Cajal bodies (CBs) and Gems are nuclear domains that contain factors responsible for spliceosomal small nuclear ribonucleoprotein (snRNP) biogenesis. The marker protein for CBs is coilin. In addition to snRNPs, coilin and other factors, canonical CBs contain the survivor of motor neuron protein (SMN). SMN can also localize to Gems. Considering the important role that coilin plays in the formation and composition of CBs, we tested the splicing efficiency of several cell lines that vary in regards to coilin level and modification using an artificial reporter substrate. We found that cells with both hypomethylated coilin and Gems are more efficient at reporter splicing compared to cells in which SMN localizes to CBs. In contrast, coilin reduction, which induces Gem formation, decreases cell proliferation and artificial reporter splicing. These findings demonstrate that coilin modifications or levels impact artificial reporter splicing, possibly by influencing snRNP biogenesis. This paper further investigates cluster synchronization in a complex dynamical network with two-cluster. Each cluster contains a number of identical dynamical systems, however, the subsystems composing the two clusters can be different, i.e., the individual dynamical system in one cluster can differ from that in the other cluster. Complete synchronization within each cluster is possible only if each node from one cluster receives the same input from nodes in other cluster. In this case, the stability condition of one-cluster synchronization is known to contain two terms: the first accounts for the contribution of the inner-cluster coupling structure while the second is simply an extra linear term, which can be deduced by the "same-input" condition. Applying the connection graph stability method, the authors obtain an upper bound of input strength for one cluster if the first account is known, by which the synchronizability of cluster can be scaled. For different clusters, there are different upper bound of input strength by virtue of different dynamics and the corresponding cluster structure. Moreover, two illustrative examples are presented and the numerical simulations coincide with the theoretical analysis.  相似文献

This paper formulates a robust stage-structured SI eco-epidemiological model with periodic constant pulse releasing of infectious pests with pathogens. The authors show that the conditions for global attractivity of the 'pest-eradication' periodic solution and permanence of the system depend on time delay, hence, the authors call it "profitless". Further, the authors present a pest management strategy in which the pest population is kept under the economic threshold level (ETL) when the pest population is uniformly persistent. By numerical analysis, the authors also show that constant maturation time delay for the susceptible pests and pulse releasing of the infectious pests can bring obvious effects on the dynamics of system.  相似文献

The authors concern robust model predictive control for linear continuous systems with polytopic uncertainties and input constraints. At each sampling time, a piecewise constant control sequence is obtained by solving a set of linear matrix inequalities. The sufficient conditions on the existence of the model predictive control are given, and the robust stability of the closed-loop systems is guaranteed. This paper deals with the development of scientific policy advice by providing a methodology to foster a social learning process. The methodology, called participatory Sustainability Impact Assessment (pSIA), aims to structure complex problem situations, to clarify interests and basic assumptions, to interpret scientific studies, to develop impact assessment, and to explore sensitivity of uncertainty and lack of information. In pSIA workshops the participants are supposed to build conceptual models with different modelling methods, like System Dynamics, Value Chain Analysis, and Morphological Analysis. A case study is presented that describes a workshop series with political as well as academic actors, applying the pSIA approach to an impact assessment of Economic Partnership Agreements between the European Union and the Eastern and Southern Africa Region.

Bile acids and bile alcohols in the form of their conjugates are amphipathic end products of cholesterol metabolism with multiple physiological functions. The great variety of bile acids and bile alcohols that are present in vertebrates are tabulated. Bile salts have an enterohepatic circulation resulting from efficient vectorial transport of bile salts through the hepatocyte and the ileal enterocyte; such transport leads to the accumulation of a pool of bile salts that cycles between the liver and intestine. Bile salt anions promote lipid absorption, enhance tryptic cleavage of dietary proteins, and have antimicrobial effects. Bile salts are signaling molecules, activating nuclear receptors in the hepatocyte and ileal enterocyte, as well as an increasing number of G-protein coupled receptors. Bile acids are used therapeutically to correct deficiency states, to decrease the cholesterol saturation of bile, or to decrease the cytotoxicity of retained bile acids in cholestatic liver disease.  相似文献

Planarians possess amazing abilities to regulate tissue homeostasis and regenerate missing body parts. These features reside on the presence of a population of pluripotent/totipotent stem cells, the neoblasts, which are considered as the only planarian cells able to proliferate in the asexual strains. Neoblast distribution has been identified by mapping the cells incorporating bromodeoxyuridine, analyzing mitotic figures and using cell proliferation markers. Recently identified molecular markers specifically label subgroups of neoblasts, revealing thus the heterogeneity of the planarian stem cell population. Therefore, the apparent totipotency of neoblasts probably reflects the composite activities of multiple stem cell types. First steps have been undertaken to understand how neoblasts and differentiated cells communicate with each other to adapt the self-renewal and differentiation rates of neoblasts to the demands of the body. Moreover, the introduction of molecular resource database on planarians now paves the way to renewed strategies to understand planarian regeneration and stem cell-related issues.  相似文献

The potential effects of synthetic unmethylated oligodeoxynucleotides (ODN) containing CpG motifs, mimicking bacterial DNA, has never been evaluated on the immune response in the teleost fish gilthead seabream (Sparus aurata), the most important fish species in Mediterranean aquaculture. First, binding and competition studies have demonstrated that binding is saturated and promiscuous, suggesting the participation of several receptors. Moreover, leucocyte cytotoxic (NCC) activity, production of ROIs (reactive oxygen intermediates), and expression of immune-relevant genes was greatly primed by ODNs. Focusing on the mechanism, the TLR9 gene is widely distributed in seabream tissues and differently regulated in vitro by several stimuli. Moreover, and for the first time in fish, TLR9 mRNA has been detected in lymphocytes as the main cell-source. To conclude, ODNs containing GACGTT, GTCGTT (optimal for mouse and human, respectively) or AACGTT motifs are the most potent inducers of seabream immunity, whilst the involvement of TLR9 is under debate.  相似文献

Olfactory ensheathing cells (OECs) have been shown previously to express Toll-like receptors and to respond to bacteria by translocating nuclear factor-kappaB from the cytoplasm to the nucleus. In this study, we show that OECs extended significantly more pseudopodia when they were exposed to Escherichia coli than in the absence of bacteria (p=0.019). Co-immunoprecipitation showed that E. coli binding to OECs was mediated by Toll-like receptor 4. Lyso-Tracker, a fluorescent probe that accumulates selectively in lysosomes, and staining for type 1 lysosome-associated membrane proteins demonstrated that endocytosed FITC-conjugated E. coli were translocated to lysosomes. They appeared to be subsequently broken down, as shown by transmission electron microscopy. No obvious adherence to the membrane and less phagocytosis was observed when OECs were incubated with inert fluorescent microspheres. The ability of OECs to endocytose bacteria supports the notion that OECs play an innate immune function by protecting olfactory tissues from bacterial infection.  相似文献
