Dictyostelium discoideum cells shed vesicles with associated DNA and vital stain Hoechst 33342

Dictyostelium discoideum cells are highly resis tant to xenobiotics. We previously observed that these primitive eukaryotic cells contain a 170-kDa P-glycoprotein, mediating multidrug resistance in mammalian cells, but nonfunctional in Dictyostelium cells. We show here that D. discoideum cells vitally stained with the DNA-specific dye, Hoechst 33342, release fluorescent material in their culture medium. Electron microscopy and lipid analysis demonstrate the vesicular nature of this material. Moreover, nucleic acids associate with these extracellular vesicles independently of Hoechst vital staining. The main vesicular DNA component exhibits a size >21 kb. Shedding of microvesicles during cell growth is not concomitant with programmed cell death. We propose that these extracellular vesicles are involved in a new cellular resistance mechanism against xenobiotics. Furthermore, since the association of DNA with vesicles occurs in physiological growth conditions and independently of vital staining, the new shedding process might be involved in a more general intercellular mechanism. Decreased number of asialoglycoprotein receptors in diabetic BB Wistar rat

Summary The binding of asialoglycoproteins by hepatic binding protein was studied in freshly isolated hepatocytes from genetically diabetic BB Wistar rats. The number of cell surface asialoglycoprotein receptors was dramatically decreased (58,000±38,000 for diabetic rats compared to 267,000±70,000 for normal rats), while the association equilibrium constant was not changed. These results parallel those obtained with streptozotocin-diabetic rats and support the hypothesis that insulin deprivation is responsible for the decrease in the receptor number.     

Fruiting ofCoprinus congregatus. Interacting effect of radiant flux density and temperature

Summary Illumination during the photoinhibited phase of the development ofCoprinus congregatus reduced fruiting. This inhibitory effect of light was dependent on the radiant flux density and on temperature. Radiant fluxes far lower than the flux provided by moonlight elicited photoresponses.     

Proliferation of adipose cells and their lipid accumulation in the rat at the perinatal stage; histoautoradiographic study]

After thymidine-3H injection into pregnant Rat, histoautoradiographic studies showed that adipose cell proliferation in different subcutaneous and visceral sites, was very important in Rat foetuses about 24 h before birth and that lipid accumulation in the labelled cells was very fast during perinatal 48 h.
