Summary Exposure of green peach aphids,Myzus persicae (Sulz.), to an atmosphere containing mercury vapor resulted in a curtailment ofembryogenesis and larviposition by adults, and in the development by larvae and adults of a cuticular darkening of their legs, head capsule, antennae, cornicles and cauda. Mortality of affected larvae resulted from molting difficulties, particularly by last-instar alatiform female and male larvae. Greenbugs,Schizaphis graminum (Rond.), and pea aphids,Acyrthosiphon pisum (Harr.), responded to mercury vapor exposure in similar ways. 相似文献

Scrutiny of the data from these studies reveals that the C58/J alcohol-preferring mice have significantly lower baseline methionine-enkephalin levels in both the corpus striatum and hypothalamus compared to C3H/CHRGL/2 non-alcohol-preferring mice. In other brain regions in these two strains, specifically, pituitary, amygdala, midbrain, and hippocampus, analysis of methionine-enkephalin levels did not show any significant differences. This suggests that the hypothalamus may indeed be a specific locus involved in the regulation of alcohol intake, via the molecular interaction between neuroamines, opioid peptides, as they are influenced by genetics and environment. 相似文献

Exposures of Chinese hamster cells to pulsing electromagnetic field (PEMF) with 0.18-2.5 mT did not influence the baseline frequency of sister-chromatid exchanges (SCE). The results suggest that PEMF with the magnetic intensity examined does not interfere with DNA replication nor produce DNA lesions, thereby leading to an increased frequency of SCE. 相似文献

The location of gene expression by hybridization histochemistry is being applied in many areas of research and diagnosis. The aim of this technique is to detect specific mRNA in cells and tissues by hybridization with a complementary DNA or RNA probe. Requirements for optimal specificity, sensitivity, resolution and speed of detection may not all be encompassed in one simple technique suitable for all applications, thus appropriate procedures should be selected for specific objectives. With reference to published procedures and our own extensive experience, we have evaluated fixatives, probes, labels and other aspects of the technique critical to the preservation and hybridization in situ of mRNA and detection and quantitation of hybrids. 相似文献

The radiosensitization brought about by oridonin, one of the Rabdosia diterpenoids, alone or in combination with misonidazole, was investigated in Chinese hamster V79 cells. The enhancement ratio of 1.92 was obtained when 0.01 mM oridonin and 1 mM misonidazole were administered to hypoxic cells under radiation. The enhancement ratios of oridonin and misonidazole for hypoxic cells were 1.16 and 1.59 respectively. Hence, a supra-additive effect was obtained by the combined treatment with these two drugs. Under aerobic conditions, no effect of 0.01 mM oridonin on the radiosensitization caused by misonidazole was observed. 相似文献

A gelatin sponge model system for tumor cell inoculation and retrieval of tumor-associated leukocytes is described. Gelatin sponges pre-implanted in nude mice harboring tumorigenic Chinese hamster ovary cells (line CHO) were examined at 2 and 11 days after injection of tumor cells for tumor cell content and leukocyte accumulation after digesting the sponge matrix in collagenase solution. The data indicate a progressive influx of host cells consisting primarily of macrophages, neutrophils and lymphocytes. The total number of viable tumor cells as well as the fraction of surviving tumor cells with clonogenic potential also increased with tumor age. Blank sponges not harboring tumor cells elicited an inflammatory response in the animals which did not change appreciably with length of sponge residence. However, when the sponges were harboring tumor cells, the accumulation of host leukocytes far exceeded that which occurred in blank sponges. This observation suggests a host response directed toward the tumor which is absent in animals bearing blank sponges. Apart from providing anchorage for injected cells, the gelatin sponge, by virtue of its digestibility in collagenase, makes possible the easy retrieval and precise quantitation of tumor-associated host cells. 相似文献

Chronic ovariectomized rats treated neonatally with MSG showed reduced circulating concentrations of LH coupled with elevated hypothalamic LHRH stores. Despite the apparent loss of LHRH secretion, the small pituitary glands showed an increased density of LHRH receptors and normal responsiveness to the releasing hormone. The positive feedback effects of progesterone on LH release in oestrogen-primed animals was greatly exaggerated reflecting the build-up of hypothalamic LHRH stores without loss of pituitary responsiveness to LHRH. 相似文献

The first tetraspanins were discovered on surface of human leucocytes, but it was rapidly demonstrated that they had a wider
tissue expression. Twenty-six molecules display sufficient homology to belong to the same superfamily. Their function is not
precisely known, but data coming from biochemical studies or knockout mice suggest that they play a major role in membrane
biology. One of their outstanding properties is their ability to form a network of multimolecular complexes, the 'tetraspanin
web', in which integrins are included. The structure of these complexes is under investigation, but some of the rules that
govern their organization have already been unraveled. The challenge is to determine how the organization of the 'tetraspanin
web' modifies the function of its constitutive molecules and consequently influences cellular behaviour. The implications
may be considerable for the understanding of basic cellular processes such as migration and also of diseases related to loss
or mutation of a single tetraspanin.

Transformation: a tool for studying fungal pathogens of plants 总被引:18,自引:0,他引:18
Plant diseases caused by plant pathogenic fungi continuously threaten the sustainability of global crop production. An effective
way to study the disease-causing mechanisms of these organisms is to disrupt their genes, in both a targeted and random manner,
so as to isolate mutants exhibiting altered virulence. Although a number of techniques have been employed for such an analysis,
those based on transformation are by far the most commonly used. In filamentous fungi, the introduction of DNA by transformation
typically results in either the heterologous (illegitimate) integration or the homologous integration of the transforming
DNA into the target genome. Homologous integration permits a targeted gene disruption by replacing the wild-type allele on
the genome with a mutant allele on transforming DNA. This process has been widely used to determine the role of newly isolated
fungal genes in pathogenicity. The heterologous integration of transforming DNA causes a random process of gene disruption
(insertional mutagenesis) and has led to the isolation of many fungal mutants defective in pathogenicity. A big advantage
of insertional mutagenesis over the more traditional chemical or radiation mutagenesis procedures is that the mutated gene
is tagged by transforming DNA and can subsequently be cloned using the transforming DNA. The application of various transformation-based
techniques for fungal gene manipulation and how they have increased our understanding and appreciation of some of the most
serious plant pathogenic fungi are discussed.

In the developing brain, postmitotic neurons exhibit dynamic changes in mode, direction, tempo and rate of migration as they traverse different cortical layers. Such changes in cell migration require orchestrated activities of multiple guidance cues and transmembrane signals. In this article, we first describe when, where and how cerebellar granule cells alter their migratory behavior during the entire course of their migration. We then present how internal (inherent) programs regulate the sequential changes in the migratory behavior of granule cells in vitro. Finally, we discuss the roles of external guidance cues and transmembrane signals in controlling granule cell migration. 相似文献
