HAb18G/CD147-mediated calcium mobilization and hepatoma metastasis require both C-terminal and N-terminal domains

HAb18G/CD147 is a heavily glycosylated protein containing two immunoglobulin superfamily domains. Our previous studies have indicated that overexpression of HAb18G/CD147 enhances metastatic potentials in human hepatoma cells by disrupting the regulation of store-operated Ca²⁺ entry by nitric oxide (NO)/cGMP. In the present study, we investigated the structure-function of HAb18G/CD147 by transfecting truncated HAb18G/CD147 fragments into human 7721 hepatoma cells. The inhibitory effect of HAb18G/CD147 on 8-bromo-cGMP-regulated thapsigargin-induced Ca²⁺ entry was reversed by the expression of either C or N terminus truncated HAb18G/CD147 in T7721C and T7721N cells, respectively. The potential effect of HAb18G/CD147 on metastatic potentials, both adhesion and invasion capacities, of hepatoma cells was abolished in T7721C cells, but not affected in T7721N cells. Release and activation of matrix metalloproteinases (MMPs), MMP-2 and MMP-9, were found to be enhanced by the expression of HAb18G/CD147, and this effect was abolished by both truncations. Thapsigargin significantly enhanced release and activation of MMPs (MMP-2 and MMP-9) in non-transfected 7721 cells, and this effect was negatively regulated by SNAP. However, no effects of thapsigargin or SNAP were observed in T7721 cells, and expression of HAb18G/CD147 enhanced secretion and activation of MMPs at a stable and high level. Taken together, these results suggest that both ectodomain and intracellular domains of HAb18G/CD147 are required to mediate the effect of HAb18G/CD147 on the secretion and activation of MMPs and metastasis-related processes in human hepatoma cells by disrupting the regulation of NO/cGMP-sensitive intracellular Ca²⁺ mobilization although each domain may play different roles.Received 1 April 2004; received after revision 15 June 2004; accepted 22 June 2004

Twisted gastrulation can function as a BMP antagonist

Bone morphogenetic proteins (BMPs), including the fly homologue Decapentaplegic (DPP), are important regulators of early vertebrate and invertebrate dorsal-ventral development. An evolutionarily conserved BMP regulatory mechanism operates from fly to fish, frog and mouse to control the dorsal-ventral axis determination. Several secreted factors, including the BMP antagonist chordin/Short gastrulation (SOG), modulate the activity of BMPs. In Drosophila, Twisted gastrulation (TSG) is also involved in dorsal-ventral patterning, yet the mechanism of its function is unclear. Here we report the characterization of the vertebrate Tsg homologues. We show that Tsg can block BMP function in Xenopus embryonic explants and inhibits several ventral markers in whole-frog embryos. Tsg binds directly to BMPs and forms a ternary complex with chordin and BMPs. Coexpression of Tsg with chordin leads to a more efficient inhibition of the BMP activity in ectodermal explants. Unlike other known BMP antagonists, however, Tsg also reduces several anterior markers at late developmental stages. Our data suggest that Tsg can function as a BMP inhibitor in Xenopus; furthermore, Tsg may have additional functions during frog embryogenesis.

光子晶体光纤熔接机理的研究

光子晶体光纤（PCF,Photonic Cpystal Fiber）的熔接技术为PCF产品的应用和开发提供了条件。本文主要介绍了影响PCF熔接的主要因素，比较了传统电弧熔接方法和激光熔接方法的优缺点，阐述了激光熔接的基本原理和工作流程，为PCF激光熔接机的制作打下基础。     

Molecular dynamics simulation and continuum modelling of granular surface flow in rotating drums

Numerical simulations of granular flows in rotating drums operated at medium to high rates (Fr=0.1― 0.2) have been carried out by using a Molecular Dynamics (MD) algorithm that incorporates inelastic particle interactions, sliding friction and rolling friction. The results indicate that the behavior of granular flow in rotating drums can be classified into two distinct zones: a shear active layer at the bed surface and a quasi-static plug flow region adjacent to the wall. The residence time of a tracer particle in the active layer is approximately a third or a half of that in the plug flow region. The thickness of the active layer at mid-chord is about 0.57―0.61 times that of the plug flow region. It is found that all cases simulated in this work are in the rolling-cascading intermediate regime instead of the pure rolling re-gime. The simulated tangential velocity at the mid-chord is also compared with experimental results reported in the literature and good agreement has been obtained. Based on the MD simulations and experimental results, a continuum approach has also been developed. It is shown that the behavior of granular solids in the plug flow region experiences plastic deformation along the radial direction from the wall with the velocity profiles well described by an exponential function, whereas the active layer velocity follows a simple expression for the Couette shear flow. Discussion has also been made on the granular temperature and concentration profiles.
