Calcitonin gene related peptide stimulates adenylate cyclase activity in rat striated muscle

Summary Rat calcitonin gene related peptide (CGRP) and salmon calcitonin (CT) stimulated adenylate cyclase activity in a dose-dependent manner in the rat diaphragm and in the kidney. The ED₅₀ value of rat CGRP was lower and that of salmon CT was higher in the diaphragm than in the kidney. These results suggest that CGRP stimulates adenylate cyclase activity in the striated muscle by reacting with sites distinct from the site in the kidney.

Adenylate cyclase activity in rat striatum decreased post mortem. Half-lives were about 2.7 h at 22 degrees C, 72 h at 4 degrees C. Differences in stability after death of adenylate cyclase in human brain and rat striatum, and possible heterogenity of the enzyme, are briefly discussed.

Summary Adenylate cyclase responses of brain cortex and neostriatum to noradrenaline and dopamine are increased in rats with jaundice in proportion to its severity. In comparing the situation with uraemia, the relevance of phenols has to be considered.

Beta-adrenergic-stimulated adenylate cyclase activity in normal and EBV-transformed lymphocytes

Summary Beta-adrenergic-associated cyclic AMP accumulation was studied in intact lymphocytes before and after transformation with Epstein-Barr virus into immortal cell lines. Although a marked reduction in isoproterenol-stimulated cyclic AMP synthesis was observed in transformed cells, forskolin-stimulated cyclic AMP accumulation was preserved. A parallel loss of¹²⁵-iodocyanopindolol binding sites suggests that the reduction in beta-adrenergic-stimulated AMP synthesis is due to receptor down regulation.

Effect of theophylline on myocardial adenylate cyclase activity

Summary Theophylline (0.01–10.0 mM) did not incease but rather decreased adenylate cyclase activity (AC) of guineapig auricles. Calcitonin stimulation of pyrophosphatase activity in rat bone

Zusammenfassung Calcitonin als neues kalzium- und phosphor-senkendes Hormon wurde auf seine Wirkung auf die inorganische Pyrophosphataseaktivität von Knochen untersucht. Eine Steigerung der alkalinen Pyrophosphataseaktivität wurde vor der starken Senkung des Serum-Kalziums beobachtet.     

5-hydroxytryptamine-sensitive adenylate cyclase activity in brain synaptosomal membrane preparations]

A serotonine-sensitive adenylate cyclase system has been observed in horse brain membrane preparations. In crude mitochondrial fraction two types of activation sites were characterized, the Ka being = 2 nM and 1 micrometer. In purified synaptosomal membranes, a single adenylate cyclase activation was observed corresponding to the highest apparent affinity (KD = 2nM). This activation site might be related to the high affinity binding site (KD = 2NM) were previously described in the same membrane preparations.     

Endotoxin inhibits the fluoride-stimulated adenylate cyclase activity of rat liver plasma membranes enriched with bile canaliculi

Summary Escherichia coli endotoxin inhibited the fluoride-stimulated adenylate cyclase activity of liver plasma membranes enriched with bile canaliculi. Inhibition was of a mixed competitive and uncompetitive type. This effect, which may result from changes of membrane organization, may have relevance in the understanding of endotoxin-cell membrane interactions.     

Tendon shortening in striated muscle.

Tendon shortening in the digastric muscle of the rabbit resulted in a short term increase in gross muscle length and sarcomere length. Subsequently, muscle and sarcomere lengths decreased to less than control values. Long-term measurements suggested that a return to control sarcomere length may have been achieved by a reduction in sarcomere number.

Glutaminase activity in rat skeletal muscle mitochondria

Isolated rat skeletal muscle mitochondria took up about 40-ng-atoms O per mg protein, with glutamine as the only respiratory substrate. The mitochondria incubated in the presence of glutamine and KCN formed both ammonia and glutamate in equivalent amounts. The experiments reported here provide suggestive evidence that rat skeletal muscle mitochondria contain glutaminase (L-glutamine amidohydrolase EC 3.5.1.2.) activity.

Vasoactive intestinal peptide (VIP): specific receptors and adenylate cyclase activation in a human prolactin-secreting pituitary tumor]

Receptors for the Vasoactive Intestinal Peptide (VIP) were characterized in particles enriched in plasma membranes obtained from a human prolactin-secreting pituiatry tumor. Native VIP inhibited competitively the binding of 125I-VIP to the particles and stimulated cyclic AMP production; both these effects were observed at concentrations of VIP as low as 10(-11)-10(-10) M, which are compatible with VIP concentrations in the hypothalamopituitary portal blood.
