将悬浮于氯化钠磷酸缓冲液(PBS),甘油和山梨醇溶液的人红细胞,在各种温度下处理60分钟,通过扫描电镜观察处理后的红细胞形态变化,同时测定红细胞的溶血作用.实验结果表明,山梨醇溶液悬浮的红细胞在48℃高温处理后,细胞形态仍保持正常状态,溶血作用也与对照细胞相似.而在这温度下处理的PBS和甘油溶液悬浮的人红细胞形态却发生明显改变,溶血作用也显著增大.经比较,在本实验条件下,山梨醇对人红细胞抗热损伤效果比甘油好.另外,本实验还测定了高温处理后人红细胞可滴定巯基和乙酰胆碱酯酶活力.初步讨论了高温下红细胞在不同介质条件下可滴定巯基含量和乙酰胆碱酯酶活力的变化.  相似文献

高温用于恶性肿瘤的治疗是近年来颇受重视的工作,但高温处理不可避免地对肿瘤组织周围正常细胞也将产生影响。为了研究高温对正常细胞的影响,本文以人红细胞为实验模型,研究其细胞热损伤的机制。红细胞经不同温度处理二十分钟后,通过扫描电镜和生化分析方法探讨红细胞的形态变化,溶血作用和膜结合酶-乙酰胆碱酯酶、Na·K·Mg-ATP酶活性的变化。结果表明,在本实验条件下细胞形态的改变对高温最敏感:48℃高温处理后,绝大部分细胞变形,50℃保温使溶血显著增加,膜结合酶热变性温度则超过54℃。  相似文献

生产、科研、生活中,有大量人群长期接触甲醛。甲醛可引起肝脏病变,致癌、致突变,已日益为人们所关注。本文采用人红细胞为材料,研究甲醛的毒害作用。人红细胞悬液,经0.1~1.0×10~(-6)mol/l甲醛处理后,通过生化分析,实验结果表明,在本实试条件下甲醛增强红细胞溶血作用,抑制乙酰胆碱酯酶活力和引起可滴定巯基含量降低。同时使磷酸甘油醛脱氢酶、乳酸脱氢酶活力提高和促进细胞内Ca~(3+)的泄漏。  相似文献

Bapta[1,2-双(邻一氨基酚氧基)乙烷-N,N,N′,N′-四乙酸]酯可以透过细胞膜,近年来已成为研究细胞Ca~(2+)生理功能的有力工具。本文以含1mM Bapta四酸甲酯的人红细胞悬液置于不同温度中保温60分钟,采用扫描电镜观察细胞形态变化,并测定细胞溶血作用,膜结合Na·K-ATP酶活力变化,分析不同温度下Bapta四酸甲酯对人红细胞的影响。实验结果表明,人红细胞形态和溶血作用实验组与对照组的变化相似,而Na·K-ATP酶活力变化实验组的均比对照组的低,其变化最大的为45℃高温处理组,对照组酶活力最高值为50℃处理组。实验结果看出,1mM Bapta四酸甲脂对人红细胞无明显的破坏作用。  相似文献

本文以人红细胞为材料,探讨还原型谷胱甘肽(GSH)对解除甲醛的毒作用效果,实验结果表明5mmol/L甲醛处理人红细胞后,磷酸甘油醛脱氢酶(GPDH)发生的变化与GSH含量的变化呈正相关,同时可滴定巯基含量变化则与Na.K-ATP酶活力变化呈负相关。在受甲醛毒害的红细胞悬液内加入GSH,GPDH活力得到恢复和提高,提示GSH有解除甲醛毒作用的效果。  相似文献

Language is a means of verbal communication. People use language to communicate with each other. In the society, no two speakers are exactly alike in the way of speaking. Some differences are due to age, gender, statue and personality. Above all, gender is one of the obvious reasons. The writer of this paper tries to describe the features of women's language from these perspectives: pronunciation, intonation, diction, subjects, grammar and discourse. From the discussion of the features of women's language, more attention should be paid to language use in social context. 理论推导与室内实验相结合,建立了低渗透非均质砂岩油藏启动压力梯度确定方法。首先借助油藏流场与电场相似的原理,推导了非均质砂岩油藏启动压力梯度计算公式。其次基于稳定流实验方法,建立了非均质砂岩油藏启动压力梯度测试方法。结果表明:低渗透非均质砂岩油藏的启动压力梯度确定遵循两个等效原则。平面非均质油藏的启动压力梯度等于各级渗透率段的启动压力梯度关于长度的加权平均;纵向非均质油藏的启动压力梯度等于各渗透率层的启动压力梯度关于渗透率与渗流面积乘积的加权平均。研究成果可用于有效指导低渗透非均质砂岩油藏的合理井距确定,促进该类油藏的高效开发。  相似文献
