Riassunto Gli estratti di pelle diBufo alvarius, rospo delle regioni desertiche del Nord America, contengono, oltre alle consuete 5-idrossiindolalchilamine (5-HT, N-metil-5-HT, bufotenina) enormi quantitativi di derivati 5-metossiindolici, rappresentati soprattutto daO-metilbufotenina. Questa può giungere a costituire fino il 16% del peso delle ghiandole cutanee secche. Si insiste sulla probabile presenza di 5-idrossiindolo-O-metiltransferasi nella pelle diBufo alvarius.

Advantages ofn-heptanol in the extraction of 5-hydroxytryptamine (5-HT)

Zusammenfassung Mitn-Heptanol als Extraktionsmittel können kleine Mengen von 5-Hydroxytryptamin (5-HT) aus biologischem Material, das 5-Hydroxytryptaphan (5-HTP) in grossen Mengen enthält, extrahiert werden.     

Post mortem increase of 5-hydroxytryptamine in rat brain after 5-hydroxytryptophan administration

Zusammenfassung Ratten mit und ohne Iproniazid-Vorbehandlung erhielten 30 min vor Dekapitation 5-Hydroxytryptophan (5HTP) i. p. Das intakte Hirn wurde bis zu 4 h bei 37° C, Zimmertemperatur und in 0·1n HCl aufbewahrt. In allen Fällen ergab sich eine starke postmortale 5-Hydroxytryptamin (5HT)-Vermehrung im Gehirn.Bei unbehandelten Kontrolltieren zeigte sich postmortal ein leichter 5HT-Abfall im Gehirn, der durch Iproniazid-Vorbehandlung aufgehoben wurde.

Behavioral effects of 5-hydroxytryptophan

Zusammenfassung Bestimmung der Verhaltensänderung bei Tauben, durch Injektion von 5-Hydroxytryptophan, der Vorstufe von Serotonin hervorgerufen. Mit steigender Dosis von 5-Hydroxytryptophan wird eine entsprechende Frequenzverminderung des Pick-Verhaltens ausgelöst.

The diterpenoid alkaloid noroxoaconitine is a Mapkap kinase 5 (MK5/PRAK) inhibitor

The mitogen-activated protein kinase-activated protein kinase MK5 is ubiquitously expressed in vertebrates and is implicated in cell proliferation, cytoskeletal remodeling, and anxiety behavior. This makes MK5 an attractive drug target. We tested several diterpenoid alkaloids for their ability to suppress MK5 kinase activity. We identified noroxoaconitine as an ATP competitor that inhibited the catalytic activity of MK5 in vitro (IC₅₀ = 37.5 μM; K _i = 0.675 μM) and prevented PKA-induced nuclear export of MK5, a process that depends on kinase active MK5. MK5 is closely related to MK2 and MK3, and noroxoaconitine inhibited MK3- and MK5- but not MK2-mediated phosphorylation of the common substrate Hsp27. Molecular docking of noroxoaconitine into the ATP binding sites indicated that noroxoaconitine binds more strongly to MK5 than to MK3. Noroxoaconitine and derivatives may help in elucidating the precise biological functions of MK5 and may prove to have therapeutic values.     

Synthesis of dinucleoside phosphates by reaction of 5′-chloro-5′-deoxynucleosides with nucleotide anions

Zusammenfassung Uridylyl-(3–5)-adenosin und Uridylyl-(3-5)-ionosin wurden durch Umsetzung eines passenden 5-chloro-5-deoxy-2, 3-O-isopropylidene-Nukleosids mit Uridin-3-phosphat synthetisiert.     

CDK5 downregulation enhances synaptic plasticity

CDK5 is a serine/threonine kinase that is involved in the normal function of the adult brain and plays a role in neurotransmission and synaptic plasticity. However, its over-regulation has been associated with Tau hyperphosphorylation and cognitive deficits. Our previous studies have demonstrated that CDK5 targeting using shRNA-miR provides neuroprotection and prevents cognitive deficits. Dendritic spine morphogenesis and forms of long-term synaptic plasticity—such as long-term potentiation (LTP)—have been proposed as essential processes of neuroplasticity. However, whether CDK5 participates in these processes remains controversial and depends on the experimental model. Using wild-type mice that received injections of CDK5 shRNA-miR in CA1 showed an increased LTP and recovered the PPF in deficient LTP of APPswe/PS1Δ9 transgenic mice. On mature hippocampal neurons CDK5, shRNA-miR for 12 days induced increased dendritic protrusion morphogenesis, which was dependent on Rac activity. In addition, silencing of CDK5 increased BDNF expression, temporarily increased phosphorylation of CaMKII, ERK, and CREB; and facilitated calcium signaling in neurites. Together, our data suggest that CDK5 downregulation induces synaptic plasticity in mature neurons involving Ca²⁺ signaling and BDNF/CREB activation.

Quantitative biological determination of 5-hydroxytryptamine

Zusammenfassung Die zwei meistgebrauchten Methoden der biologischen Auswertung von 5-Hydroxytryptamin, nämlich die am isolierten, atropinisierten Rattenuterus bzw. Rattencolon, wurden in bezug auf ihre Eignung für die quantitative Bestimmung verglichen. Für diesen Zweck bedienten wir uns der sogenannten 2 + 2-Dosen-Prozedur. Beide Methoden ergaben statistisch gleichwertige Resultate. Die Unregelmässigkeit der Uteruskontraktionen machen das Testieren öfters unmöglich, was beim Colon sehr selten der Fall ist. Ausserdem ist das Colon empfindlicher als der Uterus. Wegen dieser Vorteile ist das Colon für die quantitative biologische Auswertung von 5-Hydroxytryptamin dem Uterus vorzuziehen.     

5-Hydroxylation of kynurenine in animals

Zusammenfassung 5-Oxykynuramin (Vorläufer von Mausamin aus Maushirn) wurde aus Urin von Huhn und Maus nach Verabreichung von Kynurenin durch Kombination der Ionen-Austausch-Chromatographie mit Papier-Chromatographie isoliert. Durch Vergleich mit dem synthetischen Präparat wurde es papierchromatographisch und durch Spaltung mittels Mäuseleberhomogenat in 4,6-Dioxychinolin und 6-Oxykynurensäure identifiziert.

Some observations on substituted 5-aminothiatriazoles

Zusammenfassung Die IR-Spektren der sogenannten Thiocarbamylazide lassen mit Wahrscheinlichkeit auf Thiatriazole schliessen. Der Mechanismus der Umlagerung zu substituierten Thiazolen wurde aufgeklärt.     

Reserpine and human platelet 5-hydroxytryptamine

Zusammenfassung Nach einmaliger intramuskulärer Injektion von 1 mg Reserpin sinkt beim Menschen der 5-Oxytryptamingehalt der Blutplättchen zwei Tage lang ab, um bis zum neunten Tag auf unbestimmbarem Wert zu verharren. Während dieser Zeit ist die 5-OT-Aufnahmefähigkeit der Plättchen vermindert. Der Oxytryptamingehalt der Plättchen erreicht seinen Ausgangswert erst ungefähr 3 Wochen nach der Reserpingabe.Die Ansicht, dass die Plättchen freies 5-Oxytryptamin aus dem Kreislauf aufnehmen, wäre mit diesem Ergebnis zu vereinbaren. Die Dauer des Effektes könnte bedeuten, dass Reserpin entweder auf Plättchenvorstadien im Knochenmark wirkt oder aber dass die Lebensdauer der Plättchen in Wirklichkeit länger ist, als neuerdings angenommen wird.     

Enzymatic 5-hydroxylation of 3-methoxytyramine

Résumé Les homogénats de foie de rat et de foie de lapin contiennent un systéme enzymatique capable d'hydroxyler en position 5 la 4-hydroxy-3-méthoxy-phénéthylamine. Ce type d'oxydation pourrait représenter une voie biologique pour la transformation de catécholamines en substances analogues à la mescaline.
