During mammalian ontogeny, haematopoietic stem cells (HSCs) translocate from the fetal liver to the bone marrow, where haematopoiesis occurs throughout adulthood. Unique features of bone that contribute to a microenvironmental niche for stem cells might include the known high concentration of calcium ions at the HSC-enriched endosteal surface. Cells respond to extracellular ionic calcium concentrations through the seven-transmembrane-spanning calcium-sensing receptor (CaR), which we identified as being expressed on HSCs. Here we show that, through the CaR, the simple ionic mineral content of the niche may dictate the preferential localization of adult mammalian haematopoiesis in bone. Antenatal mice deficient in CaR had primitive haematopoietic cells in the circulation and spleen, whereas few were found in bone marrow. CaR-/- HSCs from fetal liver were normal in number, in proliferative and differentiative function, and in migration and homing to the bone marrow. Yet they were highly defective in localizing anatomically to the endosteal niche, behaviour that correlated with defective adhesion to the extracellular matrix protein, collagen I. CaR has a function in retaining HSCs in close physical proximity to the endosteal surface and the regulatory niche components associated with it. 相似文献

The upper jaws of birds, unlike those in many tetrapods, move relative to the skull and are often flexible along their length, whereas the lower jaw (mandible) is usually a rigid structure formed by the fusion of several bones, flexing only where it meets the skull. Here we describe a previously unnoticed mandibular bending movement in hummingbirds, in which the distal half of the mandible is actively flexed downwards and the gape widens to catch flying insects. The hummingbird is thought to have developed a long narrow bill as it specialized in feeding on floral nectar, but the bird's need to supplement its diet with insects must have contributed to the surprising flexibility of its jaw. 相似文献

In the central nervous system, ageing results in a precipitous decline in adult neural stem/progenitor cells and neurogenesis, with concomitant impairments in cognitive functions. Interestingly, such impairments can be ameliorated through systemic perturbations such as exercise. Here, using heterochronic parabiosis we show that blood-borne factors present in the systemic milieu can inhibit or promote adult neurogenesis in an age-dependent fashion in mice. Accordingly, exposing a young mouse to an old systemic environment or to plasma from old mice decreased synaptic plasticity, and impaired contextual fear conditioning and spatial learning and memory. We identify chemokines--including CCL11 (also known as eotaxin)--the plasma levels of which correlate with reduced neurogenesis in heterochronic parabionts and aged mice, and the levels of which are increased in the plasma and cerebrospinal fluid of healthy ageing humans. Lastly, increasing peripheral CCL11 chemokine levels in vivo in young mice decreased adult neurogenesis and impaired learning and memory. Together our data indicate that the decline in neurogenesis and cognitive impairments observed during ageing can be in part attributed to changes in blood-borne factors. 相似文献

Amyloid-β oligomers may cause cognitive deficits in Alzheimer's disease by impairing neuronal NMDA-type glutamate receptors, whose function is regulated by the receptor tyrosine kinase EphB2. Here we show that amyloid-β oligomers bind to the fibronectin repeats domain of EphB2 and trigger EphB2 degradation in the proteasome. To determine the pathogenic importance of EphB2 depletions in Alzheimer's disease and related models, we used lentiviral constructs to reduce or increase neuronal expression of EphB2 in memory centres of the mouse brain. In nontransgenic mice, knockdown of EphB2 mediated by short hairpin RNA reduced NMDA receptor currents and impaired long-term potentiation in the dentate gyrus, which are important for memory formation. Increasing EphB2 expression in the dentate gyrus of human amyloid precursor protein transgenic mice reversed deficits in NMDA receptor-dependent long-term potentiation and memory impairments. Thus, depletion of EphB2 is critical in amyloid-β-induced neuronal dysfunction. Increasing EphB2 levels or function could be beneficial in Alzheimer's disease. 相似文献

Hereditary neuralgic amyotrophy (HNA) is an autosomal dominant recurrent neuropathy affecting the brachial plexus. HNA is triggered by environmental factors such as infection or parturition. We report three mutations in the gene septin 9 (SEPT9) in six families with HNA linked to chromosome 17q25. HNA is the first monogenetic disease caused by mutations in a gene of the septin family. Septins are implicated in formation of the cytoskeleton, cell division and tumorigenesis. 相似文献

Distribution of entangled states between distant locations is essential for quantum communication over large distances. But owing to unavoidable decoherence in the quantum communication channel, the quality of entangled states generally decreases exponentially with the channel length. Entanglement purification--a way to extract a subset of states of high entanglement and high purity from a large set of less entangled states--is thus needed to overcome decoherence. Besides its important application in quantum communication, entanglement purification also plays a crucial role in error correction for quantum computation, because it can significantly increase the quality of logic operations between different qubits. Here we demonstrate entanglement purification for general mixed states of polarization-entangled photons using only linear optics. Typically, one photon pair of fidelity 92% could be obtained from two pairs, each of fidelity 75%. In our experiments, decoherence is overcome to the extent that the technique would achieve tolerable error rates for quantum repeaters in long-distance quantum communication. Our results also imply that the requirement of high-accuracy logic operations in fault-tolerant quantum computation can be considerably relaxed. 相似文献

During the two most recent deglaciations, the Southern Hemisphere warmed before Greenland. At the same time, the northern Atlantic Ocean was exposed to meltwater discharge, which is generally assumed to reduce the formation of North Atlantic Deep Water. Yet during deglaciation, the Atlantic thermohaline circulation became more vigorous, in the transition from a weak glacial to a strong interglacial mode. Here we use a three-dimensional ocean circulation model to investigate the impact of Southern Ocean warming and the associated sea-ice retreat on the Atlantic thermohaline circulation. We find that a gradual warming in the Southern Ocean during deglaciation induces an abrupt resumption of the interglacial mode of the thermohaline circulation, triggered by increased mass transport into the Atlantic Ocean via the warm (Indian Ocean) and cold (Pacific Ocean) water route. This effect prevails over the influence of meltwater discharge, which would oppose a strengthening of the thermohaline circulation. A Southern Ocean trigger for the transition into an interglacial mode of circulation provides a consistent picture of Southern and Northern hemispheric climate change at times of deglaciation, in agreement with the available proxy records. 相似文献

Tribolium castaneum is a member of the most species-rich eukaryotic order, a powerful model organism for the study of generalized insect development, and an important pest of stored agricultural products. We describe its genome sequence here. This omnivorous beetle has evolved the ability to interact with a diverse chemical environment, as shown by large expansions in odorant and gustatory receptors, as well as P450 and other detoxification enzymes. Development in Tribolium is more representative of other insects than is Drosophila, a fact reflected in gene content and function. For example, Tribolium has retained more ancestral genes involved in cell-cell communication than Drosophila, some being expressed in the growth zone crucial for axial elongation in short-germ development. Systemic RNA interference in T. castaneum functions differently from that in Caenorhabditis elegans, but nevertheless offers similar power for the elucidation of gene function and identification of targets for selective insect control. 相似文献

RNA-directed DNA methylation (RdDM) is a process in which dicer-generated small RNAs guide de novo cytosine methylation at the homologous DNA region. To identify components of the RdDM machinery important for Arabidopsis thaliana development, we targeted an enhancer active in meristems for methylation, which resulted in silencing of a downstream GFP reporter gene. This silencing system also features secondary siRNAs, which trigger methylation that spreads beyond the targeted enhancer region. A screen for mutants defective in meristem silencing and enhancer methylation retrieved six dms complementation groups, which included the known factors DRD1 (ref. 3; a SNF2-like chromatin-remodeling protein) and Pol IVb subunits. Additionally, we identified a previously unknown gene DMS3 (At3g49250), encoding a protein similar to the hinge-domain region of structural maintenance of chromosomes (SMC) proteins. This finding implicates a putative chromosome architectural protein that can potentially link nucleic acids in facilitating an RNAi-mediated epigenetic modification involving secondary siRNAs and spreading of DNA methylation. 相似文献
