The peroxisomal protein import machinery displays remarkable properties. Be it its capacity to accept already folded proteins
as substrates, its complex architecture or its energetics, almost every aspect of this machinery seems unique. The list of
unusual properties is still growing as shown by the recent finding that one of its central components, Pex5p, is transiently
monoubiquitinated at a cysteine residue. However, the data gathered in recent years also suggest that the peroxisomal import
machinery is not that exclusive and similarities with p97/Cdc48-mediated processes and with multisubunit RING-E3 ligases are
starting to emerge. Here, we discuss these data trying to distill the principles by which this complex machinery operates.

Intrinsically disordered proteins (IDPs) do not, by themselves, fold into a compact globular structure. They are extremely dynamic and flexible, and are typically involved in signalling and transduction of information through binding to other macromolecules. The reason for their existence may lie in their malleability, which enables them to bind several different partners with high specificity. In addition, their interactions with other macromolecules can be regulated by a variable amount of chemically diverse post-translational modifications. Four kinetically and energetically different types of complexes between an IDP and another macromolecule are reviewed: (1) simple two-state binding involving a single binding site, (2) avidity, (3) allovalency and (4) fuzzy binding; the last three involving more than one site. Finally, a qualitative definition of fuzzy binding is suggested, examples are provided, and its distinction to allovalency and avidity is highlighted and discussed. 相似文献

This examination of an important paper by Sydney Ross is the first in a projected series of occasional reflections on ‘Annals of Science Classic Papers’ that have had enduring utility within the field of history of science and beyond. First the messages of the paper are examined, some well known but others, particularly Ross's own contemporary concerns about the use of the word ‘scientist’, less so. The varied uses made of the paper by scholars are then traced before Ross's biography is examined in order to try to understand how a figure professionally marginal to the field of history of science came to write such a significant piece. Ross’s interest in the topic appears to have been informed by a romantically tinged scientific progressivism and a deep concern with the importance of linguistic precision in science and in public affairs. The inspirations of the author and the interests of his audience have been only partially aligned, but the paper's insights remain of broad historical interest and have wider ramifications since the denotation ‘scientist’ and its proper application are much debated today in contests over the authority of science. 相似文献

Summary Arylsulphatase, -1,3 glucanase, phosphatase and urease responded differently to -irradiation (5–50 Mrad) in air-dried and moist soils. In all instances phosphatase was the most stable. Summary The i.v. injection of bacterial endotoxin into dogs was found to cause a rapid increase in plasma levels of infused -aminoisobutyric acid. Parkinson’s disease (PD) is a progressive neurodegenerative disorder implicitly marked by the substantia nigra dopaminergic neuron degeneration and explicitly characterized by the motor and non-motor symptom complexes. Apart from the nigrostriatal dopamine depletion, the immune and endocrine study findings are also frequently reported, which, in fact, have helped to broaden the symptom spectrum and better explain the pathogenesis and progression of PD. Nevertheless, based on the neural, immune, and endocrine findings presented above, it is still difficult to fully recapitulate the pathophysiologic process of PD. Therefore, here, in this review, we have proposed the neuroimmunoendocrine (NIE) modulatory network in PD, aiming to achieve a more comprehensive interpretation of the pathogenesis and progression of this disease. As a matter of fact, in addition to the classical motor symptoms, NIE modulatory network can also underlie the non-motor symptoms such as gastrointestinal, neuropsychiatric, circadian rhythm, and sleep disorders in PD. Moreover, the dopamine (DA)–melatonin imbalance in the retino-diencephalic/mesencephalic-pineal axis also provides an alternative explanation for the motor complications in the process of DA replacement therapy. In conclusion, the NIE network can be expected to deepen our understanding and facilitate the multi-dimensional management and therapy of PD in future clinical practice. 相似文献

Ubiquitination has emerged over the years as the most sophisticated way to modify proteins to affect their fate and function.
In particular, it has been reported to be instrumental in regulating several steps of the NF-κB signalling pathway which controls
inflammation, immunity, adhesion and cell survival. Integrating ubiquitination into NF-κB activation requires the regulatory
subunit of IKK, NEMO, which not only displays affinity for polyubiquitin chains, but is also posttranslationally modified
by a complex set of reactions involving ubiquitin. Here, we examine how studies of the NEMO/ubiquitin relationship have provided
novel insights into the IKK activation process and have uncovered molecular mechanisms that should represent in the future

In this last decade, the structure and functions of the receptor for the urokinase-type plasminogen activator have been extensively
studied and characterized. This interesting receptor plays a key role in cell adhesion, migration and proliferation. It was
identified 20 years ago as the specific cell-surface molecule that could bind and concentrate urokinase on the cell membrane,
thus initiating the proteolytic cascade promoted by the activation of plasminogen. The identification of new extracellular
ligands, such as vitronectin, and of cell-surface interactors, such as integrins and fMet-Leu-Phe receptors, shed new light
on its possible roles, totally independent of the enzymatic properties of its ligand. uPAR ligands and interactors and the
functional consequences of the multiple binding capability of this intriguing receptor are reviewed here.

Alzheimer’s disease (AD) is characterized by an accumulation in the brain of amyloid β peptides (Aβ). The production of Aβ
requires two sequential cleavages induced by β- and γ-secretases on the β-amyloid precursor protein (APP). Altered activity
of these secretases is involved in the pathogenesis of AD. The expression and activity of β-secretase (BACE1) is augmented
in the brain in late-onset sporadic AD. Mutant presenilin 1 (PS1), the major genetic defect of early-onset familial AD (FAD),
alters the activity of γ-secretase, leading to increased production of Aβ42. Here we review the role of oxidative stress as
a molecular link between the β- and the γ-secretase activities, and provide a mechanistic explanation of the pathogenesis
of sporadic late-onset AD. We also discuss evidence for a role of the same mechanism in the pathogenesis of familial AD carrying

In this paper I direct attention to one feature of Hacking’s recent work on styles of reasoning and argue that this feature is of far greater philosophical significance than Hacking’s limited discussion of this suggests. The feature in question is his use of ‘legendary beginnings’ in setting out a given style, viz. the method of introducing a style of reasoning by recounting a popular and quasi-mythical narrative that ties the crystallisation of that style to a particular person in a particular place and at a particular time. Whilst Hacking both deploys and discusses this method, his comments suggest that this is primarily a stylistic device employed for reasons of expedience. In contrast, it is argued here that recounting the legendary origins of a style of reasoning affords a distinctive way of vindicating that style, a vindication from within the style itself. 相似文献

The pineal, serotoninergic and pigmented neurons are associated with light-dependent sleep/arousal, serving as a biological clock with a circadian rhythm. This rhythm is maintained by melatonin which serves to recognise the dark phase. The neural network that responds to seasonal variations in day/night length has not been identified. The present study demonstrates that melanocytes in human skin respond to changes in the duration of UV exposure, and can serve as a biological calendar. These responses are mediated by two indoleamines, serotonin and melatonin. Higher melatonin levels correspond to long nights and long days (short UV pulse), while high serotonin levels in the presence of melatonin reflect short nights and long days (long UV exposure). This response recapitulates the sleep/arousal patterns in animals exposed to large variations in day/night cycle that cause changes in coat colour from pure white in winter to complete repigmentation in summer. 相似文献
