Anti-H IV-1 Activities of 4 Telomerase Restrictors

MTT Cell Proliferation Assay was used to optimize the concentration of Telomerase Restrictors（TRs） with minimum toxicity to the selected cells. FACSort flow cytometer and Innotest P24 HIV（Human immtmodeficiency Virus） antigen mAb ELISA Kit were used to investigate the anti-HIV-1 activities of TRs. The results showed that TRs had low cytotoxicity to the PBMC （Peripheral Blood mononuclear cells） and CEM/GFP if the concentration of TRs was at 50μmol/L or below, and the supernatant from PBMC pretreated with SHIV and TR1-001 /TR1-002 could not infect the PBMC, while can infect the C8166 with reduced infectivity, which suggested that the TRs may be one of the novel resources for screening anti-HIV-1 agents.

HER2 over-expression and response to different chemotherapy regimens in breast cancer

Purpose： To exam the relationship between HER2 over-expression and different adjuvant chemotherapies in breast cancer. Patients and Methods： A total of 1625 primary breast cancer patients who received post-surgery adjuvant chemotherapy in Tianjin Cancer Hospital, China, from July 2002 to November 2005 were included in the study. Among them, 600 patients were given CMF （CTX＋MTX＋5-Fu） regimen, 600 given CEF （CTX＋E-ADM＋5-Fu） regimen, and 425 given anthracyclines plus taxanes regimen, with mean follow-up time of 42 months. Results： In CMF treatment group, the 3-year disease free survival （DFS） in HER2 over-expressed patients was lower than that of the HER2-negative ones （89.80% vs 91.24%, P=0.0348）; in node-positive subgroup, the 3-year DFS was 84.72% in HER2 over-expressed patients, and 90.18% in the HER-2-negative ones （P=0.0271）. Compared to CMF regimen, anthracyclines and anthracyclines plus taxanes regimens are more effective （P〈0.05） in node-positive HER2 over-expression than those in the node-negative. Conclusion： HER2 over-expression is an independent index for predicting poor prognosis and short DFS for breast cancer patients. HER2 over-expressed patients are resistant to CMF regimen chemotherapy, but sensitive to anthracyclines-based or anthracyclines plus taxanes regimen. HER2 expression can be taken as a marker for therapies in breast cancer.     

Molecular epidemiology and evolution of worldwide enterovirus 71 strains isolated from 1970 to 2004

Human enterovirus 71 （EV71） is one of the major etiological agents of the hand-foot-and-mouth disease （HFMD） that often causes severe neurological complications. Recently, its outbreaks mainly take place in the torrid zone of the Asia-Pacific region. To study the evolution and genetic variability, we collected 532 EV71 strains with almost complete or complete VP1 sequences （891 nt） isolated worldwide from 1970 to 2004. The pairwise homologies and genetic distances were analyzed. Most strains belong to previously identified genotype B and C. However, a unique strain R13223-1ND-01 appears not to fall into current three genotypes （A, B and C）, and probably represents a new genotype D. Some orphan strains were observed in the genotypes B and C, and their significance in the EV71 evolution was discussed. Moreover, there is a significant co-variance of 6 discrete positions on VP1 （amino acid 43, 58, 164, 184, 240 and 249）. This high co-variability is tightly related with the subgenotypes.     

Construction and visualization of high-resolution three-dimensional anatomical structure datasets for Chinese digital human

The objective of the China Digital Human Project （CDH） is to digitize and visualize the anatomical structures of human body. In the project, a database with information of morphology, physical characteristics and physiological function will be constructed. The raw data of CDH which was completed in the Southern Medical University is employed. In Huazhong University of Science and Technology （HUST）, the frozen section images are preprocessed, segmented, labeled in accordance with the major organs and tissues of human beings, and reconstructed into three-dimensional （3D） models in parallel on high performance computing clusters （HPC）. Some visualization software for 2D atlas and 3D models are developed based on the new dataset with high resolution （0.1 mm× 0.1 mm × 0.2 mm）. In order to share, release and popularize the above work, a website （www.vch.org.cn） is online. The dataset is one of the most important parts in the national information database and the medical infrastructure.
