Bone marrow cells give rise to distinct cell clones within the thymus

The thymus is the major, if not the sole site of maturation of T lymphocytes from their haematopoietic precursors. During embryonic life (at a few well-defined intervals, at least in birds) the thymus receives thymus-homing haematopoietic precursors that give rise to antigen-specific functional T lymphocytes. Although the number and thymic location of distinct T-cell lineages destined to form the peripheral T-cell pool are not yet well defined, at least two independent pathways have been proposed. First, thymic subcapsular lymphoblasts divide and differentiate to give rise to small deep cortical thymic lymphocytes, medullary lymphocytes and thymus emigrants (I.W., unpublished data) and second, the medulla contains an independent self-renewing population that contains the precursors of the peripheral T-cell pool. Following irradiation the thymus may be repopulated by injected haematopoietic cells presumably related to the thymus-homing haematopoietic cells of the embryo. Here we have reconstituted irradiated mice with limiting numbers of bone marrow cells from Thy-1 congeneic donors and have found distinct clones of cells within the thymus. The pattern of reconstitution by the precursor cells indicates that two independent thymus lineages exist: cortex plus medulla, and medulla alone.     

Spike--nucleocapsid interaction in Semliki Forest virus reconstructed using network antibodies

Using in vitro immunization, we have reconstructed three consecutive steps of an idiotype network to show that the nucleocapsid of Semliki Forest virus contains a specific 'receptor' for the cytoplasmic tail of the E2 spike glycoprotein. This interaction could be the basis for the highly selective inclusion of viral glycoproteins--and exclusion of host cell surface proteins--during virus budding.     

Incorporation of base analogues for detection of unusual base compositions in plant metaphase chromosomes (Vicia faba,Leguminosae)

Summary Bromodeoxyuridine and bromodeoxycytidine with fluorescence-plus-Giemsa staining resulted in similar patterns of asymmetric bands inVicia faba chromosomes. Cell proliferation in the rat pituitary gland: A mechanism of control in prolactin cells

Summary We investigated the relationship between prolactin content and DNA replication in the anterior pituitary gland. Thymidine incorporation in pregnant rats is significantly lower than in virgin controls. This is accompanied by a decreased activity of DNA polymerase. Sulpiride administration to pregnant rats enhances thymidine incorporation to levels similar to virgin controls. The results indicate a negative feedback between prolactin content and DNA synthesis in the rat anterior pituitary gland.We are grateful to Prof. Carlos J. Influence of pulsing electromagnetic field on the frequency of sister-chromatid exchanges in cultured mammalian cells

Exposures of Chinese hamster cells to pulsing electromagnetic field (PEMF) with 0.18-2.5 mT did not influence the baseline frequency of sister-chromatid exchanges (SCE). The results suggest that PEMF with the magnetic intensity examined does not interfere with DNA replication nor produce DNA lesions, thereby leading to an increased frequency of SCE.     

Hybridization histochemistry

Summary The location of gene expression by hybridization histochemistry is being applied in many areas of research and diagnosis. The aim of this technique is to detect specific mRNA in cells and tissues by hybridization with a complementary DNA or RNA probe. Requirements for optimal specificity, sensitivity, resolution and speed of detection may not all be encompassed in one simple technique suitable for all applications, thus appropriate procedures should be selected for specific objectives. With reference to published procedures and our own extensive experience, we have evaluated fixatives, probes, labels and other aspects of the technique critical to the preservation and hybridization in situ of mRNA and detection and quantition of hybrids.     

Measurement of urinary steroid production rates using stable-isotopes and GC-MS

Preventive effects of gliclazide on the development of cholesterol induced atherosclerosis in rabbits]

Administration of gliclazide inhibited the development of aortic and particularly coronary lesions induced by the cholesterol-containing diet; but it did not noticeably affect the evolution of plasma lipid disturbances.
