Gonatus onyx is one of the most abundant cephalopods in the Pacific and Atlantic Oceans and is an important prey species for a variety of vertebrate predators, but a full understanding of its life history has been hampered because spawning occurs at great depths, where observation is difficult. Here we describe post-spawning egg care, or brooding, in this deep-sea squid. Our finding is unexpected because this behaviour differs from the reproductive habits of all other known squid species. 相似文献

Analytical tools that have spatial resolution at the nanometre scale are indispensable for the life and physical sciences. It is desirable that these tools also permit elemental and chemical identification on a scale of 10 nm or less, with large penetration depths. A variety of techniques in X-ray imaging are currently being developed that may provide these combined capabilities. Here we report the achievement of sub-15-nm spatial resolution with a soft X-ray microscope--and a clear path to below 10 nm--using an overlay technique for zone plate fabrication. The microscope covers a spectral range from a photon energy of 250 eV (approximately 5 nm wavelength) to 1.8 keV (approximately 0.7 nm), so that primary K and L atomic resonances of elements such as C, N, O, Al, Ti, Fe, Co and Ni can be probed. This X-ray microscopy technique is therefore suitable for a wide range of studies: biological imaging in the water window; studies of wet environmental samples; studies of magnetic nanostructures with both elemental and spin-orbit sensitivity; studies that require viewing through thin windows, coatings or substrates (such as buried electronic devices in a silicon chip); and three-dimensional imaging of cryogenically fixed biological cells. 相似文献

Iron is required to produce haem and iron-sulphur (Fe-S) clusters, processes thought to occur independently. Here we show that the hypochromic anaemia in shiraz (sir) zebrafish mutants is caused by deficiency of glutaredoxin 5 (grx5), a gene required in yeast for Fe-S cluster assembly. We found that grx5 was expressed in erythroid cells of zebrafish and mice. Zebrafish grx5 rescued the assembly of grx5 yeast Fe-S, showing that the biochemical function of grx5 is evolutionarily conserved. In contrast to yeast, vertebrates use iron regulatory protein 1 (IRP1) to sense intracellular iron and regulate mRNA stability or the translation of iron metabolism genes. We found that loss of Fe-S cluster assembly in sir animals activated IRP1 and blocked haem biosynthesis catalysed by aminolaevulinate synthase 2 (ALAS2). Overexpression of ALAS2 RNA without the 5' iron response element that binds IRP1 rescued sir embryos, whereas overexpression of ALAS2 including the iron response element did not. Further, antisense knockdown of IRP1 restored sir embryo haemoglobin synthesis. These findings uncover a connection between haem biosynthesis and Fe-S clusters, indicating that haemoglobin production in the differentiating red cell is regulated through Fe-S cluster assembly. 相似文献

Shackleton crater at the Moon's south pole has been suggested as a possible site of concentrated deposits of water ice, on the basis of modelling of bi-static radar polarization properties and interpretations of earlier Earth-based radar images. This suggestion, and parallel assumptions about other topographic cold traps, is a significant element in planning for future lunar landings. Hydrogen enhancements have been identified in the polar regions, but these data do not identify the host species or its local distribution. The earlier Earth-based radar data lack the resolution and coverage for detailed studies of the relationship between radar scattering properties, cold traps in permanently shadowed areas, and local terrain features such as the walls and ejecta of small craters. Here we present new 20-m resolution, 13-cm-wavelength radar images that show no evidence for concentrated deposits of water ice in Shackleton crater or elsewhere at the south pole. The polarization properties normally associated with reflections from icy surfaces in the Solar System were found at all the observed latitudes and are strongly correlated with the rock-strewn walls and ejecta of young craters, including the inner wall of Shackleton. There is no correlation between the polarization properties and the degree of solar illumination. If the hydrogen enhancement observed by the Lunar Prospector orbiter indicates the presence of water ice, then our data are consistent with the ice being present only as disseminated grains in the lunar regolith. 相似文献

Principia (Book 1, Sect. 6), Newton's Lemma 28 on the algebraic nonintegrability of ovals has had an unusually mixed reception. Beginning
in 1691 with Jakob Bernoulli (who accepted the lemma) and Huygens and Leibniz (who rejected it and offered counterexamples),
Lemma 28 has a history of eliciting seemingly contradictory reactions. In more recent times, D.T. Whiteside in 1974 gave an
“unchallengeable counterexample,” while the mathematician V.I. Arnol'd in 1987 sided with Bernoulli and called Newton's argument
an “astonishingly modern topological proof.” This disagreement mostly stems, we argue, from Newton's vague statement of the
lemma. Indeed, we identify several different interpretations of Lemma 28, any one of which Newton may have been intending
to assert, and we then test a number of proposed counterexamples to see which, if any, are true counterexamples to one or
more of these versions of the lemma. Following this, we study Newton's argument for the lemma to see whether and where it
fails to be convincing. In the end, our study of Newton's Lemma 28 provides an answer to the question, Who is right: Huygens,
Leibniz, Whiteside and the others who reject the lemma, or Bernoulli, Arnol'd, and the others who accept it?

Summary The schistosomicides, hycanthone, oxamniquine and praziquantel, were found to inhibit the in vitro RNA synthesis using isolated hamster liver nuclei. Preincubation of the nuclei with these drugs revealed that the inhibitory effect of oxamniquine was irreversible and progressed with time, whereas that of hycanthone and parziquantel was reversible. On the other hand, hycanthone and praziquantel have a high affinity for DNA but oxamniquine does not. The data indicate that the mechanism of inhibition by oxamniquine is different from that of hycanthone and praziquantel. 相似文献

Follicular lymphoma (FL) and diffuse large B-cell lymphoma (DLBCL) are the two most common non-Hodgkin lymphomas (NHLs). Here we sequenced tumour and matched normal DNA from 13 DLBCL cases and one FL case to identify genes with mutations in B-cell NHL. We analysed RNA-seq data from these and another 113 NHLs to identify genes with candidate mutations, and then re-sequenced tumour and matched normal DNA from these cases to confirm 109 genes with multiple somatic mutations. Genes with roles in histone modification were frequent targets of somatic mutation. For example, 32% of DLBCL and 89% of FL cases had somatic mutations in MLL2, which encodes a histone methyltransferase, and 11.4% and 13.4% of DLBCL and FL cases, respectively, had mutations in MEF2B, a calcium-regulated gene that cooperates with CREBBP and EP300 in acetylating histones. Our analysis suggests a previously unappreciated disruption of chromatin biology in lymphomagenesis. 相似文献

Humans differ from other animals in many aspects of anatomy, physiology, and behaviour; however, the genotypic basis of most human-specific traits remains unknown. Recent whole-genome comparisons have made it possible to identify genes with elevated rates of amino acid change or divergent expression in humans, and non-coding sequences with accelerated base pair changes. Regulatory alterations may be particularly likely to produce phenotypic effects while preserving viability, and are known to underlie interesting evolutionary differences in other species. Here we identify molecular events particularly likely to produce significant regulatory changes in humans: complete deletion of sequences otherwise highly conserved between chimpanzees and other mammals. We confirm 510 such deletions in humans, which fall almost exclusively in non-coding regions and are enriched near genes involved in steroid hormone signalling and neural function. One deletion removes a sensory vibrissae and penile spine enhancer from the human androgen receptor (AR) gene, a molecular change correlated with anatomical loss of androgen-dependent sensory vibrissae and penile spines in the human lineage. Another deletion removes a forebrain subventricular zone enhancer near the tumour suppressor gene growth arrest and DNA-damage-inducible, gamma (GADD45G), a loss correlated with expansion of specific brain regions in humans. Deletions of tissue-specific enhancers may thus accompany both loss and gain traits in the human lineage, and provide specific examples of the kinds of regulatory alterations and inactivation events long proposed to have an important role in human evolutionary divergence. 相似文献
