Gene polymorphism in Netherton and common atopic disease.

Atopic dermatitis (AD) and asthma are characterized by IgE-mediated atopic (allergic) responses to common proteins (allergens), many of which are proteinases. Loci influencing atopy have been localized to a number of chromosomal regions, including the chromosome 5q31 cytokine cluster. Netherton disease is a rare recessive skin disorder in which atopy is a universal accompaniment. The gene underlying Netherton disease (SPINK5) encodes a 15-domain serine proteinase inhibitor (LEKTI) which is expressed in epithelial and mucosal surfaces and in the thymus. We have identified six coding polymorphisms in SPINK5 (Table 1) and found that a Glu420-->Lys variant shows significant association with atopy and AD in two independent panels of families. Our results implicate a previously unrecognized pathway for the development of common allergic illnesses.     

A method to collect cervical smears from small breeds of monkeys.

A simple method is described to collect cervical smears and to have a clear view of cervix for small breed of monkeys. This method was found useful to collect adequate cytology smears with good preservation of cellular morphology.     

High-affinity binding of beta-alanine to cerebral synaptosomes might involve glycine-receptors.

High-affinity, Na+-independent binding of beta-alanine to a synaptosomal fraction of rat brain was potently inhibited by glycine and by some other alpha-amino acids, but not by taurine or GABA. This binding mechanism, which was also sensitive to both bicuculline and strychnine, might involve synaptic receptors for both beta-alanine and glycine.     

Progressive resistance against the male recombination factor 31.1 MRF acquired by Drosophila melanogaster.

A genetically abnormal but structurally normal second chromosome (31.1 MRF) which induces recombination in the male sex, was introduced by outcrossing into the cytoplasm of a normal strain. In this new combination, male recombination frequency induced by females dropped from 3.81% to 0.17% within 10 generations, indicating a gradual acquisition of resistance against the activities of the 31.1 MRF.     

Lipoprotein lipase activator deficiency in very low density lipoproteins in rat nephrotic syndrome.

Marked urinary loss of lipoprotein lipase activator in experimental rat nephrotic syndrome may be partly responsible for its deficiency in plasma very low density lipoproteins.     

Effects of chronic treatment with ACTH on the intracellular levels of cyclic-AMP and cyclic-GMP in the rat adrenal cortex.

The effects of chronic ACTH treatment on the increase in the intracellular concentration of cyclic-AMP and cyclic-GMP acutely elicited by ACTH in the rat adrenal cortex were investigated. The results are consistent with the hypothesis that chronic ACTH treatment stimulates a) the de novo synthesis of adenylate- and guanylate-cyclase or b) the synthesis of new specific membrane receptors for ACTH.     

Criteria for the appearance of post-tetanic potentiation of transmission at central synapses in Helix aspersa.

Long-lasting potentiation of inhibitory post-synaptic potentials occurs at 2 identifiable synapses in Helix brain. It appears only after tetanic stimulation and after a minimum of 20 impulses. Its amplitude and duration depends on the number of stimuli.     

Glutathione peroxidase in dried blood spots.

A new procedure utilizing dried blood spots was developed for detecting glutathione peroxidase deficiency. Samples from a known patient with a partial defect and from rats with an induced deficiency were distinguished from respective control groups by their longer defluorescence endpoints. Samples from 100 patients with anemia and 2 phenyl-ketonuric infants on low-protein diets contained glutathione peroxidase activity similar to that in 82 controls, when screened for the enzyme defect by the new procedure.     

Effects of the antiprotease Trasylol on peripheral blood leucocytes.

Binding of the antiprotease Trasylol to human peripheral blood lymphocytes and polymorphonuclear leucocytes (PMNs) was demonstrated at the ultrastructural level using an indirect immunoperoxidase technique. This also revealed endocytosis of membrane bound Trasylol by PMNs. Trasylol inhibited PHA- and ConA-induced lymphocyte stimulation, and was cytotoxic to unstimulated cells.     

Further characterization of the effects of ultraviolet irradiation of the amphibian egg

UV-irradiation of the vegetal hemisphere of amphibian eggs leads to developmental abnormalities in neural morphogenesis. The possibility that the egg's transient sensitivity to irradiation could be due to pigmentation changes was examined in albino eggs. The tissue specificity of the effects of irradiation was analyzed by exchanging the ectoderm between irradiated and control embryos.
