甘蔗组织培养中培养基的优化   总被引:2,自引:2,他引:2  
比较了不同基本培养基 ,不同激素及其不同质量浓度的组合对甘蔗愈伤组织的诱导 ,不定芽的分化及其生根的效果 .结果表明 :MS + 2 ,4 -D 1~ 5mg·L-1培养基均可诱导愈伤组织的增殖 ;MS +KT 1.5~ 2 .0mg·L-1+ 6 -BA 2mg·L-1培养基对愈伤组织分化不定芽的效果最佳 ;1 2MS +NAA 1~ 4mg·L-1培养基对生根较好 .  相似文献

选用观赏凤梨不同品种的老叶鞘、嫩叶鞘、顶芽、侧芽、茎段、叶片等不同组织作外植体 ,接种在诱导培养基为改良 MS+ 2 .0 mg· L- 1的 BA+ 5 .0 mg· L- 1的 2 ,4- D+ 0 .2 mg· L- 1的 IBA,及分化培养基为改良 MS+ 4 .0 mg· L- 1的 BA+ 0 .2 mg·L- 1的 IBA,培养 60 d进行统计 ,老叶鞘的愈伤组织三个品种平均诱导率为 74.6% ,丛生芽的诱导率平均数达 1 7.1 % ,次之是侧芽、顶芽 ,嫩叶鞘、茎段、叶片 ,后者能诱导出愈伤组织 ,不能诱导出丛生芽  相似文献

以一品红幼茎为外植体,接种在培养基(MS+ZT)0.5mg·L-1、NAA0.5mg·L-1、IBA0.5mg·L-1、2,4 D1.0mg·L-1、6 BA1.0mg·L-1诱导愈伤组织,其中以NAA0.5mg·L-1时愈伤组织诱导率最高,为96.4%.将继代培养的愈伤组织,接种到(MS+ZT)1.0mg·L-1+NAA0~2.0mg·L-1、IBA0~2.0mg·L-1、2,4 D0~2.0mg·L-1;(MS+6 BA)1.0mg·L-1+NAA0~2.0mg·L-1、2,4 D0 2.0mg·L-1培养基上,进行胚状体的分化培养,其中以ZT1.0mg·L-1+2,4 D0.5mg·L-1效果最好.  相似文献

报道了火炬姜叶片离体培养的研究结果 ,试验表明 :诱导叶片产生愈伤组织效果较好的激素配比是 6 -BA 0 .5mg·L- 1+ 2 ,4 -D 0 .5mg·L- 1;最适于愈伤组织分化的基本培养基是MS ;6 -BA、ZT和KT均能诱导愈伤组织分化不定芽 ,其中以 6 -BA的效果较好和经济 ;在所试的 6 -BA质量浓度中 ,以 3mg·L- 1的效果较佳 ;添加 80 0mg·L- 1的CH有利于火炬姜愈伤组织的分化 ;生根培养阶段 ,以附加w =0 .15 %的活性炭的 1/ 2MS培养基效果较好 ,生根率达 10 0 % ,且植株生长健壮 ;试管苗移栽在表土上 ,成活率可达 10 0 %  相似文献

曼地亚红豆杉愈伤组织诱导和继代培养中抑制褐化的研究   总被引:15,自引:0,他引:15  
对曼地亚红豆杉茎段和叶片的愈伤组织诱导条件和愈伤组织继代培养条件进行了探讨,同时对愈伤组织继代培养中的褐化问题进行了分析.结果表明:具有较强分生能力的新生茎段和叶片能够得到很高的诱导率,与NAA相比,2,4 D在诱导中作用显著.含有3mg/L2,4 D+0 5mg/LNAA+0 2mg/L6 BA的MS培养基有利于愈伤组织的诱导.在继代培养中,2,4 D比NAA对愈伤组织生长的促进作用显著,6 BA对愈伤组织生长的促进作用极显著.在继代培养中发现在第20天就开始转移继代和提高愈伤组织与培养基接触面的氧气供给能够较好克服褐化.在含有5mg·L-12,4 D5+0 5mg·L-16 BA+1g·L-1AC的B5培养基上,愈伤组织细胞增殖倍数最大,达到4 7,褐化也得到部分控制.  相似文献

比较了不同基本培养基,不同激素及其不同浓度组合对番茄愈伤组织的增殖、不定芽分化及其生根的效果.结果表明:MS+NAA0.02~0.10mg·L-1+6-BA3.0mg·L-1+蔗糖30g·L-1(pH=5.8)均可诱导愈伤组织的增殖,但以MS+6-BA3.0mg·L-1+NAA0.05mg·L-1+蔗糖30g·L-1最佳;MS+IAA0.1mg·L-1+6-BA2.0mg·L-1+蔗糖30g·L-1培养基对愈伤组织分化不定芽的效果最佳;MS+IAA0.1mg·L-1+蔗糖20g·L-1培养基对生根较好.  相似文献

小立碗藓(Physcomitrella patens(Hewd.)B.S.G.)作为独特的模式植物在分子生物学研究中被广泛应用.本实验以改良的Knop培养基为基本培养基,研究添加不同种类和浓度配比的植物激素对小立碗藓愈伤组织诱导及分化的影响.实验结果表明当糖浓度为2%,添加KT 0.05 mg·L-1或添加6-BA 0.05 mg·L-1,在温度(25±1)℃,光照强度3000~4000lx,光照周期12/12 h/d条件下培养,诱导小立碗藓愈伤组织的效果最为理想.诱导愈伤组织分化的实验结果表明未添加任何植物激素的Knop培养基上分化出正常的配子体且数量最多.添加2,4-D或IAA的培养基上能长出较多的原丝体.  相似文献

报道了青天葵叶片离体培养的研究结果 ,试验表明 :青天葵叶片在MS附加BA 0 .3mg·L- 1+ 2 ,4 D 1mg·L- 1的培养基上可产生俞伤组织 ;将愈伤组织转入附加BA和NAA的KC培养基中可产生原球茎 ;原球茎在KC附加BA 2 .5mg·L- 1与NAA 0 .2mg·L- 1的培养基上可形成走茎 ;将走茎转入附加NAA 3mg·L- 1的H培养基中 ,可形成大量新的球茎 ;新的球茎转入KC附加w =0 .1%的活性炭和 φ =10 %的椰子水的培养基中 ,可形成完整植株 ;试管苗移栽在椰糠上成活率可达 83.3% .  相似文献

以何首乌为材料, 叶片愈伤组织为外植体, 采用L9 (34)正交设计, 研究不同因素对降低愈伤组织褐化的作用. 3因素为基本培养基(A)、培养基中活性炭(B)和硝酸银(C)浓度, 每因素设3个水平, A分别为MS, 1/2 MS和1/4 MS无机盐浓度, B分别为0 g·L-1, 5 g·L-1和10 g·L-1, C分别为5 mg·L-1, 0 mg·L-1和10 mg·L-1.外植体接种在不同的MS基本培养基中(含2,4 D 1 mg·L-1、琼脂8 g·L-1、蔗糖25 g·L-1, pH=5 8), 于(25±1) ℃、2 000 lx光照培养, 30 d后统计愈伤组织的褐化率. 试验结果表明: 降低愈伤组织褐化的最优水平组合为A2B3C1, B对试验结果的影响达显著水平(p<0 05). 以1/2 MS为基本培养基, 并于1/2 MS+活性炭10 g·L-1+AgNO3 5 mg·L-1培养基(附含2,4 D 1 mg·L-1)中培养, 褐化率可降至13 9%.  相似文献

以蕨类植物全缘贯众根茎、叶片和叶柄为外植体诱导愈伤组织,选取诱导率最高的根茎愈伤组织为材料诱导分化形成幼叶,继续培养成生根试管苗,建立起全缘贯众的无性系.结果表明,1/2 MS NH4H2PO4(100 mg·L-1) BA(0.5 mg·L-1) 2,4-D(1.2 mg·L-1) NAA(1.2 mg·L-1)是诱导根茎愈伤组织的最佳培养基;愈伤组织分化形成幼叶的最佳培养基是1/2 MS BA(0.2 mg·L-1) NAA(0.1 mg·L-1);试管苗生根培养的最佳培养基是1/3 MS IAA(0.6 mg·L-1);移栽后根茎上的须根比对照野生苗增加1.5~2.5倍.  相似文献

Language is a means of verbal communication. People use language to communicate with each other. In the society, no two speakers are exactly alike in the way of speaking. Some differences are due to age, gender, statue and personality. Above all, gender is one of the obvious reasons. The writer of this paper tries to describe the features of women's language from these perspectives: pronunciation, intonation, diction, subjects, grammar and discourse. From the discussion of the features of women's language, more attention should be paid to language use in social context. 理论推导与室内实验相结合,建立了低渗透非均质砂岩油藏启动压力梯度确定方法。首先借助油藏流场与电场相似的原理,推导了非均质砂岩油藏启动压力梯度计算公式。其次基于稳定流实验方法,建立了非均质砂岩油藏启动压力梯度测试方法。结果表明:低渗透非均质砂岩油藏的启动压力梯度确定遵循两个等效原则。平面非均质油藏的启动压力梯度等于各级渗透率段的启动压力梯度关于长度的加权平均;纵向非均质油藏的启动压力梯度等于各渗透率层的启动压力梯度关于渗透率与渗流面积乘积的加权平均。研究成果可用于有效指导低渗透非均质砂岩油藏的合理井距确定,促进该类油藏的高效开发。  相似文献
