Summary Rat calcitonin gene related peptide (CGRP) and salmon calcitonin (CT) stimulated adenylate cyclase activity in a dose-dependent manner in the rat diaphragm and in the kidney. The ED50 value of rat CGRP was lower and that of salmon CT was higher in the diaphragm than in the kidney. These results suggest that CGRP stimulates adenylate cyclase activity in the striated muscle by reacting with sites distinct from the site in the kidney.  相似文献

Summary Beta-adrenergic-associated cyclic AMP accumulation was studied in intact lymphocytes before and after transformation with Epstein-Barr virus into immortal cell lines. Although a marked reduction in isoproterenol-stimulated cyclic AMP synthesis was observed in transformed cells, forskolin-stimulated cyclic AMP accumulation was preserved. A parallel loss of125-iodocyanopindolol binding sites suggests that the reduction in beta-adrenergic-stimulated AMP synthesis is due to receptor down regulation.  相似文献

Summary Theophylline (0.01–10.0 mM) did not incease but rather decreased adenylate cyclase activity (AC) of guineapig auricles. A serotonine-sensitive adenylate cyclase system has been observed in horse brain membrane preparations. In crude mitochondrial fraction two types of activation sites were characterized, the Ka being = 2 nM and 1 micrometer. In purified synaptosomal membranes, a single adenylate cyclase activation was observed corresponding to the highest apparent affinity (KD = 2nM). This activation site might be related to the high affinity binding site (KD = 2NM) were previously described in the same membrane preparations.  相似文献

Summary Escherichia coli endotoxin inhibited the fluoride-stimulated adenylate cyclase activity of liver plasma membranes enriched with bile canaliculi. Inhibition was of a mixed competitive and uncompetitive type. Subsequently, muscle and sarcomere lengths decreased to less than control values. The mitochondria incubated in the presence of glutamine and KCN formed both ammonia and glutamate in equivalent amounts. Receptors for the Vasoactive Intestinal Peptide (VIP) were characterized in particles enriched in plasma membranes obtained from a human prolactin-secreting pituiatry tumor. Native VIP inhibited competitively the binding of 125I-VIP to the particles and stimulated cyclic AMP production; both these effects were observed at concentrations of VIP as low as 10(-11)-10(-10) M, which are compatible with VIP concentrations in the hypothalamopituitary portal blood.  相似文献
