Summary The circadian clock in the unicellular algaGonyaulax polyedra is accelerated by a substance in extracts from the cells themselves. The extracts have been fractionated using the circadian rhythm of bioluminescence as bioassay. The active substance, termed gonyauline, has been isolated and characterized as a novel low molecular weight cyclopropanecarboxylic acid (S-methyl-cis-2-(methylthio) cyclopropanecarboxylic acid). Synthetic gonyauline has a similar shortening effect on the period of the circadian clock.  相似文献

In this article we model the log of the US inflation rate by means of fractionally integrated processes. We use the tests of Robinson (1994) for testing this type of hypothesis, which include, as particular cases, the I(0) and I(1) specifications, and which also, unusually, have standard null and local limit distributions. A model selection criterion is established to determine which may be the best model specification of the series, and the forecasting properties of the selected models are also examined. The results vary substantially depending on how we specify the disturbances. Thus, if they are white noise, the series is I(d) with d fluctuating around 0.25; however, imposing autoregressive disturbances, the log of the US inflation rate seems to be anti‐persistent, with an order of integration smaller than zero. Looking at the forecasting properties, those models based on autocorrelated disturbances (with d < 0) predict better over a short horizon, while those based on white noise disturbances (with d > 0) seem to predict better over longer periods of time. The prothoracicotropic hormones (PTTHs) are cerebral peptides that control insect postembryonic development by stimulating the prothoracic glands to synthesize ecdysteroids. InManduca sexta, the tobacco hornworm, two classes of PTTH are distinguished by their Mr, small (ca. 7 kDa) and big PTTH (ca. 25–30 kDa). Little is known about the physical nature of the PTTHs and this study takes a first step towards defining characteristics of theManduca big PTTH. The neurohormone has a Stokes radius of 2.59 nm and a sedimentation coefficient of 2.76 S. Based on these data, an Mr of 29,443.7 and anf/f 0 of 1.27 were calculated. Combined, the physical data revealManduca big PTTH is an asymmetrical acidic homodimeric peptide with intra- and intermolecular disulfide bonds.  相似文献

Summary The mechanism of the in vitro inhibition of Ca2+-, phosphatidylserine-dependent protein kinase C (PK-C)2 by the purifiedholo (ligand-saturated) forms of cellular retinol-binding protein (cRBP) and cellular retinoic acid-binding protein (cRABP) was studied. We report here that i) the PK-C-inhibitory action ofholo-cRBP andholo-cRABP is due to their respective ligands, all-trans-retinol and all-trans-retinoic acid; ii) the reduced phosphorylation of theholo-retinoid-binding proteins and brain cytosolic proteins is not the result of a retinoid-induced soluble phosphatase or protease activity; iii) retinoids reduce PK-C affinity for calcium and phosphatidylserine in vitro; and iv) the structure-function activity of the retinoids and the specific interaction of these effect of retinoids on plasma membrane-associated PK-C activity pays a significant role in defining the early epigenetic aspects of PK-C-dependent tumor promotion and may be a physiological mechanism by which retinoids induce terminal differentiation in cell types that do not express soluble retinoid-binding proteins.We would like to thank Dr L.M. Two classes of sponges (animal phylum Porifera) possess a siliceous skeleton which is composed of spicules. Studying the optical fiber-mechanical properties of large spicules from hexactinellid sponges (> 5 cm) it was demonstrated that they are effective light-collecting optical fibers. Here, we report that the demosponge Suberites domuncula is provided with a biosensor system composed of the (organic) light producing luciferase and the (inorganic) light transducing silica spicules. The light transmission feature of these smaller spicules (200 μm) has been demonstrated and the ability of sponge tissue to generate light has been proven. Screening for a luciferase gene in S. domuncula was successful; the recombinant luciferase was prepared and shown to be bioactive. The luciferase protein is abundantly present in the close neighborhood of the spicules. The expression of the luciferase gene is under the control of light. It has been suggested that a common sex pheromone composition may account for interspecific sexual interactions observed with certain moths in the Arctiidae. In this study, it is demonstrated that the sex pheromones released by females of the Scarlet Tiger Moth,Callimorpha dominula L., and the Cinnabar Moth,Tyria jacobaeae L., have similar activities and elute at the same retention time on analysis by coupled gas chromatography (GC)-electrophysiology with males from each species. Peak enhancement on GC, chiral GC and coupled GC-mass spectrometry using authentic compounds show that the sex pheromone for bothC. dominula andT. jacobaeae is (3Z,6Z,9S,10R)-9,10-epoxyheneicosa-3,6-diene.  相似文献

Summary Aqueous extracts from spermatophores ofAcanthoscelides obtectus (Coleoptera, Bruchidae) have been chromatographed on Sephadex G 25 and G 15 columns, the active fractions being further purified by preparative SiO2 TLC. The isolation and properties of 2 biologically active compounds are reported. The paragonial substance A has a favourable action upon oogenesis, is a low molecular weight molecule (500–1000) and gives 1 single amino acid on hydrolysis, corresponding to glycin on TLC. The paragonial substance B is toxic, or an inhibitor of oogenesis at lower concentration, thus showing an antagonistic effect.  相似文献

DNA probes containing the repeated rDNA region ofDrosophila melanogaster (coding for e.g. 28S and 18S rRNA) hybridized in situ to distinct regions of two heterologous mitotic chromosomes of the honeybee, identifying the nucleolus organizing regions (NORs). The method allows a rapid establishment of a physical map ofApis mellifera using other DNA probes ofDrosophila. This is the first report on well-defined chromosomal markers in the honeybee.  相似文献

This is the second installment of a two-part paper on developments in quantum dispersion theory leading up to Heisenberg’s Umdeutung paper. In telling this story, we have taken a 1924 paper by John H. Van Vleck in The Physical Review as our main guide. In this second part we present the detailed derivations on which our narrative in the first part rests. The central result that we derive is the Kramers dispersion formula, which played a key role in the thinking that led to Heisenberg’s Umdeutung paper. We derive classical formulae for the dispersion, emission, and absorption of radiation and use Bohr’s correspondence principle to construct their quantum counterparts both for the special case of a charged harmonic oscillator (Sect. 5) and for arbitrary non-degenerate multiply-periodic systems (Sect. 6). We then rederive these results in modern quantum mechanics (Sect. 7). This paper was written as part of a joint project in the history of quantum physics of the Max Planck Institut für Wissenschaftsgeschichte and the Fritz-Haber-Institut in Berlin. Mining education was one of the areas of technical savoir transformed during the eighteenth century. Mining academies arose and spread through Europe in the second half of that century. This happened first in the German states and the Austrian dominions, due to the cameralistic system, and soon developed elsewhere through a transfer of the German model to France (Ecole des mines) as well as to other francophone and Spanish-speaking areas (Belgium and Piedmont-Savoy, Spain and America). The mining academies may rightly be considered among the prototypes of technical high schools (Polytechniques and Technische Hochschulen) established during the nineteenth century. In the course of exploring some of the details of this development, the present paper aims at suggesting outlines to be followed in pursuing a comparative analysis of special schools for the training of technicians and mining officers (Bergschulen and Bergakademien). A second aim is to examine more closely the Italian case as a typical example of adaptation, although on a smaller scale, of French and German experiences. This can provide a framework within which to place further microanalyses and case-studies.  相似文献

The conventional growth rate measures (such as month‐on‐month, year‐on‐year growth rates and 6‐month smoothed annualized rate adopted by the US Bureau of Labor Statistics and Economic Cycle Research Institute) are popular and can be easily obtained by computing the growth rate for monthly data based on a fixed comparison benchmark, although they do not make good use of the information underlying the economic series. By focusing on the monthly data, this paper proposes the k‐month kernel‐weighted annualized rate (k‐MKAR), which includes most existing growth rate measures as special cases. The proposed k‐MKAR measure involves the selection of smoothing parameters that are associated with the accuracy and timeliness for detecting the change in business turning points. That is, the comparison base is flexible and is likely to vary for different series under consideration. A data‐driven procedure depending upon the stepwise multiple reality check test for choosing the smoothing parameters is also suggested in this paper. The simple numerical evaluation and Monte Carlo experiment are conducted to confirm that our measures (in particular the two‐parameter k‐MKAR) improve the timeliness subject to a certain degree of accuracy. The business cycle signals issued by the Council for Economic Planning and Development over the period from 1998 to 2009 in Taiwan are taken as an example to illustrate the empirical application of our method. The empirical results show that the k‐MKAR‐based score lights are more capable of reflecting turning points earlier than the conventional year‐on‐year measure without sacrificing accuracy. This paper presents the main features of the treatise on magnetism written by the Jesuit Leonardo Garzoni (1543–92). The treatise was believed to be lost, but a copy of it has been recently recovered. The treatise is briefly described and analysed. The results of a comparison between Garzoni's treatise, Della Porta's Magia Naturalis (1589), and Gilbert's De Magnete (1600) are also summarized. As claimed in the seventeenth century by Niccolò Cabeo and Niccolò Zucchi, the treatise contains quite a lot of the material to be found subsequently in the Magia Naturalis and in the De Magnete. Most importantly, the treatise presents so many interesting features, well before Gilbert's work, which make it the first example of a modern treatment of magnetic phenomena.  相似文献

Summary It is reported that concanavalin A (conA) and wheat germ agglutinin (WGA) have a differential binding pattern on normal mouse spleen lymphocytes and the surface of Dalton's lymphoma cells. It is suggested that sialic acid on the cell surface controls the expression of lectin binding sites. Further, it has been observed that the increased release of sialic acid from cell surfaces aftercis-dichlorodiammine platinum (II) (cis-Platin) treatment is due to the increased activity of sialidase.To whom reprint requests should be addressed.  相似文献

Summary The DNA interspersion pattern in 3 Selachians (R. asterias, T. marmorata andS. stellaris) has been studied through the reassociation kinetics of short (0.3 Kb) and long (2.5 Kb) DNA fragments. Preliminary results show that most of the DNA (approximately 80%) of these organisms is arranged according to a short-period interspersion pattern. A notable resemblance to the pattern previously described in the teleosteanSalmo trutta has been observed.  相似文献

System-based combination weights for series r/step-length h incorporate relative accuracy information from other forecast step-lengths for r and from other series for step-length h. Such weights are examined utilizing the West and Fullerton (1996) data set-4275 ex ante employment forecasts from structural simultaneous equation econometric models for 19 metropolitan areas at 10 quarterly step-lengths and a parallel set of 4275 ARIMA forecasts. The system-based weights yielded combined forecasts of higher average accuracy and lower risk of large inaccuracy than seven alternative strategies: (1) averaging; (2) relative MSE weights; (3) outperformance (per cent best) weights; (4) Bates and Granger (1969) optimal weights with a convexity constraint imposed; (5) unconstrained optimal weights; (6) select a ‘best’ method (ex ante) by series and; (7) experiment in the Bischoff (1989) sense and select either method (2) or (6) based on the outcome of e experiment. Accuracy gains of the system-based combination were concentrated at step-lengths two to five. Although alternative (5) was generally outperformed, none of the six other alternatives was systematically most accurate when evaluated relative to each other. This contrasts with Bischoff's (1989) results that held promise for an empirically applicable guideline to determine whether or not to combine.  相似文献

Summary The average duration of larval life in the anadromous lamprey,Geotria australis (the sole representative of the Geotriidae) is estimated as 41/4 years. Under anaerobic conditions and in the presence of nitrate, the facultative anaerobe Escherichia coli synthesises an electron-transport chain comprising a primary dehydrogenase and the terminal membrane-bound nitrate reductase A (NarGHI). This review focuses on recent advances obtained on the structure and function of the three protein subunits of membrane-bound nitrate reductases. We discuss a global architecture for the Mo-bisMGD-containing subunit (NarG) and a coordination model for the four [Fe–S] centres of the electron-transfer subunit (NarH) and for the two b-type haems of the anchor subunit NarI.  相似文献

A forecasting model for yt based on its relationship to exogenous variables (e.g. x?t) must use x?t, the forecast of x?t. An example is given where commercially available x?t's are sufficiently inaccurate that a univariate model for yt appears preferable. For a variety of types of models inclusion of an exogenous variable x?t is shown to worsen the yt forecasts whenever x?t must itself be forecast by x?t and MSE (x?t) > Var (x?t). Tests with forecasts from a variety of sources indicate that, with a few notable exceptions, MSE (x?t) > Var (x?t) is common for macroeconomic forecasts more than a quarter or two ahead. Thus, either:
  • (a) available medium range forecasts for many macroeconomic variables (e.g. the GNP growth rate) are not an improvement over the sample mean (so that such variables are not useful explanatory variables in forecasting models), and/or
  • (b) the suboptimization involved in directly replacing x?t by x?t is a luxury that we cannot afford.

Subunit interaction in sorbitol dehydrogenase (SDH) has been studied with in vitro and in silico methods identifying a vital hydrogen-bonding network, which is strictly conserved among mammalian SDH proteins. Mutation of one of the residues in the hydrogen-bonding network, Tyr110Phe, abolished the enzymatic activity and destabilized the protein into tetramers, dimers and monomers as judged from gel filtration experiments at different temperatures compared to only tetramers for the wild-type protein below 307 K. The determined equilibrium constants revealed a large difference in Gibbs energy (8 kJ/mol) for the tetramer stability between wild-type SDH and the mutated form Tyr110Phe SDH. The results focus on a network of coupled hydrogen bonds in wild-type SDH that uphold the protein interface, which is specific and favorable to electrostatic, van der Waals and hydrogen-bond interactions between subunits, interactions that are crucial for the catalytic power of SDH.
