Summary In the endoplasmic reticulum of cells of spleen and lymphnodes of different monkeys, crystalloid inclusions were often found. These inclusions show a pattern of small balls with a diameter of 250 Å, which are connected by 100 Å long bridges. It is supposed that these crystalloid inclusions are produced by the lymphatic cells as a reaction to virus infection, which remains clinically non-apparent, because the same pattern was observed in cells, which were experimentally infected with yellow fever virus, West Nile virus, and rubella virus.  相似文献

The REp cells of the bursa follicle medulla of chicken were isolated in vitro. Culture of the REp cells was maintained over a period of 10 days and the cells were observed at 3 and 10 days by means of transmission electron microscopy (TEM) and immunofluorescence. The use of an anticytokeratin monoclonal antibody confirmed their epithelial nature. TEM observations showed the presence of desmosomes and tonofilaments, which are characteristic of epithelial cells. Furthermore, to some extent the cells regenerated in vitro the network they form in vivo. Though the growth rate becomes slower with time, the features of the REp cells do not significantly change.  相似文献

目的观察成年大鼠脊髓损伤后内源性神经前体细胞的增殖与分化,探讨内源性神经前体细胞的自然变化规律。方法制作脊髓压迫损伤模型,Brdu腹腔注射标记神经前体细胞,免疫荧光法(Immunofluoreseence)检测大鼠脊髓Brdu、GFAP、MBP阳性细胞数的变化。结果 1)正常组可观察到少量Brdu阳性细胞,脊髓损伤后Brdu阳性细胞显著增加(p0.05),并在第7天达到最大值,21天时仍高水平表达。2)正常组可见少量Brdu/GFAP和Brdu/MBP阳性细胞,脊髓损伤后Brdu/GFAP,Brdu/MBP双标阳性细胞数显著增加(p0.05)。结论脊髓损伤后神经前体细胞的数量在第7天达到最大值,我们认为,一周内可能是神经前体细胞增殖分化调控的关键时期。此外,新生星形胶质细胞和少突胶质细胞大量增殖,并与神经前体细胞的迁移、后肢功能恢复表现出一定的同步性,提示新生胶质细胞可能参与了脊髓损伤后神经功能的修复作用。  相似文献

Summary Part of the lymphocytes of the thymus were phagocytized by epithelial and mesenchymal reticular cells in rats after X-ray irradiation of the whole body. The intracellular decomposition is obviously brought about by the two cellular forms in a different way. Pearl-string-like structures — probably nucleoproteids — were found in the cytoplasma in the case of the epithelial cells. On the other hand, numerous cytoplasma inclusions of different size, which are evidently reduced lymphocytic nuclei, could be observed in the case of the mesenchymal cells.  相似文献

Homeostasis of solid tissue is characterized by a low proliferative activity of differentiated cells while special conditions like tissue damage induce regeneration and proliferation. For some cell types it has been shown that various tissue-specific functions are missing in the proliferating state, raising the possibility that their proliferation is not compatible with a fully differentiated state. While endothelial cells are important players in regenerating tissue as well as in the vascularization of tumors, the impact of proliferation on their features remains elusive. To examine cell features in dependence of proliferation, we established human endothelial cell lines in which proliferation is tightly controlled by a doxycycline-dependent, synthetic regulatory unit. We observed that uptake of macromolecules and establishment of cell–cell contacts was more pronounced in the growth-arrested state. Tube-like structures were formed in vitro in both proliferating and non-proliferating conditions. However, functional vessel formation upon transplantation into immune-compromised mice was restricted to the proliferative state. Kaposi’s sarcoma-associated herpes virus (KSHV) infection resulted in reduced expression of endothelial markers. Upon transplantation of infected cells, drastic differences were observed: proliferation arrested cells acquired a high migratory activity while the proliferating counterparts established a tumor-like phenotype, similar to Kaposi Sarcoma lesions. The study gives evidence that proliferation governs endothelial functions. This suggests that several endothelial functions are differentially expressed during angiogenesis. Moreover, since proliferation defines the functional properties of cells upon infection with KSHV, this process crucially affects the fate of virus-infected cells.  相似文献

Summary In the epidermic cells ofAllium cepa stained with a benzopyrene-glycerol-serum medium the fluorescence microscope shows a blue cytoplasmic fluorescence. The cells which have absorbed benzopyrene plasmolize and deplasmolize in the same time as the control cells. Numerous intramitochondrial paracrystalline inclusion bodies (ICIB) were observed in the germinal plasm of a mid-blastula, and in primordial germ cells (PGCs) after their migration to the germinal ridges, in Rana ridibunda, R. lessonae and R. esculenta. In oogonia the number of ICIB decreases rapidly. Single ICIB are observed in the germ cells up to the leptotene stage; they have never been observed in pachytene oocytes. In diplotene oocytes that have reached a diameter of about 100 microns ICIB are visible again, and their number increases concomitantly with oocyte growth.  相似文献

Summary Chemical studies in chick embryo have indicated the existence of proteoglycan in notochordal sheath. Primordial germ cells were observed with scanning electron microscope on the notochord dorsal face, surrounded with perichordal material. We postulate the identification of such a material with proteoglycan which could attract primordial germ cells to the notochord.  相似文献

Summary In 150 out of 164 human hypothalami of all ages, nerve cells with indirect intranuclear inclusions and other karyological specialities were observed. These inclusions existing of intranuclear located cytoplasmic portions must be interpreted as a consequence in a disturbance of the spindle apparatus which took place during the last preceeding mitosis. Summary Cold-stored blood cells of rats, which were depleted of potassium and had gained an equivalent amount of sodium instead, were incubated with 500 mg% glucose at 37°C in serum. Under these conditions an extrusion of sodium from and a re-entry of potassium into the cells could be observed. Desoxycorticosterone, when added in a concentration of 1 mg per ml, was able to slow down the rate of this exchange to three fifths of the rate of untreated cells. With blood of adrenalectomised rats, the same effect of DOC was observed, although the rate without DOC was somewhat lower than in the blood of normal animals. There was no statistically significant difference between the DOC-effect in blood of normal and adrenalectomised rats.  相似文献

Different substrates were used to coat plastic petri dishes for the cultivation of dissociated fetal rat brain cells. Only on surfaces which were coated with a mixture of serum and non-reconstituted collagen, did the majority of the inoculated cells attach singly or as aggregates within 24 h. The attachment of the cells was followed by the outgrowth of cellular processes either from single cells or from aggregates in the same time period. This did not occur on collagen or serum treated or on regular plastic dishes. Under the latter conditions a similar outgrowth was observed only after 3–5 days.  相似文献

Bacterially fermented mistletoe preparations (BFMP) were tested on rat hepatoma tissue culture (HTC) cells and human leukemia Molt 4 cells. A dose-dependent inhibition of the growth rate of the cells was observed. For both cell lines, cytostatic concentrations, expressed in weight of fresh plant, were 0.5 mg/ml culture medium for oak BFMP and 1 mg/ml for apple tree BFMP. However, the action of the two preparations was markedly different on each cell line. Non-viable HTC cells were not stained by trypan blue while non-viable Molt 4 cells were fully colored by this reagent. A lysis of cellular membranes of HTC cells was observed by electron microscopy. Furthermore, oak BFMP inhibited the growth of virus transformed 3T3-SV40 cells more than that of non-transformed 3T3 cells. In contrast to BFMP, non-fermented extracts and a purified mistletoe lectin showed a greater inhibition of the growth of Molt 4 cells than of HTC cells. Samples withdrawn at different times during fermentation gradually lost their inhibitory effect on the growth of Molt 4 cells while their action on HTC cells increased up to the 4th day of fermentation. These results are discussed in relation to the cytotoxic substances of mistletoe already characterized.  相似文献

Childhood nephrotic syndrome is mainly caused by minimal change disease which is named because only subtle ultrastructural alteration could be observed at electron microscopic level in the pathological kidney. Glomerular podocytes are presumed to be the target cells whose protein sieving capability is compromised by a yet unidentified permeability perturbing factor. In a cohort of children with non-hereditary idiopathic nephrotic syndrome, we found the complement fragment C5a was elevated in their sera during active disease. Administration of recombinant C5a induced profound proteinuria and minimal change nephrotic syndrome in mice. Purified glomerular endothelial cells, instead of podocytes, were demonstrated to be responsible for the proteinuric effect elicited by C5a. Further studies depicted a signaling pathway involving Rho/Rho-associated kinase/myosin activation leading to endothelial cell contraction and cell adhesion complex breakdown. Significantly, application of Rho-associated kinase inhibitor, Y27632, prevented the protein leaking effects observed in both C5a-treated purified endothelial cells and mice. Taken together, our study identifies a previously unknown mechanism underlying nephrotic syndrome and provides a new insight toward identifying Rho-associated kinase inhibition as an alternative therapeutic option for nephrotic syndrome.  相似文献

Summary As is shown by electron-microscopic examinations of the thymus, the implantation of ascites-tumour cells usually causes degenerative changes of thymus cells in mice with simultaneous involution of the organ. These changes are already perceptible on the second day. After 7 days most of the lymphocytes give evidence of a so-called segregation of karyoplasm, whereas simultaneously a high dissociation of cells is partially observed. Pyknotic cells could not be observed. Obviously, however, the cellular reaction of the separate thymi is different, which is possibly in relation with tumour growth.  相似文献
