Summary It is possible to dissociate the activity of different enzymatic systems of the bacteriumEscherichia coli by the action of high hydrostatic pressures. This activity is studied in the microbes after having been subjected to high hydrostatic pressure. Each enzymatic system is inhibited at different pressures. The enzymes of the bacteria submitted to pressures lower than the inhibiting limit show a residual enzymatic activity; sometimes this activity appears after a phase of lag.  相似文献

Summary The vital importance of the vitamins resides in theircoferment function, which enables them to catalyse enzymatic processes in living tissues. The term coferment is used in this connection to designate organic compounds of relatively low molecular weight, which in conjunction with the specific proteinous apoferments bring about the transformation of definite substrates. The vitamins are, however, catabolized like ordinary substrates when they come into contact with enzymatic systems in which they have no coferment function. This explains why normal life cannot be sustained, if the living organisms do not receive asnutritional factors the vitamins which they cannot synthesize. From a biochemical point of view not only exogenous hypo- or avitaminoses but all pathological changes may be regarded as the sequelae of disturbances in one or several enzymatic systems. The vitamins may exert a regulating influence also in these enzymatic disturbances which are not directly caused by vitamin deficiency. In such cases they will act asrèmedies.  相似文献

Subunit interaction in sorbitol dehydrogenase (SDH) has been studied with in vitro and in silico methods identifying a vital hydrogen-bonding network, which is strictly conserved among mammalian SDH proteins. Mutation of one of the residues in the hydrogen-bonding network, Tyr110Phe, abolished the enzymatic activity and destabilized the protein into tetramers, dimers and monomers as judged from gel filtration experiments at different temperatures compared to only tetramers for the wild-type protein below 307 K. The determined equilibrium constants revealed a large difference in Gibbs energy (8 kJ/mol) for the tetramer stability between wild-type SDH and the mutated form Tyr110Phe SDH. The results focus on a network of coupled hydrogen bonds in wild-type SDH that uphold the protein interface, which is specific and favorable to electrostatic, van der Waals and hydrogen-bond interactions between subunits, interactions that are crucial for the catalytic power of SDH. Summary Diphenylamine (DPA) inhibition of carotenoid biosynthesis with concomitant appearance of detectable amounts of the colourless and fluorescent polyene, phytofluene, is reported inChlorella rubescens Chod.Preliminary negative results with the hydrosoluble DPA-sulphonic acid suggest that anticarotenogenic activity of the hydro-and liposoluble DPA may well be related to the faculty of this molecule to accumulate at the aqueous-lipidic interphases in the cytoplasm where it would interfere with the enzymatic dehydrogenation of the carotenoid precursors through its noted antioxidant properties.  相似文献

Summary Chicken liver fructose 1,6-bisphosphatase is readily immobilized on CNBr-activated Sepharose. The immobilization alters some enzymatic properties. They include broader pH activity curve, loss of activation by K+ or NH 4 + , increased resistance to inactivation by trypsin, decreased sensitivity to AMP inhibition, and loss of cooperative interaction among AMP-binding sites. The immobilized enzyme retains about 38% or 19% of the specific activity of the native enzyme when the activity is measured in the absence or presence of K+, resepctively.This work was supported by grant RR-8006 from the General Research Branch, Division of Research Resources, NIH (USA).  相似文献

Summary Brain homogenates ofLocusta migratoria are found to possess enzyme capable of catalyzing the N-acetylation of tryptamine. A main product of the enzymatic reaction is isolated and identified as N-acetyltryptamine by chromatography analyse. Both insecticides, chlordimeform and lindane, inhibit enzyme activity. A direct correlation between the degree of intoxication and acetylation of tryptamine is described.  相似文献

The Atlantic salmon (Salmo salar) goose-type lysozyme gene was isolated and revealed alternative splicing within exon 2 affecting the signal peptide-encoding region. The lysozyme was produced in Escherichia coli, and the recombinant enzyme showed a high specific lytic activity that was stimulated by low or moderate concentrations of mono- or divalent cations. Relative lytic activities of 70 and 100% were measured at 4°C and 22°C, respectively, and there was no detectable activity at 60°C. However, 30% activity was retained after heating the enzyme for 3 h at 90°C. This unique combination of thermal properties was surprising since the salmon goose-type lysozyme contains no cysteines for protein structure stabilization through disulphide bond formation. The results point to a rapid reversal of inactivation, probably due to instant protein refolding. Summary Different purified polysaccharides, mucopolysaccharides, phosphatides and carbohydrat-sulfonic acids have been investigated for their effect on stimulation of leucocytic migrationin vitro. The bacterial polysaccharides of proteus,S. marcescens, shiga and shiga fullantigen, have been found to be of high specific activity. All other compounds have much less or no activity. Therefore it might be concluded that the type of bacterial polysaccharides which promote leucocytic migration belong to a highly specific group.  相似文献

The digestive tract of worker ants was dissected and the enzymatic activity tested by incubation with different carbohydrates. An RNA hairpin is an essential secondary structure of RNA. It can guide RNA folding, determine interactions in a ribozyme, protect messenger RNA (mRNA) from degradation, serve as a recognition motif for RNA binding proteins or act as a substrate for enzymatic reactions. In this review, we have focused on cis-acting RNA hairpins in metazoa, which regulate histone gene expression, mRNA localization and translation. We also review evolution, mechanism of action and experimental use of trans-acting microRNAs, which are coded by short RNA hairpins. Finally, we discuss the existence and effects of long RNA hairpin in animals. We show that several proteins previously recognized to play a role in a specific RNA stem-loop function in cis were also linked to RNA silencing pathways where a different type of hairpin acts in trans. Such overlaps indicate that the relationship between certain mechanisms that recognize different types of RNA hairpins is closer than previously thought. Summary Ginkgo biloba extract is known to be efficient in diseases associated with free radical generation. The purpose of this work was to study, under in vitro conditions, the action ofGinkgo biloba extract (Gbe) against superoxide anion ( ), which is directly or indirectly implicated in cell damage.Gbe appears to have both an scavenging effect and also a superoxide dismutase activity. Its antiradical effect was demonstrated by low temperature electron spin resonance and in a non-enzymatic system (phenazine methosulfate-NADH), and its enzymatic activity was shown by polarographic determination.  相似文献

Summary Prodigiosin condensing enzyme (PCE) activities were present inSerratia marcescens wild type 08, mutants OF, WF and 9-3-3. Their specific activities exhibited different maxima and at different times during the late log phase or the early stationary phase of cell growth. The levels of prodigiosin and its precursors also showed a significant increase at this period. The results support that prodigiosin and/or its precursors are secondary metabolites. The ubiquity of the PCE activity in mutants deficient in prodigiosin biosynthesis suggest that this particular enzyme may also be present in non-pigmented clinical isolates.  相似文献
