Mutations in CTC1, encoding conserved telomere maintenance component 1, cause Coats plus

Coats plus is a highly pleiotropic disorder particularly affecting the eye, brain, bone and gastrointestinal tract. Here, we show that Coats plus results from mutations in CTC1, encoding conserved telomere maintenance component 1, a member of the mammalian homolog of the yeast heterotrimeric CST telomeric capping complex. Consistent with the observation of shortened telomeres in an Arabidopsis CTC1 mutant and the phenotypic overlap of Coats plus with the telomeric maintenance disorders comprising dyskeratosis congenita, we observed shortened telomeres in three individuals with Coats plus and an increase in spontaneous γH2AX-positive cells in cell lines derived from two affected individuals. CTC1 is also a subunit of the α-accessory factor (AAF) complex, stimulating the activity of DNA polymerase-α primase, the only enzyme known to initiate DNA replication in eukaryotic cells. Thus, CTC1 may have a function in DNA metabolism that is necessary for but not specific to telomeric integrity.

Requirement of Src kinases Lyn, Hck and Fgr for BCR-ABL1-induced B-lymphoblastic leukemia but not chronic myeloid leukemia

The Abl kinase inhibitor imatinib mesylate is the preferred treatment for Philadelphia chromosome-positive (Ph(+)) chronic myeloid leukemia (CML) in chronic phase but is much less effective in CML blast crisis or Ph(+) B-cell acute lymphoblastic leukemia (B-ALL). Here, we show that Bcr-Abl activated the Src kinases Lyn, Hck and Fgr in B-lymphoid cells. BCR-ABL1 retrovirus-transduced marrow from mice lacking all three Src kinases efficiently induced CML but not B-ALL in recipients. The kinase inhibitor CGP76030 impaired the proliferation of B-lymphoid cells expressing Bcr-Abl in vitro and prolonged survival of mice with B-ALL but not CML. The combination of CGP76030 and imatinib was superior to imatinib alone in this regard. The biochemical target of CGP76030 in leukemia cells was Src kinases, not Bcr-Abl. These results implicate Src family kinases as therapeutic targets in Ph(+) B-ALL and suggest that simultaneous inhibition of Src and Bcr-Abl kinases may benefit individuals with Ph(+) acute leukemia.     

The effect of anoxia and of muscle activity on the lymphatic and venous transport of lactate dehydrogenase

Zusammenfassung Nach 3 1/2 stündiger Muskelischämie wurde bei Hunden mit Ductus thoracicus-Fistel eine signifikante Zunahme der Milchsäuredehydrogenase-Aktivität im Blutserum und in der Lymphe der anoxischen Extremität bei gleichzeitiger signifikanter arterovenöser Aktivitätsdifferenz beobachtet. Auch Muskelarbeit verursachte eine Erhöhung der Enzymaktivität im Serum und in der Lymphe. Durch Unterbrechen der Verbindung zwischen Venen und Lymphsystem konnte das direkte Eindringen des Enzymproteins in die Blutkapillaren bewiesen werden.     

Identification of an emerin–β-catenin complex in the heart important for intercalated disc architecture and β-catenin localisation

How mutations in the protein emerin lead to the cardiomyopathy associated with X-linked Emery-Dreifuss muscular dystrophy (X-EDMD) is unclear. We identified emerin at the adherens junction of the intercalated disc, where it co-localised with the catenin family of proteins. Emerin bound to wild type β-catenin both in vivo and in vitro. Mutating the GSK3β phosphorylation sites on β-catenin abolished this binding. Wild type but not mutant forms of emerin associated with X-EDMD were able to reduce β-catenin protein levels. Cardiomyocytes from emerin-null mice hearts exhibited erroneous β-catenin distribution and intercalated disc architecture. Treatment of wild type cardiomyocytes with phenylephrine, which inactivates GSK3β, redistributed emerin and β-catenin. Emerin was identified as a direct target of GSK3β activity since exogenous expression of GSK3β reduced emerin levels at the nuclear envelope. We propose that perturbation to or total loss of the emerin–β-catenin complex compromises both intercalated disc function and β-catenin signalling in cardiomyocytes.

Uptake of3H-noradrenaline and3H-5-hydroxytryptamine in cultured rat brainstem

Zusammenfassung Die zelluläre Lokalisation der Aufnahme von³H-NA und³H-5HT in den Hirnstamm von Ratten wurde am Modell der Nervengewebskultur untersucht. Im Gegensatz zu den Aminosäure-Transmittern, welche in Neurone und Gliazellen aufgenommen werden, zeigen Monoamine nur eine Aufnahme in Zellkörper und Fortsätze von Neuronen, nicht jedoch in Gliazellen. Es besteht eine Korrelation zwischen den Ergebnissen von fluoreszenzmikroskopischen Versuchen und unseren autoradiographischen Untersuchungen.

A histochemical study of immune destruction of the lymphopoietic system

Zusammenfassung Eine auffallende Zunahme von -Galactosidase kommt im lymphoetischen Säuger-Gewebe im Zusammenhang mit der immunologischen Auflösung dieses Systems vor. Die hernach auftretende Proliferation retikulärer Zellen steht im Zusammenhang mit der Regeneration dieses Organs. Diese Zellen enthalten eine reichliche Anzahl lysosomatischer Enzyme (z.B. -Galactosidase).
