抗人体乳腺癌单克隆抗体6C_6杂交瘤细胞株的建立

用乳腺癌患者的原发瘤组织制备的细胞膜抗原免疫BALB/C小鼠,取致敏脾脏细胞与SP_(2/0)骨髓瘤细胞融合。融合率为42％,经克隆化培养后得到一株分泌抗人体乳腺癌单克隆抗体杂交瘤细胞,命名为6C_6。其分泌的抗体和乳腺癌细胞系MCF-7呈阳性反应,与其它5种人癌细胞系,1种人正常细胞系及4种鼠类肿瘤细胞系呈阴性反应。免疫组织化学实验表明:6C_6单克隆抗体(简称6C_6McAb)与乳腺癌的反应多呈阳性(30/36),与7例正常乳腺均呈阴性反应。6C_6McAb类别为IgC_1。其抗体经ZetaPrep大容量离子交换圆盘纯化,对乳腺癌原发瘤组织仍具有较高的活性。     

抗人绒毛膜促性腺激素（HCG）单克隆抗体的研制和特性分析

以HCG为抗原,免疫BALB/c小鼠,取其脾细胞与小鼠Sp2/0—Ag14骨髓瘤细胞融合,经反复筛选和再克隆化,获得8株能稳定分泌抗HCG特异单克隆抗体的杂交瘤细胞株。对其中6个高效价单抗进行了特异性和抗原决定簇检测,选出两个抗相距较远不同决定簇的单抗(2D_4,4F_4)作进一步特性分析。结果显示:免疫球蛋白亚类鉴定,2D_4为IgGl亚类,4F_4为IgG2a亚类;亲和力常数(K_(affi)),2D_4为5.41±0.94×10~(-10)mol/L,4F_4为1.37±0.33×10~(-9)mol/L。证实该两种单克隆抗体可用于研制有关临床诊断试剂盒。     

免疫组化方法检测猪和人皮肤通用抗原标志

目的　探讨猪与人皮肤组织蛋白抗原性的差异。方法　人和小猪全厚正常皮肤作冰冻切片 ,用抗人皮肤组织相关抗原 (HLA ABC ,HLA DR ,Vimentin ,Desmin ,Actin ,FⅧRA ,Cytokeratin)、细胞因子及其受体和胞外基质 (Hyalu ronicacid ,Fibronectin ,Laminin ,TypeⅣcollagen ,Estradiol 17β和Testosterone)及Ⅰ Ⅲ Ⅶ型胶原等抗原成分的单克隆或多克隆抗体进行免疫组化染色 ,比较猪与人皮肤组织抗原性异同。结果　在猪皮肤组织可检测到I Ⅲ Ⅳ Ⅶ型胶原和Hyaluronicacid、Fibronectin、Laminin、FⅧRA、Cytokeratin、IL 6、IL 8、Estradiol 17β、Testosterone及ICAM 1;多克隆抗体阳检率明显高于单克隆抗体 ;其中细胞外基质成分和分子量较小的细胞因子 ,以及结构或功能原始的蛋白分子在猪与人皮肤有较好的同源性 ,而某些蛋白分子 (如细胞因子或激素的受体和淋巴细胞CD分子等 )则多为阴性 ,显示抗体 抗原结合反应的特异性及其种族差异。结论　猪与人皮肤组织抗原之间存在很大的差异 ,猪血管内皮细胞表达人ICAM 1,提示二者粘附分子可能存在物种间的交叉 ,可能与异种皮肤移植难以存活有关。本研究为异种脱细胞真皮基质的临床应用提供了有用的资料     

山慈菇酯提物对4T1乳腺癌免疫微环境的影响

为探究山慈菇酯提物(Cremastra appendiculata,CrAp)体内外抗4T1乳腺癌的作用以及对4T1乳腺癌荷瘤小鼠免疫相关的分子机制的影响.采用四甲基偶氮唑盐微量酶反应比色法(MTT法)检测不同浓度(12500、1250、125、12.5、1.25、0.125μg/mL)的CrAp在24、48、72 h对4T1乳腺癌细胞的增殖抑制作用.100只BABL/c雌性小鼠植瘤成功后,随机分成空白对照组、模型组、阿霉素(adriamycin,ADM)阳性对照组、CrAp组以及CrAp+ADM组,每隔7 d小鼠称重一次,绘制体重曲线;每3 d测量小鼠肿瘤长径及短径,绘制肿瘤曲线;剥离癌组织,计算抑瘤率;取癌组织做苏木精-伊红(hematoxylin-eosin staining,HE)染色;ELISA法检测白介素-2(interleukin-2,IL-2)、肿瘤坏死因子-α(tumor necrosis factor-α,TNF-α)、干扰素-γ(interferon-γ,IFN-γ)和白介素-10(interleukin-10,IL-10)含量的变化.TMT分析CrAp调控与4T1乳腺癌免疫相关的差异蛋白.结果表明:在MTT实验中,不同浓度的CrAp抑制率范围为13.48％ ～74.47％,其中CrAp(125μg/mL)在72 h抑制作用最强,抑制率为74.47％.乳腺癌荷瘤鼠体重明显降低;CrAp能明显增加乳腺癌小鼠的体重(P<0.01),并明显降低乳腺癌肿瘤体积(P<0.01).癌组织HE染色:模型组肿瘤细胞形态完好,数目较多,坏死区域少见;CrAp及CrAp+ADM组肿瘤细胞破裂以及胞核溶解,密度降低,并存在不同程度的死亡区域,且各给药组增加了乳腺癌中免疫细胞的数量.在癌组织中,与模型组相比,各药物组明显升高IL-2和IFN-γ的含量(P<0.01),而明显降低TNF-α和IL-10的含量(P<0.01).在体外,CrAp可抑制4T1乳腺癌细胞的增殖.在体内,CrAp能抑制4T1乳腺癌的增长,增加荷瘤小鼠的体重,改善乳腺癌荷瘤小鼠的生存.CrAp提高癌组织中免疫因子IL-2、IFN-γ的表达而降低TNF-α、IL-10的表达.此外,CrAp可以增强ADM抗4T1乳腺癌及免疫调节作用.     

乳腺癌中Fas抗原与C-erbB-2表达及其意义

目的检测乳腺癌中Fas抗原与C-erbB-2表达,探讨两者的相关性及与临床病理参数的关系.方法应用免疫组化SP法检测51例乳腺癌组织中Fas抗原与C-erbB-2表达,结果进行统计学分析.结果Fas抗原阳性率为56.9%,C-erbB-2阳性率为60.8%.结论Fas抗原与乳腺癌组织学分级、淋巴结转移呈负相关;C-erbB-2与组织学分级、淋巴结转移呈正相关;Fas抗原、C-erbB-2检测对判断乳腺癌预后、淋巴结转移有重要意义.     

VEGF 和 PCNA在乳腺癌组织中表达的意义

为了探讨血管内皮生长因子（VEGF）和增殖细胞核抗原（PCNA）在乳腺癌组织中的表达及其与病理学分级关系，采用免疫组织化学SP法检测102例乳腺癌及其中48例癌旁正常乳腺组织中VEGF和PCNA的表达，实验结果显示VEGF在乳腺癌组织中的阳性表达率为84．31％，癌乳腺组织中则为12．50％，两者间具有高度显著性差异（P＜0．01），PCNA在乳腺癌组织中的阳性表达率为78．43％，癌旁正常乳腺组织中则为8．33％，它们之间的差异有高度显著性（P＜0．01），VEGF和PCNA的阳性表达率与乳腺癌病理学分级呈正相关，说明VEGF和PCNA阳性表达作为乳腺 辅助诊断指标。     

瓜蒌薤白半夏汤对动脉粥样硬化小鼠炎症因子、ICAM-1、VCAM-1表达的影响

目的:观察瓜蒌薤白半夏汤(GXBD)对Apo-E-/-小鼠动脉粥样硬化模型(AS)C反应蛋白(CRP)、白介素-6(IL-6)、肿瘤坏死因子(TNF-α)、细胞间黏附分子-1(ICAM-1)和血管细胞黏附分子-1(VCAM-1)表达的影响,并探讨其可能的机制.方法:以高脂饲料饲喂雄性Apo-E-/-小鼠建立AS模型,将AS模型小鼠随机分为模型对照组(MS)、辛伐他汀组(XFTT)、GXBD高、低剂量组,选取C57BL/6J雄性小鼠作为正常对照组,每组10只,连续灌胃(ig)给药8周.末次给药24h后,检测血清总胆固醇(TC)、甘油三酯(TG)、低密度脂蛋白(LDL-C)、高密度脂蛋白(HDL-C)、及血清CRP、IL-6、TNF-α水平,HE染色观察主动脉组织病理学变化;Western-Blot法检测主动脉VCAM-1和ICAM-1蛋白表达水平.结果:模型组小鼠TC、TG、LDL-C、HDL-C、及血清CRP、IL-6、TNF-α显著高于正常对照组(P0.05),主动脉VCAM-1、ICAM-1蛋白表达显著高于正常对照组(P0.05),模型组小鼠主动脉出现明显粥样硬化斑块;GXBD各组和辛伐他汀组小鼠血脂及血清CRP、IL-6、TNF-α水平显著低于模型组(P0.05),主动脉组织VCAM-1、ICAM-1蛋白表达水平显著低于模型组(P0.05),且主动脉组织粥样硬化病变明显减轻.结论:GXBD对Apo-E-/-小鼠AS病变有较好的治疗作用,其机制与其调节血脂代谢、抑制炎症因子及主动脉组织VCAM-1、ICAM-1蛋白的表达相关.     

新生牛肝中低分子抑瘤物体内抑瘤作用的研究

采用离子交换柱分离分子量小于1.0 kD的新生牛肝提取液,得到9份样品.在体外抑制肿瘤细胞增殖的基础上,选用体外抑瘤明显的离子交换柱层析获得的〈5〉组分进行体内对荷S-180肉瘤小鼠的肉瘤生长抑制实验.并从白细胞数、胸腺指数和脾指数三个角度分析了低分子抑瘤物对荷瘤小鼠免疫功能的影响;同时用斑点印记法,比较荷瘤前后重要脏器和肉瘤细胞内p27/k ip1细胞周期调控蛋白表达水平的变化.实验结果表明低分子新生牛肝提取液和经离子交换柱层析分离得到的〈5〉组分对荷S-180肉瘤小鼠的肉瘤生长有明显的抑制作用,实验同时发现来源于新生牛肝中的低分子抑瘤物,可通过增强荷瘤小鼠的免疫力,升高肉瘤细胞中的p27/k ip1蛋白表达水平的方式抑制肉瘤细胞的增殖,阻碍瘤细胞向正常组织的浸润和转移.     

长爪沙鼠仙台病毒( SeV) 抗体ELISA 检测方法的建立与应用

摘要: 目的建立长爪沙鼠仙台病毒( SeV) 抗体ELISA 检测方法,用于长爪沙鼠体内仙台病毒抗体的检测。方法孵育鸡胚,接种SeV,制备鸡胚尿囊液正常抗原和SeV 特异抗原,滴定酶结合物和抗原最佳工作浓度,并进行特异性、敏感性、精密性、稳定性实验。结果正常抗原、特异抗原和酶结合物最佳工作浓度分别为0. 1 μg /mL、2 μg /mL 和1∶ 5 000; 正常抗原、特异抗原批内变异系数分别为9. 6% 和8. 4%,批间平均变异系数分别为9. 0% 和5. 9%; 检测灵敏度为1∶ 10 240; 与沙鼠小鼠肝炎病毒( MHV) 、小鼠肺炎( PVM) 、呼肠孤病毒3 型( Reo3) 均无交叉反应。稳定性试验相对偏差小于25%。结论建立的ELISA 方法特异性、敏感性强,重复性、稳定性好,检测结果,准确、可靠。可用于长爪沙鼠体内SeV 抗体的检测。     

乳腺癌组织芯片的应用与HER-2/neu的表达

目的 :构建乳腺癌及乳腺癌 -良性病变组织微阵列 ,研究HER 2 /neu在原发性乳腺癌和乳腺良性病变中的表达并探讨HER 2 /neu的判断标准。方法 :制作乳腺癌 -乳腺良性病变组织微阵列 (tissuemicroarray)。用免疫组织化学方法检测HER 2 /neu在 180例乳腺癌 ,32例乳腺良性病变组织中的表达。结果 :构建乳腺癌及乳腺癌 -乳腺良性病变组织微阵列 4个 ,180例乳腺癌中HER 2 /neu阳性率为 37.78% (6 8/ 180 ) ,32例乳腺良性病变不表达。乳腺癌组织中HER 2 /neu的过表达率显著高于乳腺良性病变组织 (P <0 .0 5 )。结论 :乳腺癌 -乳腺良性病变组织微阵列的建立使乳腺癌相关基因及其蛋白产物的筛选工作简便、快捷。HER 2 /neu的过表达与乳腺癌的发生密切相关     

Markedness and its Interpretation in Language Use

Language markedness is a common phenomenon in languages, and is reflected from hearing, vision and sense, i.e. the variation in the three aspects such as phonology, morphology and semantics. This paper focuses on the interpretation of markedness in language use following the three perspectives, i.e. pragmatic interpretation, psychological interpretation and cognitive interpretation, with an aim to define the function of markedness.

Lower Paleozoic oil relationships within Williston Basin, Canada

The Williston Basin is a significant petroleum province, containing oil production zones that include the Middle Cambrian to Lower Ordovician, Upper Ordovician, Middle Devonian, Upper Devonian and Mississippian and within the Jurassic and Cretaceous. The oils of the Williston Basin exhibit a wide range of geochemical characteristics defined as "oil families", although the geochemical signature of the Cambrian Deadwood Formation and Lower Ordovician Winnipeg reservoired oils does not match any "oil family". Despite their close stratigraphic proximity, it is evident that the oils of the Lower Palaeozoic within the Williston Basin are distinct. This suggests the presence of a new "oil family" within the Williston Basin. Diagnostic geochemical signatures occur in the gasoline range chromatograms, within saturate fraction gas chromatograms and biomarker fingerprints. However, some of the established criteria and cross-plots that are currently used to segregate oils into distinct genetic families within the basin do not always meet with success, particularly when applied to the Lower Palaeozoic oils of the Deadwood and Winnipeg Formation.     

低渗透非均质砂岩油藏启动压力梯度研究

理论推导与室内实验相结合,建立了低渗透非均质砂岩油藏启动压力梯度确定方法。首先借助油藏流场与电场相似的原理,推导了非均质砂岩油藏启动压力梯度计算公式。其次基于稳定流实验方法,建立了非均质砂岩油藏启动压力梯度测试方法。结果表明:低渗透非均质砂岩油藏的启动压力梯度确定遵循两个等效原则。平面非均质油藏的启动压力梯度等于各级渗透率段的启动压力梯度关于长度的加权平均;纵向非均质油藏的启动压力梯度等于各渗透率层的启动压力梯度关于渗透率与渗流面积乘积的加权平均。研究成果可用于有效指导低渗透非均质砂岩油藏的合理井距确定,促进该类油藏的高效开发。
