NaOH改性花生壳去除水中亚甲基蓝的吸附特性研究

利用亚甲基蓝水溶液进行实验,测定了NaOH改性处理的花生壳对其的吸附能力.运用单因素实验方法,探讨了改性花生壳的物理化学特性、吸附时间、吸附剂添加量、pH值、温度和离子强度等因素对吸附性能的影响以及等温吸附过程.结果表明,改性能够提高吸附剂的吸附效果.在低温碱性条件下,改性花生壳粉投加量为0.6 g·L~(-1),吸附时间为1 h时,亚甲基蓝的去除率可达94.43%,饱和吸附容量为294.12 mg·g~(-1),等温吸附过程遵循Langmuir方程.     

Melatonin and mitochondrial function during ischemia/reperfusion injury

Ischemia/reperfusion (IR) injury occurs in many organs and tissues, and contributes to morbidity and mortality worldwide. Melatonin, an endogenously produced indolamine, provides a strong defense against IR injury. Mitochondrion, an organelle for ATP production and a decider for cell fate, has been validated to be a crucial target for melatonin to exert its protection against IR injury. In this review, we first clarify the mechanisms underlying mitochondrial dysfunction during IR and melatonin’s protection of mitochondria under this condition. Thereafter, special focus is placed on the protective actions of melatonin against IR injury in brain, heart, liver, and others. Finally, we explore several potential future directions of research in this area. Collectively, the information compiled here will serve as a comprehensive reference for the actions of melatonin in IR injury identified to date and will hopefully aid in the design of future research and increase the potential of melatonin as a therapeutic agent.     

<Emphasis Type="Italic">Yersinia pseudotuberculosis</Emphasis> supports Th17 differentiation and limits de novo regulatory T cell induction by directly interfering with T cell receptor signaling

Adaptive immunity critically contributes to control acute infection with enteropathogenic Yersinia pseudotuberculosis; however, the role of CD4⁺ T cell subsets in establishing infection and allowing pathogen persistence remains elusive. Here, we assessed the modulatory capacity of Y. pseudotuberculosis on CD4⁺ T cell differentiation. Using in vivo assays, we report that infection with Y. pseudotuberculosis resulted in enhanced priming of IL-17-producing T cells (Th17 cells), whereas induction of Foxp3⁺ regulatory T cells (Tregs) was severely disrupted in gut-draining mesenteric lymph nodes (mLNs), in line with altered frequencies of tolerogenic and proinflammatory dendritic cell (DC) subsets within mLNs. Additionally, by using a DC-free in vitro system, we could demonstrate that Y. pseudotuberculosis can directly modulate T cell receptor (TCR) downstream signaling within naïve CD4⁺ T cells and Tregs via injection of effector molecules through the type III secretion system, thereby affecting their functional properties. Importantly, modulation of naïve CD4⁺ T cells by Y. pseudotuberculosis resulted in an enhanced Th17 differentiation and decreased induction of Foxp3⁺ Tregs in vitro. These findings shed light to the adjustment of the Th17-Treg axis in response to acute Y. pseudotuberculosis infection and highlight the direct modulation of CD4⁺ T cell subsets by altering their TCR downstream signaling.     

基于园林卓越人才培养的观赏植物组织培养课程实践改革

以园林专业卓越人才培养为目标,提出教学过程以应用与创新为导向,更新教学内容体系,探索引入项目式教学形式,调整考核方法.改革后的课程体系侧重学生实践动手能力与创新能力的培养,学生的学习主动性显著提高,在学生中反馈效果好,教学效果显著.     

灰色关联度在甜高粱材料筛选中的应用

对甘肃省兰州市榆中县中试基地种植的11种甜高粱材料进行试验,以筛选出适合在兰州榆中地区种植的甜高粱材料。采用灰色关联度对材料的茎干鲜重、生物产量、茎杆平均含糖量进行分析,同时参考株高、穗长、穗颈长、物候期等指标,选定适合榆中地区不同用途的甜高粱材料。灰色关联度分析结果:茎秆鲜重、生物产量、茎秆平均含糖量三个指标中,茎秆平均含糖量灰色关联度最大,对结果影响最大;灰色综合评判排序显示08-2、2043、09-1、雅津43号分别排列前4位。08-2、2043、09-1、雅津43号综合表现最好,适合兰州榆中地区种植,可用于饲料生产和生物质能源储备种植。     

过江隧道穿越既有铁路相互影响研究

过江隧道穿越既有铁路的相互影响是一个难点.依托重庆长江隧道穿越襄渝铁路工程进行研究,研究结果表明：过江隧道施工对既有铁路两个方向地表沉降的影响都很小,最大值为2.549 mm,故受影响的既有铁路附近地层不需做特殊加固处理,但要加强监控;既有铁路对过江隧道结构安全性及洞周位移影响也较小,在列车荷载作用下结构最小安全系数为6.8,过江隧道结构仍是偏于安全的,故隧道结构可不作加固处理,只需加强监控.研究成果为保证既有铁路的正常运营及过江隧道的结构安全性提供了理论基础,并可为相似工程提供参考.     

Yersinia pestis genome sequencing identifies patterns of global phylogenetic diversity

Plague is a pandemic human invasive disease caused by the bacterial agent Yersinia pestis. We here report a comparison of 17 whole genomes of Y. pestis isolates from global sources. We also screened a global collection of 286 Y. pestis isolates for 933 SNPs using Sequenom MassArray SNP typing. We conducted phylogenetic analyses on this sequence variation dataset, assigned isolates to populations based on maximum parsimony and, from these results, made inferences regarding historical transmission routes. Our phylogenetic analysis suggests that Y. pestis evolved in or near China and spread through multiple radiations to Europe, South America, Africa and Southeast Asia, leading to country-specific lineages that can be traced by lineage-specific SNPs. All 626 current isolates from the United States reflect one radiation, and 82 isolates from Madagascar represent a second radiation. Subsequent local microevolution of Y. pestis is marked by sequential, geographically specific SNPs.     

Granzyme B is recovered by natural killer cells via clathrin-dependent endocytosis

When they recognize a target cell, natural killer (NK) cells mount an attack to kill the target by exerting their cytotoxicity via the exocytosis of cytotoxic granules. Although the details of this process (which includes the movement of cytotoxic granules in the immune synapse and their fusion with the plasma membrane, releasing granzymes and perforin into the synaptic cleft) are relatively better understood, the post-exocytosis regulation of the process is still largely unknown. Here we show that a clathrin-dependent endocytosis stimulated by target cell occurs in NK92 cell line, which is closely correlated with granzyme B recovery. Inhibition of the endocytosis significantly attenuates the cytotoxicity of NK92 cells. The NK cell recovery of its released effector molecules, in turn, suggests that endocytosis may well play a key role in the post exocytosis regulation of immune cells.     

Lysosomal chymotrypsin B potentiates apoptosis via cleavage of Bid

We have reported that chymotrypsin B (CtrB) is not just a digestive enzyme but is also stored in lysosomes. Herein, we demonstrated a broad distribution of CtrB and explored the involvement of CtrB in apoptosis. Exposure of RH-35 cells to H₂O₂ or palmitate induced the redistribution of lysosomal CtrB into the cytoplasm as a result of lysosomal membrane permeabilization (LMP). Suppression of CtrB significantly blocked apoptosis, while overexpression of CtrB sensitized apoptosis markedly. CtrB could cleave Bid under neutral conditions. In RH-35 cells with Bid silenced, apoptosis induced by CtrB protein was attenuated, suggesting that CtrB mediates apoptosis of RH-35 cells mainly through processing Bid. Our data also suggest that LMP occurs earlier than mitochondrial outer membrane permeabilization; Bid activation initiated by caspase-8 might be reinforced by CtrB in consequence of LMP, which causes a positive feedback loop leading to the accumulation of tBid, and results in lysosome- and mitochondrion-dependent apoptosis.
