The antigenic saturation of lobster arginine kinase (38 000 daltons) by its specific antibodies has been studied. It was found that seven antigenic binding sites are simultaneously reactive on the surface of the enzyme in the presence of a large excess of antibodies or of their Fab fragments. The research described in this paper is undertaken under the banner of the smart city, a concept that captures the way urban spaces are re-made by the incursion of new technology. Much of smart is centred on converting everyday activities into data, and using this data to generate knowledge mediated by technology. Ordinary citizens, those that may have their lives impacted by the technology, usually are not properly involved in the ‘smartification’ process. Their perceptions, concerns and expectations should inform the conception and development of smart technologies at the same extent. How to engage general public with smart cities research is the central challenge for the Making Metrics Meaningful (MMM) project. Applying a rapid participatory method, ‘Imagine’ over a five-month period (March – July) the research sought to gain insights from the general public into novel forms of information system innovation. This brief paper describes the nature of the accelerated research undertaken and explores some of the themes which emerged in the analysis. Generic themes, beyond the remit of an explicit transport focus, are developed and pointers towards further research directions are discussed. Participatory methods, including engaging with self-selected transport users actively through both picture creation and programmatically specific musical ‘signatures’ as well as group discussion, were found to be effective in eliciting users’ own concerns, needs and ideas for novel information systems. 相似文献

Nonsense-mediated messenger RNA decay (NMD) is triggered by premature translation termination, but the features distinguishing premature from normal termination are unknown. One model for NMD suggests that decay-inducing factors bound to mRNAs during early processing events are routinely removed by elongating ribosomes but remain associated with mRNAs when termination is premature, triggering rapid turnover. Recent experiments challenge this notion and suggest a model that posits that mRNA decay is activated by the intrinsically aberrant nature of premature termination. Here we use a primer extension inhibition (toeprinting) assay to delineate ribosome positioning and find that premature translation termination in yeast extracts is indeed aberrant. Ribosomes encountering premature UAA or UGA codons in the CAN1 mRNA fail to release and, instead, migrate to upstream AUGs. This anomaly depends on prior nonsense codon recognition and is eliminated in extracts derived from cells lacking the principal NMD factor, Upf1p, or by flanking the nonsense codon with a normal 3'-untranslated region (UTR). Tethered poly(A)-binding protein (Pab1p), used as a mimic of a normal 3'-UTR, recruits the termination factor Sup35p (eRF3) and stabilizes nonsense-containing mRNAs. What happens when you take the idea of the biblical Adam—the first human – and apply it to insects? You create an origin story for Nature’s tiniest creatures, one that gives them ‘a Pedigree as ancient as the first creation’. This the naturalist Robert Hooke argued in his treatise, the Micrographia (1665). In what follows, I will retrace how Hooke endeavoured to show that insects—then widely believed to have arisen out of the dirt – were the products of an ancient lineage. These genealogies, while constructed from empirical observation, were conjectures of the imagination. Section 2 shows how Hooke introduced the concept of a ‘prime parent’ (an Adam-insect) to explain the anatomical similarities between ‘mites’. Section 3 demonstrates how Hooke defined the family of “gnats” as tiny machines built from the same components and relates Hookean genealogies to contemporary ideas about Noah’s Ark. Section 4 shows how Hooke outlined the morphology of ‘insects’ (delineating what we now call arthropods). Section 5 explores how Hooke used fossils to study these animals in the distant past. Understanding the mechanisms of evolution requires identification of the molecular basis of the multiple (pleiotropic) effects of specific adaptive mutations. We have characterized the pleiotropic effects on protein levels of an adaptive single-base pair substitution in the coding sequence of a signaling pathway gene in the bacterium Pseudomonas fluorescens SBW25. We find 52 proteomic changes, corresponding to 46 identified proteins. None of these proteins is required for the adaptive phenotype. Instead, many are found within specific metabolic pathways associated with fitness-reducing (that is, antagonistic) effects of the mutation. The affected proteins fall within a single coregulatory network. The mutation 'rewires' this network by drawing particular proteins into tighter coregulating relationships. Although these changes are specific to the mutation studied, the quantitatively altered proteins are also affected in a coordinated way in other examples of evolution to the same niche. 相似文献
