The effect of a small amount of Zr addition on the temperature-dependent stability of Laves phase particles and mechanical properties of Fe-13.5Cr-4.73Al-2.07Mo-(0.34–0.5)Nb-(0.65–0.98)Ta-(0–0.33)Zr (wt. %) ferritic alloys was investigated in the present study. The designed alloy ingots were hot-rolled, aged at 1073 ?K for 24 ?h, and then re-treated at 1273 ?K, 1323 ?K, 1373 ?K, and 1473 ?K for 1 ?h, respectively. It was found that the Zr addition could not only stabilize the Fe2M Laves phase (M ?= ?Mo,Nb,Ta,Zr) to a much higher temperature, but also induce the formation of stable Fe23Zr6 phase. The high-temperature (HT) microstructural stability of the alloys significantly was improved, as evidenced by the fact that a certain amount (0.66–1.19%) of precipitates (Fe2M, Fe23Zr6, and core(Fe23Zr6)-shell(Fe2M)-structured particles) with an appropriate size (~1.0 ?μm) uniformly distributed in the ferritic matrix even after being re-treated at 1473 ?K. Particularly, the formation of core-shell-structured particles at HTs was studied from the viewpoint of both solid solubility and diffusion coefficient of M in the matrix. All these aged alloys exhibited prominent mechanical properties at both room and elevated temperatures, showing high yield strength with σYS ?= ?490–560 ?MPa at room-temperature and σYS ?= ?80–85 ?MPa at 1073 ?K. The strengthening effect was further discussed in light of various strengthening mechanisms, and the calculated strength are in good agreement with the experimental results.  相似文献

The effects of CO2 enrichment on the growth and glucosinolate (GS) concentrations in the bolting stem of Chinese kale (Brassica alboglabra L.) treated with three nitrogen (N) concentrations (5, 10, and 20 mmol/L) were investigated. Height, stem thickness, and dry weights of the total aerial parts, bolting stems, and roots, as well as the root to shoot ratio, significantly increased as CO2 concentration was elevated from 350 to 800 μl/L at each N concentration. In the edible part of the bolting stem, 11 individual GSs were identified, including 7 aliphatic and 4 indolyl GSs. GS concentration was affected by the elevated CO2 concentration, N concentration, and CO2×N interaction. At 5 and 10 mmol N/L, the concentrations of aliphatic GSs and total GSs significantly increased, whereas those of indolyl GSs were not affected, by elevated atmospheric CO2. However, at 20 mmol N/L, elevated CO2 had no significant effects on the concentrations of total GSs and total indolyl GSs, but the concentrations of total aliphatic GSs significantly increased. Moreover, the bolting stem carbon (C) content increased, whereas the N and sulfur (S) contents decreased under elevated CO2 concentration in the three N treatments, resulting in changes in the C/N and N/S ratios. Also the C/N ratio is not a reliable predictor of change of GS concentration, while the changes in N and S contents and the N/S ratio at the elevated CO2 concentration may influence the GS concentration in Chinese kale bolting stems. The results demonstrate that high nitrogen supply is beneficial for the growth of Chinese kale, but not for the GS concentration in bolting stems, under elevated CO2 condition.  相似文献

Ciliate Euplotes octocarinatus centrin (EoCen) is an EF-hand calcium-binding protein closely related to the prototypical calcium sensor protein calmodulin. The first amino acid of the Ca2+-binding loops found in the EF-hand calcium-binding proteins is a highly conserved aspartic acid residue. The D37K mutant was produced to elucidate the metal binding role of the first aspartic acid of the EF-loop I of EoCen. Aromatic-sensitized Tb3+ fluorescence results indicated that the metal binding ability of loop I was lost due to the D37K mutation. Based on fluorescence titration curves of Lu2-D37K, the conditional binding constants of the EoCen loop II were quantitatively found to be K II = (1.61 ± 0.04) × 105 L mol?1 and K II = (3.52 ± 0.08) × 102 L mol?1 with Tb3+ and Ca2+, respectively. Using 2-p-toluidinylnaphthalene-6-sulfonate as a hydrophobic probe, exposure of the hydrophobic surface upon metal binding was found to be significantly reduced for the metal ion-saturated EoCen D37K mutant.  相似文献

来自陆地大量有机碳源的输入,以及三峡库区调度运行和筑坝蓄水导致CO_2、CH_4等碳源气体的排放成为近来关注的热点。以三峡库区干流中段为研究对象,于2017年8月—2018年11月对忠县(ZX),万州(WZ),涪陵(FL)进行每月采样分析,探求水-气界面CO_2、CH_4通量在水库不同运行工况下的变化特征。研究发现,在观测期间内,ZX采样点CO_2浓度在0.008 6~0.115 3 mmol/L波动,WZ采样点CO_2浓度在0.003 5~0.116 8 mmol/L波动,FL采样点0.006 3~0.098 6 mmol/L,整体上3个采样点的CO_2浓度变化趋于一致,都在库区泄水期运行4—5月CO_2浓度达到峰值,在低水位运行期的6—9月降到最低值。CH_4通量在水库低水位运行期表现为"汇",FL点位在泄水期达到"源"的最大值;3个采样点的CO_2通量在泄水期均表现为"源"的特征,低水位运行期的CO_2通量表现为"源"与"汇"的转换。通过与国外已有水库监测数据比较分析,CO_2扩散通量在全球水库序列中处于中等水平,CH_4扩散通量在全球水库序列中处于中等偏下水平。  相似文献

采用DEAE-Sephadex A-50及磷酸纤维素柱层析,用底物亲和洗脱法从萌发油菜(Brassica napus)种子中分离纯化了焦磷酸:果糖-6-磷酸1-磷酸转移酶(PFP).纯化倍数679.7倍,比活力为21.75 单位/毫克.蛋白,活力回收率22.9%.酶的最适pH值为7.5,Mg2+和M2+n对酶有激活作用.进行酶的初级动力学及稳态动力学研究,对果糖-6-磷酸(F6P)表现为典型的米氏规律,Km值为3.33 mmol/L;对焦磷酸(PPi)的活力变化,在PPi浓度小于1.0 mmol/L时具有部分米氏酶特点(Km=1.0 mmol/L),大于1.0 mmol/L时,PPi对酶有抑制作用.从两底物F6P和PPi的相互作用以及产物磷酸(Pi)与底物(F6P和PPi)的相互关系分析,初步推断油菜籽PFP的催化反应为双底物双产物的有序机制.  相似文献

We recorded slow vacuolar (SV-type) channel currents of Radish vacuoles successfully for the first time by using the whole-vacuolar patch-clamp recording mode. SV- type currents would increase and threshold potentials of activation would shift towards more negative values with the increase of concentrations of cytosolic Ca2+. When 2.5 mmol/ L LaCl3 and 4 mmol/L EGTA were added to bath solutions, SV-type currents were suppressed remarkably. Then adding LaCl3 with different concentrations to pipette solutions, we found that LaCl3 with higher concentrations (>4 ´ 10-7 mol/L) had a strong inhibitory effect on SV-type currents, while LaCl3 with lower concentrations (≤4 ´ 10-7 mol/L) promoted channel currents. This promoting effect provides an important basis at channel level for researching further the effects of rare earth on physiological activities of plants and the production-increase effects of rare earth fertilizers on crops.  相似文献

淫羊藿无性系种群构件生物量分配格局   总被引:4,自引:0,他引:4  
淫羊藿为具有克隆生长习性的多年生根茎型草本植物,是重要的药用资源植物.该文以野生淫羊藿为研究材料,于营养生长期和有性生殖期对无性系分株种群进行取样,比较分析不同时期、不同构件生物量分配格局.结果表明,营养期与有性生殖期无性系分株种群生物量差异不显著,但各构件生物量百分比差异显著.营养期根生物量比(RMR)、支持结构生物量比(SBR)和根冠比(R/C)均大于有性生殖期;而叶生物量比(LMR)和叶重分数(LMF)小于有性生殖期.在有性生殖期,营养分株(VR)生物量(83.93%)远远大于生殖分株(RR)生物量(16.07%),用于生殖器官(花)的生物量投资比例非常小(1.45%),叶构件生物量分配比例(50.68%)最大,地下根茎(32.03%)和支持结构(15.84%)生物最分配比例次之.高比例的叶构件生物量有利于合成大量有机物质,这些物质储存于地下根茎中以维持基株在资源缺乏季节的生长,保证种群的生存和繁衍.这些都暗示着淫羊藿不仅有很强的克隆生长能力,而且采取克隆生长为主,有性繁殖为辅的手段维持种群的繁衍与扩散.  相似文献

利用分子动力学模拟了Zr50Cu50金属玻璃的形成过程,并获得了不同温度下合金的原子构型.借助金属玻璃中自由体积量等于金属玻璃与对应晶体的体积差理论提出一种自由体积湮没速度法,对Zr50Cu50金属玻璃形成过程中的临界玻璃态转变温度Tc以及热力学玻璃态转变温度Tg进行预测.用该方法确定出的Tc(969.5K)与利用模式耦合理论计算获得的Tc(978.4K)接近;Tg(731K)与利用平均原子体积随温度变化关系曲线确定的Tg(725K)相近.运用自由体积湮没速度法计算的Tc和Tg无需计算各温度下的原子扩散系数,节省了计算时间.  相似文献

Nucleoside phosphorylases (NPases) were found to be induced in Enterobacter aerogenes DGO-04, and cytidine and cytidine 5'-monophosphate (CMP) were the best inducers. Five mmol/L to fifteen mmol/L cytidine or CMP could distinctly increase the activities ofpurine nucleoside phosphorylase (PNPase), uridine phosphorylase (UPase) and thymidine phosphorylase (TPase) when they were added into medium from 0 to 8 h. In the process of enzymatic synthesis of adenine arabinoside from adenine and uracil arabinoside with wet cells ofEnterobacter aerogenes DGO-04 induced by cytidine or CMP, the reaction time could be shortened from 36 to 6 h. After enzymatic reaction the activity of NPase in the cells induced remained higher than that in the cells uninduced.  相似文献

Long-chain alkenones (LCAs) can potentially be used as indicators to understand past variations in lacustrine environments.Previous research has suggested that the relationship between the temperature and the unsaturation index of LCAs should be calibrated individually,because of the possible variations in the alkenone-producing algal species in the lacustrine environment.In this work,we have calibrated U37K’ of water filter samples against the in-situ water temperature in Lake Qinghai,Tibetan Plateau.There are significant relationships between U37K’ and the water temperature,a non-linear relationship was derived.Because the U37K’ values did not respond sensitively at lower temperatures,we suggested that a quadratic regression (U37K’ =0.0011×T2-0.0201×T+0.1959,n=15,r2=0.74) was appropriate than linear regression to represent the relationship between the in-situ temperatures and U37K’.Meanwhile,the U37K correlation relationship was not more significant than U37K’ index in our study.Because of the C37:4 effects by salinity change,we suggest U37K is not as robust as the U37K’ index as a temperature proxy,at least for the salt lake in the Tibetan Plateau.The calibration of the U37K’ index in this work has provided a new understanding of historic climatic changes in the Tibetan Plateau.  相似文献

Photosynthetic inorganic carbon utilization was investigated in two cyanobacteria Microcystis aeruginosa and Synechocystis PCC6803 grown in standing culture. Photosynthetic rates for the two algae reached about 10 times the theoretical CO2 supply rate at low dissolved inorganic carbon (DIC) of 100 μmol/L, and the rates were unaffected by the addition of 20 mmol/L Na+, indicating that the two algae possessed Na+-independent HCO-3 utilization for photosynthesis under low DIC. Their photo- synthetic rates at low DIC were inhibited by higher Cl- and the degrees of inhibition were increased with the rise of CI- concentration, and in the presence of Diphenylamine-2-carboxylate (DPC), a reported Cl- channel inhibitor, the rates decreased by 74%-82%, implying that putative DPC-sensitive Cl- channels participate in Na+-independent HCO3- uptake for photosynthesis. The experiment of intracellular 14C-DIC accumulation for photosynthesis showed that internal DIC pools decreased by about 80% with 200 μmol/L DPC and by 64%-70% with 100 mmol/L Cl-. The experiment of chlorophyll a fluorescence quenching showed that initial rates and extents of fluorescence quenching obviously decreased with 200 μmol/L DPC or 100 mmol/L Cl-. The two experiments gave further evidence that putative DPC-sen- sitive Cl- channels participate in Na+-independent HCO-3 uptake for photosynthesis in the two algae grown in standing culture.  相似文献

The composition characteristics of maraging stainless steels were studied in the present work investigation using a cluster-plus-glue-atom model. The least solubility limit of high-temperature austenite to form martensite in basic Fe–Ni–Cr corresponds to the cluster formula [NiFe12]Cr3,where NiFe12is a cuboctahedron centered by Ni and surrounded by 12 Fe atoms in FCC structure and Cr serves as glue atoms. A cluster formula [NiFe12](Cr2Ni) with surplus Ni was then determined to ensure the second phase(Ni3M) precipitation,based on which new multicomponent alloys [(Ni,Cu)16Fe192](Cr32(Ni,Mo,Ti,Nb,Al,V)16) were designed. These alloys were prepared by copper mould suction casting method,then solid-solution treated at 1273 K for 1 h followed by water-quenching,and finally aged at 783 K for 3 h. The experimental results showed that the multi-element alloying results in Ni3M precipitation on the martensite,which enhances the strengths of alloys sharply after ageing treatment. Among them,the aged [(Cu4Ni12)Fe192](Cr32(Ni8.5Mo2Ti2Nb0.5Al1V1)) alloy(Fe74.91Ni8.82Cr11.62Mo1.34Ti0.67Nb0.32Al0.19V0.36Cu1.78wt%) has higher tensile strengths with YS?1456 MPa and UTS?1494 MPa. It also exhibits good corrosion-resistance in 3.5 wt% NaCl solution.  相似文献

The Sinorhizobium meliloti C4-dicarboxylate transport (Dct) system is essential for symbiotic nitrogen fixation. The dctA gene, encoding the C4-dicarboxylate permease, is expressed in both free living and symbiotic cells. But in free living cells expression of dctD and dctB is absolutely required for the expression of dctA. In this study, in order to investigate the effect of oxygen concentration on the induction of Dct system, E. coli DH5a strain which carries the plasmid-encoded dctABD operon was used in tube assays. It was found that the specific induction of Dct system oc- curred only at a certain depth under the surface of M63-0.6% agar media, suggesting that Dct system could respond to oxygen concentration during succinate-induced expression. Furthermore, when measured at different oxygen concentrations, the highest expression level was observed at oxygen concentration of 2%. Thus, we predict that in addition to dicarboxylates, the induction of Dct system may also regulated by oxygen concentration.  相似文献

随着经济的发展,酸雨已成为全球性的环境污染问题,我国酸雨污染出现的频率和强度也在逐年增加。如何在酸雨影响下保护农作物产量是当前酸雨相关研究的热点问题。实验选择了我国东北地区典型的农作物小白菜作为研究对象。通过模拟实验,分析了酸雨胁迫下小白菜的生理生长指标的变化,并验证土壤中Ca(NO_3)_2的添加是否可以维持酸雨条件下小白菜的产量。研究结果表明酸雨和土壤中的Ca(NO_3)_2均会影响小白菜的部分生长生理特性,酸雨的存在会影响小白菜的可食用部分的产量等指标。但是当土壤中Ca(NO_3)_2浓度增加时,会降低酸雨的影响,间接地减弱了酸雨对小白菜产量的影响。因此可在酸雨影响较为强烈的地区,通过增加土壤中Ca(NO_3)_2浓度,以降低酸雨对小白菜的影响。  相似文献

The Pb-17wt% Sb alloy was directionally solidified under two solidification conditions: with different temperature gradients (G=0.93–3.67 K/mm) at a constant growth rate (V=17.50 μm/s) and with different growth rates (V=8.3–497 μm/s) at a constant temperature gradient (G=3.67 K/mm) in a Bridgman furnace. Microstructure parameters, such as primary dendrite arm spacing (λ1), secondary dendrite arm spacing (λ2), and dendrite tip radius (R), were measured. The microhardness (Hv) and ultimate tensile strength (σ) of the directional solidification samples were also measured. The influences of solidification and microstructure parameters on Hv and σ were investigated. The results obtained in this work were compared with similar experimental researches in literatures. It is shown that the Hv and σ values increase with the increase of G and V, but decrease with the increase of λ1, λ2, and R.  相似文献

长梗吊石苣苔Lysionotus longipedunculatus (W.T. Wang) W.T. Wang是我国吊石苣苔属中的一个比较罕见的物种,目前濒危等级被评估为极危CR Blac (ⅰ,ⅱ,ⅴ),而同属的吊石苣苔L. pauciflorus Maxim.分布范围非常广泛、生境变化多样,濒危等级为无危LC。本研究通过筛选出16个微卫星位点对长梗吊石苣苔的两个居群进行遗传多样性研究,并对比相近地区的吊石苣苔居群,比较研究导致长梗吊石苣苔濒危的原因。结果显示二者的遗传多样性都比较低,有较高自交现象,但吊石苣苔具有明显的走茎,可以有效地克隆繁殖,相较之下,长梗吊石苣苔较弱的克隆繁殖能力可能是导致其濒危的原因之一。  相似文献

The dense granule protein 4 (GRA4) is a granular protein from Toxoplasma gondii, and is a candidate for vaccination against this parasite. In this study, the plasmid pcDNA3.1-GRA4 (pGRA4), encoding for the GRA4 antigen, was incorporated by the dehydration-rehydration method into liposomes composed of 16 mmol/L egg phosphatidylcholine (PC), 8 mmol/L dioleoyl phosphatidylethanolamine (DOPE), and 4 mmol/L 1,2-diodeoyl-3-(trimethylammonium) propane (DOTAP). C57BL/6 mice and BALB/c mice were immunized intramuscularly three times with liposome-encapsulated pGRA4 to determine whether DNA immunization could elicit a protective immune response to T. gondii. Enzyme-linked immunosorbent assay (ELISA) of sera from immunized mice showed that liposome-encapsulated pGRA4 generated high levels of IgG antibodies to GRA4. Production of primary interferon (IFN)-γ and interleukin (IL)-2 in GRA4-stimulated splenocytes from vaccinated mice suggested a modulated Th1-type response. 72.7% of C57BL/6 mice immunized with liposome-encapsulated pGRA4 survived the challenge with 80 tissue cysts of ME49 strain, whereas C57BL/6 mice immunized with pGRA4 had only a survival rate of 54.5%. When immunized BALB/c mice were intraperitoneally challenged with 103 tachyzoites of the highly virulent RH strain, the survival time of mice immunized with liposome-encapsulated pGRA4 was markedly longer than that of other groups. Our observations show that liposome-encapsulated pGRA4 enhanced the protective effect against infection of T. gondii.  相似文献

对湘西地区凤凰、永顺、龙山3个不同居群的箭叶淫羊藿叶片的超氧阴离子自由基(·O2)的产生速率和丙二醛(MDA)的含量进行测定.结果表明:永顺的箭叶淫羊藿的超氧阴离子自由基的产生速率最大,为(0.059 1±0.003 3) μmol·min-1·g-1,而龙山的箭叶淫羊藿的超氧阴离子自由基的产生速率最小,为(0.053 0±0.000 9) μmol·min-1·g-1;在3个不同居群的箭叶淫羊藿叶片的MDA的含量中,永顺的箭叶淫羊藿的含量最高,为(0.014 9±0.001 4) nmol·g-1,而龙山的箭叶淫羊藿的含量最低,为(0.007 5±0.001 4) nmol·g-1.  相似文献

In this paper,we report that the thermoelectric performance of n-type Pb Se could be improved through synergistically optimizing electrical and thermal transport properties via Sb doping and Mg alloying.The carrier concentration was firstly optimized through Sb doping,resulting in a maximum power factor of~15.4μW cm~(-1)K~(-2)and maximum ZT of~0.9 at 873 K in Pb_(0.99)Sb_(0.01)Se.Then,Mg was selected for alloying in Pb sites to produce point defects,which can largely intensify the phonon scattering and lower thermal conductivity.After Mg alloying,the thermal conductivity at 300 K(873 K)was significantly suppressed from~4.6 Wm~(-1)K~(-1)(1.5 Wm~(-1)K~(-1))for Pb_(0.99)Sb_(0.01)Se to~2.9 Wm~(-1)K~(-1)(1.1 Wm~(-1)K~(-1))for Pb_(0.99)Sb_(0.01)Se-6%Mg Se.Through combining Sb doping and Mg alloying,a maximum ZT of~1.1 was achieved at 873 K for Pb_(0.99)Sb_(0.01)Se-6%Mg Se,and the average ZT(ZT_(ave))was increased by 28.6%from~0.42 for Pb_(0.99)Sb_(0.01)Se to~0.54 for Pb_(0.99)Sb_(0.01)Se-6%Mg Se.The results indicate that Pb Se is a robust candidate for medium-temperature thermoelectric applications.  相似文献
