Compared with traditional structure-based approaches for the identification of species-specific ligands, the ab initio approach, based on large-scale protein sequences from different species, has been used to locate specific sites that may be important to the molecular selectivity of species. Statistically significant differences in the distribution of residues in different species and differences in the physicochemical properties of residue-specific sites may largely account for species selectivity. The nicotinic acetylcholine receptor (nAChR), an important neuro-receptor with significantly different ligand selectivity in different species, was used to test our method. Because of the lack of nAChR structural information, the mechanism of ligand discrimination is unclear which hinders attempts at novel molecular design. In this study, the specific site residues 186 and 189 in the principal subunits and residues 34, 55, 56, 57, 106 and 112 in complementary subunits of nAChR were identified by applying our method with stringent statistical cutoffs. These sites were predicted to contribute to ligand selectivity and this result coincides well with the known experimental data. 相似文献

To quantify the characteristics of the power spectrum of plant electrical signals, we defined the following concepts:spectral edge frequency (SEF), spectral center frequency (SCF), power index (PI) and power spectral entropy (PSE). These parameters were used to examine and quantify changes in the power spectrum of electrical signals in maize leaves under osmotic stress. In the absence of osmotic stress, the SEF of the electrical signal in maize leaves was approx. 0.2 Hz and the SCF was approx. 0.1 Hz. The electrical signal in maize leaves was mainly a slow wave signal with a frequency of 0-0.1 Hz. After 2 h osmotic stress, the SEF and SCF of the electrical signal increased to higher frequencies. The proportion of the fast wave frequency also increased to 0.1-0.2 Hz, resulting in a dramatic increase in PSE. We also found that the changes in PSE and SCF were significantly correlated during osmotic stress. We propose that the changes in the PSE and SCF in maize leaves can be used as a sensitive signal indicating water deficit in leaf cells under osmotic stress. Thus, measurement of SCF or PSE of electrical signals in maize leaves could be used to develop early warning and rapid diagnosis techniques for the water demands of plants. 相似文献

Compound machining of silicon carbide ceramics by high speed end electrical discharge milling and mechanical grinding 总被引:1,自引:0,他引:1
A compound process that integrates end electrical discharge (ED) milling and mechanical grinding to machine silicon carbide (SiC) ceramics is developed in this paper. The process employs a turntable with several uniformly-distributed cylindrical copper electrodes and abrasive sticks as the tool, and uses a water-based emulsion as the machining fluid. End electrical discharge milling and mechanical grinding happen alternately and are mutually beneficial, so the process is able to effectively machine a large surface area on SiC ceramic with a good surface quality. The machining principle and characteristics of the technique are introduced. The effects of polarity, pulse duration, pulse interval, open-circuit voltage, discharge current, diamond grit size, emulsion concentration, emulsion flux, milling depth and tool stick number on performance parameters such as the material removal rate, tool wear ratio, and surface roughness have been investigated. In addition, the microstructure of the machined surface under different machining conditions is examined with a scanning electron microscope and an energy dispersive spectrometer. The SiC ceramic was mainly removed by end ED milling during the initial rough machining mode, whereas it is mainly removed by mechanical grinding during the later finer machining mode; moreover, the tool material can transfer to the workpiece surface during the compound process. 相似文献

High-speed all-optical logic circuits have attracted much attention because of their important roles in signal processing in next-generation optical networks.The digital encoder is widely used in binary calculation,multiplexing,demultiplexing,address recognition and data encryption.A priority encoder allows the existence of multiple valid inputs simultaneously,identifies the priority of the request signals and encodes the priority.We propose and experimentally demonstrate an all-optical 4-bit priority encoder for return-to-zero signals at 40 Gbit/s based on cross-gain modulation in semiconductor optical amplifiers.Detuning fil-ters after semiconductor optical amplifiers are employed to improve the output performance.Correct logic bit sequences and clear open eye patterns with extinction ratios exceeding 10 dB are achieved. 相似文献

The electromagnetic interaction of light with polar materials shows a sharp and well defined electromagnetic response in the infrared (IR) region that consists mainly of excitation of optical phonons. Similar to surface plasmons in the visible region, surface phonons can couple efficiently to infrared light in micron-sized antennas made of polar materials. We applied the boundary element method to calculating the infrared electromagnetic response of single SiC disks acting as effective infrared antennas as a function of different parameters such as disk size and thickness. We also analyzed the effect of locating a probing metallic tip near the SiC disk to scatter light in the proximity of the SiC disk, thereby obtaining new spectral peaks connected with localized modes between the tip and the SiC disk. We then further investigated their application in IR scanning probe microscopy. A near-field map of the phononic resonances enhances the understanding of the nature of the IR extinction peaks. 相似文献

A new type of chiral bisoxazoline ligands 1 based on spiro[4,4]-1,6-nonadiene backbone was easily prepared in six steps from racemic spiro[4,4]-nonane-1,6-dione, with the Pd-catalyzed coupling of the enol triflates with CO and amino alcohols as the key steps for the construction of the oxazoline moiety. The structure of the ligand (R,S,S)-1b was unambiguously established by X-ray crystallographic analysis. The chiral Cu(II) complex generated in situ from the combination of spiro bisoxazoline ligand (S,S,S)- 1c and Cu(OTf)2 was effective in the catalysis of asymmetric chlorination of the β-ketoester, methyl 1-oxo-2,3-dihydro-1H- in-dene-2-carboxylate, affording the corresponding chlorinated derivative in 99% yield with 17% ee. 相似文献
