Genetic recombination occurs during meiosis, the key developmental programme of gametogenesis. Recombination in mammals has been recently linked to the activity of a histone H3 methyltransferase, PR domain containing 9 (PRDM9), the product of the only known speciation-associated gene in mammals. PRDM9 is thought to determine the preferred recombination sites--recombination hotspots--through sequence-specific binding of its highly polymorphic multi-Zn-finger domain. Nevertheless, Prdm9 knockout mice are proficient at initiating recombination. Here we map and analyse the genome-wide distribution of recombination initiation sites in Prdm9 knockout mice and in two mouse strains with different Prdm9 alleles and their F(1) hybrid. We show that PRDM9 determines the positions of practically all hotspots in the mouse genome, with the exception of the pseudo-autosomal region (PAR)--the only area of the genome that undergoes recombination in 100% of cells. Surprisingly, hotspots are still observed in Prdm9 knockout mice, and as in wild type, these hotspots are found at H3 lysine 4 (H3K4) trimethylation marks. However, in the absence of PRDM9, most recombination is initiated at promoters and at other sites of PRDM9-independent H3K4 trimethylation. Such sites are rarely targeted in wild-type mice, indicating an unexpected role of the PRDM9 protein in sequestering the recombination machinery away from gene-promoter regions and other functional genomic elements.  相似文献

Harlequin (Hq) mutant mice have progressive degeneration of terminally differentiated cerebellar and retinal neurons. We have identified the Hq mutation as a proviral insertion in the apoptosis-inducing factor (Aif) gene, causing about an 80% reduction in AIF expression. Mutant cerebellar granule cells are susceptible to exogenous and endogenous peroxide-mediated apoptosis, but can be rescued by AIF expression. Overexpression of AIF in wild-type granule cells further decreases peroxide-mediated cell death, suggesting that AIF serves as a free radical scavenger. In agreement, dying neurons in aged Hq mutant mice show oxidative stress. In addition, neurons damaged by oxidative stress in both the cerebellum and retina of Hq mutant mice re-enter the cell cycle before undergoing apoptosis. Our results provide a genetic model of oxidative stress-mediated neurodegeneration and demonstrate a direct connection between cell cycle re-entry and oxidative stress in the ageing central nervous system.  相似文献

Red giants are evolved stars that have exhausted the supply of hydrogen in their cores and instead burn hydrogen in a surrounding shell. Once a red giant is sufficiently evolved, the helium in the core also undergoes fusion. Outstanding issues in our understanding of red giants include uncertainties in the amount of mass lost at the surface before helium ignition and the amount of internal mixing from rotation and other processes. Progress is hampered by our inability to distinguish between red giants burning helium in the core and those still only burning hydrogen in a shell. Asteroseismology offers a way forward, being a powerful tool for probing the internal structures of stars using their natural oscillation frequencies. Here we report observations of gravity-mode period spacings in red giants that permit a distinction between evolutionary stages to be made. We use high-precision photometry obtained by the Kepler spacecraft over more than a year to measure oscillations in several hundred red giants. We find many stars whose dipole modes show sequences with approximately regular period spacings. These stars fall into two clear groups, allowing us to distinguish unambiguously between hydrogen-shell-burning stars (period spacing mostly ~ 50 seconds) and those that are also burning helium (period spacing ~ 100 to 300 seconds).  相似文献

This paper summarizes some of the typical riser vortex-induced vibration (VIV) problems in subsea oil and gas developments,and presents the corresponding computational fluid dynamics (CFD) time domain simulation results to address these problems. First,the CFD time domain simulation approach was applied to analyze the wake field behind a stationary cylinder and a vibrating cylinder. Then a vertical riser VIV response under uniform current was studied. The VIV response time histories revealed some valuable clues that could lead to explanation of the higher harmonics. After that,a vertical riser VIV response under shear current was investigated. A 3 000 ft (1 ft=0.304 8 m) water depth top tensioned riser was sized,and its VIV responses under uniform and shear current were studied. Then this paper continues to discuss one catenary flexible riser VIV response during normal lay. Last,the time domain simulation approach was applied to a partially submerged flexible jumper,to study the jumper VIV behavior,and dynamic motion envelopes. It was demonstrated that the time domain simulation approach is able to disclose details of the flow field,vortex shedding pattern,and riser dynamic behavior,and han-dle different types of risers under different type of currents.  相似文献

Traumatomutilla duplicata duplicata (Gerstaecker, 1874), T. duplicata feia Casal, 1969, T. bruchi André, 1908, T. cristata (Gerstaecker, 1874), T. aterrima (Gerstaecker, 1874) and T. lorena (Cresson, 1902) are proposed as junior synonyms of Traumatomutilla bivittata (Gerstaecker, 1874). Traumatomutilla immaculiceps André, 1901, T. bivittata rubroguttata André, 1901 and T. estrella (Cresson, 1902) are proposed as junior synonyms of T. juvenilis (Gerstaecker, 1874). Traumatomutilla bispiculata (André, 1907) is proposed as a junior synonym of T. miniata (Gerstaecker, 1874). The previously unknown males of T. guarata Casal, 1969 and T. juvenilis are described and illustrated. All species of the T. juvenilis species group are redescribed and illustrated. A new species, Traumatomutilla juvenindica Bartholomay & Williams sp. nov., is described based on males and females from northern South America. Additionally, identification keys to the species and known colour forms of the T. juvenilis species group are provided.

The paper reviews the evolution of Interactive Management, later referred to as Structured Democratic Dialogue, starting from the early 1970s up to this date. The authors propose a generational classification scheme consisting of five periods based primarily on whether some or all stages of the process were implemented synchronously or asynchronously and whether the participants' presence was physical, virtual or hybrid. Other aspects such as modifications in the steps of the process; the evolution of the software; domains of applications; file management; methods of collecting or recording contributions, votes, clarifications and preparation of reports; and key players are also considered and reported within the context of the primary scheme. The paper considers key advances achieved at each generational stage in terms of process or software, discusses associated challenges and concludes with a view towards the future of the emerging fifth generation.  相似文献

Viewing investment projects in new technologies as real options, this paper studies the effects ofendogenous competition and asymmetric information on the strategic exercise of real options. We firstdevelop a multi-period, game-theoretic model and show how competition leads to early exercise andaggressive investment behaviors and how competition erodes option values. We then relax the typicalfull-information assumption found in the literature and allow information asymmetry to exist acrossfirms. Our model shows, in contrast to the literature that payoff is independent of the ordering ofexercise, that the sequential exercise of real options may generate both informational and payoffexternalities. We also find some surpising but interesting results such as having more information isnot necessarily better.  相似文献
