Experimental autoimmune encephalomyelitis (EAE) induced by immunization to the basic protein of central nervous system myelin (BP) is a paralytic disease in which T lymphocytes attack the individual's own central nervous system. As the target is in white matter, EAE has been considered an experimental model of some aspects of human disease such as multiple sclerosis. To investigate whether autoimmune T lymphocytes could produce paralysis, we studied the effects on the electrophysiology of isolated nerves produced by T-lymphocyte lines reactive specifically to BP or other antigens. We now report that propagation of action potentials evoked by electrical stimulation was blocked by incubating optic nerves with specific anti-BP T cells. This blockade could be reversed for up to two hours by removing the anti-BP line cells from the optic nerve. The anti-BP line cells had no effect on conduction along allogeneic optic nerves or syngeneic peripheral nerves. This indicates that disruption of the function of myelin in neuroimmunological disease may result from an immunologically specific interaction between autoimmune T lymphocytes and myelin antigens.  相似文献

Acute emetic response to relatively low-doses of X-irradiation on suncus (Suncus murinus) was examined. The behaviors recorded for each subject, using a video-cassette recorder system, were (1) the number of emesis, during exposure to a dose of 3.0 Gy; (2) emesis threshold. Results showed that the emetic threshold was observed at 0.85 Gy, and the number of radioemesis during exposure was 29. However, the observed threshold dose became 2.22 Gy following olfactory bulbectomy. The emetic number decreased significantly (p<0.01), and reached a value one-fourth of the sham-control. The bulbectomized suncus showed a resistance to X-irradiation. Furthermore, I examined whether the animals could also acquire radio-resistance when they were subjected to a brief of dose X-ray (0.3 Gy) prior to a exposure to 3.0 Gy. Results showed that brief pre-exposure increased the observed threshold, a pattern that was exactly the same as shown in the bulbectomized animals. Increasing the pre-exposure dose further to 0.45–0.60 Gy, however, resulted in the complete disappearance of the effect. These results suggest that only the mice pre-irradiated with 0.30 Gy acquired resistance to radiation-induced emesis.  相似文献

Summary Ectopic pituitary transplantation induced a high incidence of adenomyosis in SHN mice. Early signs of the development of adenomyosis were the penetration of stromal connective tissue into myometrium followed by uterine gland invasion. Soluble dipeptidyl peptidase IV (EC 3.4.14.5) was purified from the 100,000 X g supernatant fraction of pig liver homogenate. The purified enzyme had the same properties as, and immunological identity with, the membrane-bound enzyme which was described previously. However, the purified enzyme had a pattern of molecular heterogeneity different from the membrane-bound enzyme; this was shown by isoelectric focusing. Carbohydrate analysis revealed that the soluble enzyme contained glucose, which is not found in the membrane-bound one, and less fucose, mannose, and sialic acid than the latter. From these results, we conclude that the soluble form of dipeptidyl peptidase IV in pig liver is closely related to the membrane-bound enzyme, but is not simply a proteolytically solubilized product of it.  相似文献

Summary 14C-Indomethacin (ID) was applied to the skin of guinea-pigs as an ointment. After several applications, the concentration of ID in the skin and muscle under the applied site reached a constant level. The concentration of ID in the muscle as well as the skin may be enough to exert an anti-inflammatory effect.  相似文献
