggered neurotransmitter release. Neurotransmitter release is impaired because synaptic Ca2+ channel function is markedly reduced, although the number of cell-surface Ca2+ channels appears normal. These data suggest that alpha-neurexins organize presynaptic terminals by functionally coupling Ca2+ channels to the presynaptic machinery.  相似文献

Many palaeoclimate records from the North Atlantic region show a pattern of rapid climate oscillations, the so-called Dansgaard-Oeschger events, with a quasi-periodicity of approximately 1,470 years for the late glacial period. Various hypotheses have been suggested to explain these rapid temperature shifts, including internal oscillations in the climate system and external forcing, possibly from the Sun. But whereas pronounced solar cycles of approximately 87 and approximately 210 years are well known, a approximately 1,470-year solar cycle has not been detected. Here we show that an intermediate-complexity climate model with glacial climate conditions simulates rapid climate shifts similar to the Dansgaard-Oeschger events with a spacing of 1,470 years when forced by periodic freshwater input into the North Atlantic Ocean in cycles of approximately 87 and approximately 210 years. We attribute the robust 1,470-year response time to the superposition of the two shorter cycles, together with strongly nonlinear dynamics and the long characteristic timescale of the thermohaline circulation. For Holocene conditions, similar events do not occur. We conclude that the glacial 1,470-year climate cycles could have been triggered by solar forcing despite the absence of a 1,470-year solar cycle.  相似文献

We propose a nonlinear time series model where both the conditional mean and the conditional variance are asymmetric functions of past information. The model is particularly useful for analysing financial time series where it has been noted that there is an asymmetric impact of good news and bad news on volatility (risk) transmission. We introduce a coherent framework for testing asymmetries in the conditional mean and the conditional variance, separately or jointly. To this end we derive both a Wald and a Lagrange multiplier test. Some of the new asymmetric model's moment properties are investigated. Detailed empirical results are given for the daily returns of the composite index of the New York Stock Exchange. There is strong evidence of asymmetry in both the conditional mean and the conditional variance functions. In a genuine out‐of‐sample forecasting experiment the performance of the best fitted asymmetric model, having asymmetries in both conditional mean and conditional variance, is compared with an asymmetric model for the conditional mean, and with no‐change forecasts. This is done both in terms of conditional mean forecasting as well as in terms of risk forecasting. The subduction of oceanic plateaux, which contain extraordinarily thick basaltic crust and are the marine counterparts of continental flood-basalt provinces, is an important factor in many current models of plate motion and provides a potential mechanism for triggering plate reorganization. To evaluate such models, it is essential to decipher the history of the collision between the largest and thickest of the world's oceanic plateaux, the Ontong Java plateau, and the Australian plate, but this has been hindered by poor constraints for the arrival of the plateau at the Melanesian trench. Here we present (40)Ar-(39)Ar geochronological data on hotspot volcanoes in eastern Australian that reveal a strong link between collision of the Greenland-sized Ontong Java plateau with the Melanesian arc and motion of the Australian plate. The new ages define a short-lived period of reduced northward plate motion between 26 and 23 Myr ago, coincident with an eastward offset in the contemporaneous tracks of seamount chains in the Tasman Sea east of Australia. These features record a brief westward deflection of the Australian plate as the plateau entered and choked the Melanesian trench 26 Myr ago. From 23 Myr ago, Australia returned to a rapid northerly trajectory at roughly the same time that southwest-directed subduction began along the Trobriand trough. The timing and brevity of this collisional event correlate well with offsets in hotspot seamount tracks on the Pacific plate, including the archetypal Hawaiian chain, and thus provide strong evidence that immense oceanic plateaux, like the Ontong Java, can contribute to initiating rapid change in plate boundaries and motions on a global scale.  相似文献
