The schizophrenia susceptibility gene, Rgs4, is one of the most intensively studied regulators of G-protein signaling members, well known to be fundamental in regulating neurotransmission. However, little is known about its role in the developing nervous system. We have isolated zebrafish rgs4 and shown that it is transcribed in the developing nervous system. Rgs4 knockdown did not affect neuron number and patterning but resulted in locomotion defects and aberrant development of axons. This was confirmed using a selective Rgs4 inhibitor, CCG-4986. Rgs4 knockdown also attenuated the level of phosphorylated-Akt1, and injection of constitutively-activated AKT1 rescued the motility defects and axonal phenotypes in the spinal cord but not in the hindbrain and trigeminal neurons. Our in vivo analysis reveals a novel role for Rgs4 in regulating axonogenesis during embryogenesis, which is mediated by another schizophrenia-associated gene, Akt1, in a region-specific manner. 相似文献

本文提出一个亚90nm沟道MOSFET在亚阈值状态下的二维电势和阈值电压的半解析模型.文章首先根据短沟道MOSFET在亚闽值状态下的物理模型提出定解问题,然后用特征函数将由氧化层和空间电荷区衔接条件所得到的超越方程组作正交展开,得到关于未知量的线性代数方程组.求出了氧化层和空间电荷区的二维电势、耗尽层厚度和阈值电压的表达式.该模型不需要适配参数,运算量小,避免了方程离散化,计算精度与数值解精度相同.文章给出了沟道长度为90nm以下MOSFET的电势分布、表面势、耗尽层厚度和阈值电压计算结果.计算值与二维数值模拟值高度吻合. 相似文献

Curcumin, a natural polyphenol, has been described to exhibit effects on signaling pathways, leading to induction of apoptosis.
In this study, we observed that curcumin inhibited Hsp90 activity causing depletion of client proteins implicated in survival
pathways. Based on this observation, this study was designed to investigate the cellular effects of curcumin combination with
the pan-HDAC inhibitors, vorinostat and panobinostat, which induce hyperacetylation of Hsp90, resulting in inhibition of its
chaperone function. The results showed that, at subtoxic concentrations, curcumin markedly sensitized tumor cells to vorinostat-
and panobinostat-induced growth inhibition and apoptosis. The sensitization was associated with persistent depletion of Hsp90
client proteins (EGFR, Raf-1, Akt, and survivin). In conclusion, our findings document a novel mechanism of action of curcumin
and support the therapeutic potential of curcumin/HDAC inhibitors combination, because the synergistic interaction was observed
at pharmacologically achievable concentrations, which were ineffective when each drug was used alone. 相似文献

In vascular smooth muscle cells, IGF-I stimulates SHPS-1/SHP2/Src complex formation which is required for IGF-I-stimulated cell proliferation. Using SHP2/Src silencing and a Pyk2/Y402F mutant, we showed that Pyk2 was also recruited to the SHPS-1 complex. Pyk2 recruitment to SHPS-1 is mediated via the interaction of Pyk2 Tyr402 and the Src in response to IGF-I. Following Src/Pyk2 association, Src phosphorylates Pyk2 on Tyr881 providing a binding site for Grb2. Cells expressing Pyk2/Y881F showed decreased Grb2 recruitment to SHPS-1 and impaired Shc/Grb2 association. This change led to reduced Erk1/2 (MAP kinase) activation and cell proliferation in response to IGF-I. Our results show that, following its recruitment to the SHPS-1 signaling complex, Pyk2 localizes Grb2 in close proximity to Shc thereby facilitating Shc/Grb2 association which leads to Erk1/2 activation in response to IGF-I. Thus, Pyk2 recruitment to SHPS-1 plays an important role in regulating the IGF-I-stimulated mitogenic response. 相似文献

Here we show that a small GTPase, Rab32, is a novel protein required for the formation of autophagic vacuoles. We found that
the wild-type or GTP-bound form of human Rab32 expressed in HeLa and COS cells is predominantly localized to the endoplasmic
reticulum (ER), and overexpression induces the formation of autophagic vacuoles containing an autophagosome marker protein
LC3, the ER-resident protein calnexin and endosomal/lysosomal membrane protein LAMP-2, even under nutrient-rich conditions.
The recruitment of Rab32 to the ER membrane was necessary for autophagic vacuole formation, suggesting involvement of the
ER as a source of autophagosome membranes. In contrast, the expression of the inactive form of, or siRNA-specific for, Rab32
caused the formation of p62/SQSTM1 and ubiquitinated protein-accumulating aggresome-like structures and significantly prevented
constitutive autophagy. We postulate that Rab32 facilitates the formation of autophagic vacuoles whose membranes are derived
from the ER and regulates the clearance of aggregated proteins by autophagy.

Proof for the role of triacylglycerol-rich lipoproteins (TRLs) in the development of cardiovascular events is accumulating. We recently reported that postprandial TRLs bind to and internalize into human aortic vascular smooth muscle cells (HA-VSMCs) by a lipid-dependent mechanism. We now show that postprandial TRLs triggered hydrolysis of sphingomyelin and stimulation of the sphingosine kinase producing sphingosine 1-phosphate (S1P). In addition, postprandial TRLs exhibited survival and mitogenic effects. Interestingly, the signals were modulated by the nature of the fatty acids located at the sn-2 position in the triacylglycerol molecules of TRL. This lipid-stereospecific regulation of S1P cellular levels in HA-VSMCs provides a novel insight into the intrinsic role of dietary fatty acids and the mechanism mediated by triacylglycerol-containing postprandial lipoproteins in the pathogenesis of atherosclerosis.Received 14 August 2003; received after revision 8 October 2003; accepted 15 October 2003 相似文献

Sprouty2 is an important inhibitor of cell proliferation and signal transduction. In this study, we found a bimodal expression
of Sprouty2 protein during cell cycle progression after exit from quiescence, whereas elevated Sprouty4 expression in the
G1 phase stayed high throughout the rest of the cell cycle. Induction of the mitogen-activated protein kinase via activated
Ras was crucial for increased Sprouty2 expression at the G0/G1 transition. Following the first peak, accelerated proteasomal
protein degradation caused a transient attenuation of Sprouty2 abundance during late G1. Since the decline in its expression
was abolished by dominant negative c-Cbl and the timely restricted interaction between Sprouty2 and c-Cbl disappeared at the
second peak of Sprouty2 expression, we conclude that the second phase in the cell cycle-specific expression profile of Sprouty2
is solely dependent on ubiquitination by c-Cbl. Our results suggest that Sprouty2 abundance is the result of strictly coordinated

Selenocystine (SeC), a naturally occurring selenoamino acid, has been shown to be a novel compound with broad-spectrum anticancer activity. In this study, we showed that SeC triggered time- and dose-dependent apoptosis in A375 human melanoma cells by activating the mitochondria-mediated and death receptor-mediated apoptosis pathways. Pretreatment of cells with a general caspase inhibitor z-VAD-fmk significantly prevented SeC-induced apoptosis. A375 cells exposed to SeC showed an increase in levels of total p53 and phosphorylated p53 (serine-15). Silencing of p53 expression with RNA interference significantly suppressed SeC-induced p53 phosphorylation, caspase activation and apoptotic cell death. Moreover, generation of reactive oxygen species and subsequent induction of DNA strand breaks were found to be upstream mediators of p53 activation induced by SeC. In a nude mice xenograft experiment, SeC significantly inhibited the tumor growth of A375 cells via induction of apoptosis. Taken together, these results suggest the potential applications of SeC in cancer chemoprevention. 相似文献

Summary Polarized UV light from a platform on the ground can attract backswimmers and elicit plunge reactions. Threshold intensity is independent of the overall UV-intensity in the surrounding space. Unpolarized UV light attracts the animals only when the intensity is higher than that of the surroundings. Menkes disease (MD) is an X-linked recessive disorder characterized by copper deficiency resulting in a diminished function
of copper-dependent enzymes. Most MD patients die in early childhood, although mild forms of MD have also been described.
A diversity of mutations in the gene encoding of the Golgi-resident copper-transporting P1B-type ATPase ATP7A underlies MD. To elucidate the molecular consequences of the ATP7A mutations, various mutations in ATP7A associated with distinct phenotypes of MD (L873R, C1000R, N1304S, and A1362D) were analyzed in detail. All mutants studied
displayed changes in protein expression and intracellular localization parallel to a dramatic decline in their copper-transporting
capacity compared to ATP7A the wild-type. We restored these observed defects in ATP7A mutant proteins by culturing the cells
at 30°C, which improves the quality of protein folding, similar to that which as has recently has been demonstrated for misfolded
ATP7B, a copper transporter homologous to ATP7A. Further, the effect of the canine copper toxicosis protein COMMD1 on ATP7A
function was examined as COMMD1 has been shown to regulate the proteolysis of ATP7B proteins. Interestingly, in addition to
adjusted growth temperature, binding of COMMD1 partially restored the expression, subcellular localization, and copper-exporting
activities of the ATP7A mutants. However, no effect of pharmacological chaperones was observed. Together, the presented data
might provide a new direction for developing therapies to improve the residual exporting activity of unstable ATP7A mutant
proteins, and suggests a potential role for COMMD1 in this process. 相似文献
