In order to elucidate the role of the hypophysis in the regulation of cholesterol 7alpha-hydroxylase activity, male and female rats hypophysectomized on day 50 of life were treated with different hypophyseal hormones and tested on day 85 of life. Only a crude extract from human hypophysis and ovine prolactin was able to enhance the enzyme activity, thereby restoring the level of intact controls.  相似文献

Immunological properties of ovine chorionic somatomammotropin (OSC) are studied immunodiffusion, radio-immunoassays and by comparative studies of serum hormonal changes during pregnancy. OCS shows no immunological cross-reaction with ovine prolacin, ovine, bovine and human growth hormone and human placental lactogen. Structural analogies of these hormones are discussed.  相似文献

Summary The time requirement is treated for a longitudinal fission by Brownian movement of a very long particle consisting of two or more filaments twisted a great number of times round each other to form a double spiral. It is shown that a comparatively swift disintegration is obtained by partial rotation or torsion round the axis of the spiral, resulting in a loosing of the spiral structure and subsequent separation of the constituents by translational Brownian movement. The time required to separate a double spiral consisting of about 900 turns of a height of 3.4 × 10−7 cm and a radius of 10−7 cm, thus having a length of 3 × 10−4 cm being realized approximately by deoxyribonucleic acid is found by this mechanism to be about 50 to 80 s. The time required to undo the same spiral by unwrapping it turn by turn would be about 150 days. The result of the considerations is related to observations published byAlexander andSteacy on deoxyribonucleic acid. An additional remark stresses the importance of stereochemical asymmetry for the practicability of the mechanism and therefore the importance of optical activity for the time requirement of such disintegrations or transformations of high polymer material occurring in living organisms.   相似文献

Summary The intestine of the ferret (Putorius furo) is unusual in that there is no external anatomical division between ileum and colon. Up to 8–10 cm from the anus the electrical activity was organized into migrating myoelectric complexes typical of the small intestine. At this point the pattern of electrical activity changed abruptly to that characteristic of the colon, namely short and long spike burst activity. Histological examination showed that at this point the muscular layers were interupted by a band of connective tissue sufficient to permit the functional autonomy of the last part of the intestine.Acknowledgments. Rabbit antisera raised against a strain of E. coli 013, with a strong antiglycogen activity, were tested on human fetal and normal adult colons, on colon carcinomas, and on colon tumor cells in culture (HT29). Only very rare granules were present in adult normal colons when tested with the immunofluorescence method. In faetal colons, in 12 out of 14 carcinomas, and on HT29 cells, the immunofluorescent reactions were similar to those observed in normal liver. The reactions were negative after previous treatment with alpha-amylase. They were inhibited with glycogen, with phenol-alcohol, perchloric, and trichloroacetic extracts from faetal colons, and with a tumor trichloroacetic extract. The extracts precipitated with anti-E. coli 013 antisera. They had a strong inhibiting activity in a radioimmunoassay test with labeled glycogen. The extracts from normal adult colons did not precipitate with the antisera and they had no inhibiting activity in either immunofluorescence and radioimmunoassay tests.  相似文献

Several different cell types constitute the intestinal wall and interact in different manners to maintain tissue homeostasis. Elegant reports have explored these physiological cellular interactions revealing that glial cells and neurons not only modulate peristalsis and mechanical stimulus in the intestines but also control epithelial proliferation and sub-epithelial angiogenesis. Although colon carcinoma arises from epithelial cells, different sub-epithelial cell phenotypes are known to support the manifestation and development of tumors from their early steps on. Therefore, new perspectives in cancer research have been proposed, in which neurons and glial cells not only lead to higher cancer cell proliferation at the tumor invasion front but also further enhance angiogenesis and neurogenesis in tumors. Transformation of physiological neural activity into a pro-cancer event is thus discussed for colon carcinogenesis herein.  相似文献

Summary Explants derived from mammary carcinomas of DMBA-treated female Sprague-Dawley rats were cultured for 5 days in Medium 199 containing insulin and corticosterone. The addition of ovine prolactin to the culture media resulted in a consistent significant increase in H3-thymidine incorporation into DNA. DNA synthesis of explants treated with either ovine or human growth hormone was intermediary to prolactin-treated cultures and control cultures. The adrenal weight of the ovine fetus increases dramatically during late pregnancy. This increase is achieved in three periods: one of DNA duplication between two phases of cellular hypertrophy. The continuous cell lines T 24 and HT-29, derived from human bladder and colon carcinomas, produce term-placental and intestinal alkaline phosphatase, respectively. Growth in hyperosmolar medium or exposure to prednisolone or sodium butyrate induces increased enzyme levels, and combinations of inducers elicit synergistic activity increases. The effect of the inducing agents is strikingly diminished when cells are grown in the presence of high concentrations of human serum, and the synergistic increases are essentially abolished. Major human serum protein fractions do not affect alkaline phosphatase induction.  相似文献

Purified gonadotropin subunits of a teleost Fish, the Carp, reassociated in vitro, according to a second order rate reaction. The rate constant was temperature dependent. Compared to ovine LH, the Q10 was about three times less and the rate constant much greater (160 times for instance at 20 degrees C). 目的从人结肠腺癌组织中分离、鉴定结肠癌干细胞,并初步观察其生物学特性。方法利用新鲜结肠腺癌组织,无血清悬浮成球培养,流式细胞检测ESA、CD44表达情况,体外观察其诱导分化及CK20、Muc表达情况,Balb/C小鼠移植观察其成瘤情况。结果从人原代结肠腺癌中分离、纯化EpCAM^high CD44^+结肠癌干细胞,结肠癌原代细胞中EpCAM^highCD44^+细胞比例为1.7%~38%(平均5.4%)。单克隆形成实验证实结肠癌组织中存在肿瘤干细胞。其比例为(2.07±0.11)%,分离获得的EpCAM^highCD44^+细胞能在无血清培养基中“成球”,在血清诱导下能贴壁分化;将EpCAM^highCD44^+细胞移植在Balb/C裸鼠体内,表现出很强的致瘤性,移植瘤中EpCAM^highCD44^+细胞比例为3.6%~43.2%(平均15.2%),所有的移植瘤经组织学测定,均形成腺管样结构,表达结肠特异性分化标志物CK-20、中性上皮粘蛋白(neutral epithelial mucins,Muc)。结论人结肠腺癌组织中存在EpCAM^highCD44^+细胞群,具有和普通干细胞相类似的无限增殖、自我更新和分化能力。  相似文献

The rates of fluid and NaCl absorption are greater in the proximal than in the distal colon. Prolactin treatment caused significant increases in fluid and NaCl absorption in the proximal but not in the distal colon. Occurrence of ovine Chorionic Gonadotropin (oCG) is demonstrated in placenta and amniotic fluid with the use of a radioreceptor assay (corpus luteum membranes) in ewes. Identification of oCG is possible as early as 15th day of pregnancy. It should be secreted at a constant rate, and its maximum concentration is recorded on the 130th day. This hormonal factor might be one of the major components accountable for high progesteronemia observed during the 100 last days of pregnancy.  相似文献

Purple acid phosphatase (PAP), also known as tartrate-resistant acid phosphatase (TRAP), uteroferrin or type 5 acid phosphatase (Acp5) is synthesized as an N-glycosylated monomeric latent precursor, which can be processed by limited proteolysis to a disulfide-linked two-subunit form with increased enzyme activity. In this study, we disclosed that the proteolytically processed two-subunit form constitutes the major PAP/TRAP variant in monocytic cells in spleen, thymus, liver and colon. In addition significant expression of the monomeric PAP/TRAP, indicating a non-enzymatic function, was detected in epithelial cells of colon, lung and kidney. Interestingly, proteolytic processing alone did not activate the enzyme but rendered the enzyme more susceptible to activation by reductants. Thus, beside limited proteolysis, the subcellular redox state could also be a determinant of enzyme action in vivo. The co-localization of PAP/TRAP and the cysteine protease cathepsin L could suggest a role for cathepsin L in the in vivo proteolytic processing of PAP/TRAP in monocytic cells.Received 10 December 2004; received after revision 19 January 2005; accepted 9 February 2005  相似文献

The production of testosterone (measured by radioimmuno-assay) by the 18-day-old mouse fetal testis may be stimulated specifically by ovine LH (1 ng, p less than 0.005) and HCG in organ culture. A stimulation by FSH is observed only with high doses (10 mug, p less than 0.0005). Prolactin and ACTH have no effect. Age-matched fetal pituitaries increase significantly the testosterone production in the culture medium (p less than 0.0005).  相似文献
