Summary The absorption of ouabain and digitoxin into the gut of the milkweed bug was studied by a tissue-accumulation technique. Uptake of both compounds obeyed diffusion kinetics, was Na+ sensitive, and for digitoxin, was concentrative.The authors express their appreciation to Dr.James A. Slater and Ms.Flavia O'Rourke, Biological Sciences Group, University of Connecticut, Storrs, Conn. (USA) for supplying milkweed bugs. Rossbank functioned from 1840 to 1854 as one of a chain of British Colonial Observatories which combined with European and Asian observatories in the study of terrestrial magnetism. It was established in Hobart, Tasmania, by the Governor of Van Diemen's Land, Sir John Franklin, and Captain James Clark Ross, R.N., commanding H.M. ships Erebus and Terror. The history and operation of the Rossbank Observatory is related, its instruments described, and the results discussed.

Biographical notes on the Observatory staff, with lists of its archives and instruments are provided. A number of previously unpublished pictures show the Observatory in use and the surviving buildings and instruments today.  相似文献

Many studies have assessed genetic changes in exotic plant species in their native and introduced ranges, but none have focused on genetic variation in native plant species in response to exotic invasion. We examine characteristics of native plant communities within and outside old (> 25 year) invasions of Acroptilon repens (Russian knapweed) and Cardaria draba (hoary cress). We also document genetic variability of 4 native grass populations ( Hesperostipa comata [needle and thread], Achnatherum hymenoides [Indian ricegrass], Sporobolus airoides [alkali sacaton], and Poa secunda [Sandberg bluegrass]) from 2 areas: adjacent to and within weed invasions. Native plant species richness and diversity did not differ between invaded and noninvaded areas. Inter-simple sequence repeat (ISSR) analysis of individual native perennial grasses of each of the 4 species suggests that populations exposed to long-term coexistence with exotics may differ from adjacent noninvaded populations. We suggest that future research efforts should focus on intraspecific diversity of native plant species to identify possible candidates for restoration following weed control.  相似文献

The most unique feature of Earth is the existence of life, and the most extraordinary feature of life is its diversity. Approximately 9 million types of plants, animals, protists and fungi inhabit the Earth. So, too, do 7 billion people. Two decades ago, at the first Earth Summit, the vast majority of the world's nations declared that human actions were dismantling the Earth's ecosystems, eliminating genes, species and biological traits at an alarming rate. This observation led to the question of how such loss of biological diversity will alter the functioning of ecosystems and their ability to provide society with the goods and services needed to prosper.  相似文献

Tissue development and regeneration depend on cell-cell interactions and signals that target stem cells and their immediate progeny. However, the cellular behaviours that lead to a properly regenerated tissue are not well understood. Using a new, non-invasive, intravital two-photon imaging approach we study physiological hair-follicle regeneration over time in live mice. By these means we have monitored the behaviour of epithelial stem cells and their progeny during physiological hair regeneration and addressed how the mesenchyme influences their behaviour. Consistent with earlier studies, stem cells are quiescent during the initial stages of hair regeneration, whereas the progeny are more actively dividing. Moreover, stem cell progeny divisions are spatially organized within follicles. In addition to cell divisions, coordinated cell movements of the progeny allow the rapid expansion of the hair follicle. Finally, we show the requirement of the mesenchyme for hair regeneration through targeted cell ablation and long-term tracking of live hair follicles. Thus, we have established an in vivo approach that has led to the direct observation of cellular mechanisms of growth regulation within the hair follicle and that has enabled us to precisely investigate functional requirements of hair-follicle components during the process of physiological regeneration.  相似文献

Cancer immunoediting is a process by which immune cells, particularly lymphocytes of the adaptive immune system, protect the host from the development of cancer and alter tumour progression by driving the outgrowth of tumour cells with decreased sensitivity to immune attack. Carcinogen-induced mouse models of cancer have shown that primary tumour susceptibility is thereby enhanced in immune-compromised mice, whereas the capacity for such tumours to grow after transplantation into wild-type mice is reduced. However, many questions about the process of cancer immunoediting remain unanswered, in part because of the known antigenic complexity and heterogeneity of carcinogen-induced tumours. Here we adapted a genetically engineered, autochthonous mouse model of sarcomagenesis to investigate the process of cancer immunoediting. This system allows us to monitor the onset and growth of immunogenic and non-immunogenic tumours induced in situ that harbour identical genetic and histopathological characteristics. By comparing the development of such tumours in immune-competent mice with their development in mice with broad immunodeficiency or specific antigenic tolerance, we show that recognition of tumour-specific antigens by lymphocytes is critical for immunoediting against sarcomas. Furthermore, primary sarcomas were edited to become less immunogenic through the selective outgrowth of cells that were able to escape T lymphocyte attack. Loss of tumour antigen expression or presentation on major histocompatibility complex I was necessary and sufficient for this immunoediting process to occur. These results highlight the importance of tumour-specific-antigen expression in immune surveillance, and potentially, immunotherapy.  相似文献

Biodegradation of crude oil in subsurface petroleum reservoirs is an important alteration process with major economic consequences. Aerobic degradation of petroleum hydrocarbons at the surface is well documented and it has long been thought that the flow of oxygen- and nutrient-bearing meteoric waters into reservoirs was necessary for in-reservoir petroleum biodegradation. The occurrence of biodegraded oils in reservoirs where aerobic conditions are unlikely, together with the identification of several anaerobic microorganisms in oil fields and the discovery of anaerobic hydrocarbon biodegradation mechanisms, suggests that anaerobic degradation processes could also be responsible. The extent of anaerobic hydrocarbon degradation processes in the world's deep petroleum reservoirs, however, remains strongly contested. Moreover, no organism has yet been isolated that has been shown to degrade hydrocarbons under the conditions found in deep petroleum reservoirs. Here we report the isolation of metabolites indicative of anaerobic hydrocarbon degradation from a large fraction of 77 degraded oil samples from both marine and lacustrine sources from around the world, including the volumetrically important Canadian tar sands. Our results therefore suggest that anaerobic hydrocarbon degradation is a common process in biodegraded subsurface oil reservoirs.  相似文献

非英语专业学生口语能力有待提高。如何提高学生的口语水平始终是英语教学中的难点和重点。传统教学模式下的大学英语教学束缚了学生学习潜能的发挥。大学英语教学模式的改革可以考虑采用“任务型教学法”。任务型教学模式是20年来交际教学思想的一种发展形态,它把语言运用的基本理念转化为具有实践意义的课堂教学模式。本研究将任务型教学理念应用于非英语专业英语口语教学之中,通过对任务型教学理论的定义、特征和理论基础等方面的阐析,提出任务型口语教学的总体设计思路和具体教学改革措施。  相似文献

The mouse mutation fragilitas ossium (fro) leads to a syndrome of severe osteogenesis and dentinogenesis imperfecta with no detectable collagen defect. Positional cloning of the locus identified a deletion in the gene encoding neutral sphingomyelin phosphodiesterase 3 (Smpd3) that led to complete loss of enzymatic activity. Our knowledge of SMPD3 function is consistent with the pathology observed in mutant mice and provides new insight into human pathologies.  相似文献
