Physiologic and genetic analysis of a mutant excreting alkaline phosphatase in Escherichia coli K 12

A physiological analysis of an E. coli K 12 periplasmic-leaky mutant shows that specific excretion of periplasmic proteins increases with time and temperature of incubation when cells are grown in rich L medium. The associated lky mutation was mapped by use of conventional genetic techniques near the gal locus.     

Possible role of intestinal alkaline phosphatase activity in thiamine transport

Summary Thiamine deficiency caused a marked decrease of intestinal alkaline phosphatase (al-Pase) activity, but had no effect on the Ca⁺⁺-ATPase activity and Ca⁺⁺-absorption in rats. The al-Pase activity was significantly decreased 1 h after oral administration of ethanol at 0.5 and 2.5 g/kg. In contrast, Mg⁺⁺-, Ca⁺⁺- and (Na⁺+K⁺)-ATPase activities did not change after the administration of ethanol. These findings show that the al-Pase activity, unlike the Ca⁺⁺-ATPase activity, is not related to Ca⁺⁺-absorption. A possible role of al-Pase activity in the active transport of thiamine in the intestine was discussed.

Zusammenfassung Durch KMnO₄-Injektionen hervorgerufene lokale Hautverkalkung kann durch Pyrophosphatinjektionen verhindert werden. Durch alkalische Phosphatase wird die Pyrophosphatwirkung aufgehoben.

Zusammenfassung Zu Beginn der Entwicklung der Wirbelsäure im Hühnerembryo findet sich die alkalische Phosphatase intrazellulär. Mit der späteren Verkalkung und Entwicklung des Knochenmarks ist die Reaktion extrazellulär nachweisbar.     

The localization and functional significance of alkaline phosphatase in the vertebrate ovary

Zusammenfassung Es zeigt sich, dass die Wirkung der alkalischen Phosphatase in der Theca interna, im interstitiellen glandulären Gewebe und im Corpora lutea liegt und eine bedeutende Transportfunktion bei den ovariellen Zellmembranen der Wirbeltiere hat.     

Ultrastructure of the particles reconstituted by complementation of extracts from chl-r mutants of Escherichia coli K 12]

By freeze-fracturing it is shown that the vesicles reconstituted by complementation of the chlA and chlB mutants of E. coli K 12 extracts are characterized by an asymmetric membrane bilayer. In a feature quite similar to the original intact plasma membranes, the membrane splits in two halves and the intramembranous particles are asymmetrically distributed on the two facture faces. It is proposed that the process of membrane reconstitution, which is also associated with the restoration of nitrate-reductase activity, relies on a sequence of increasing complexity of the molecular organisation.

Stimulation of the synthesis and non-competitive inhibition of alkaline phosphatase by theophylline in normal hamster fibroblasts and absence of response in transformed fibroblasts]

Theophylline increases the synthesis of alkaline phosphatase in normal Hamster fibroblasts (activity X6 after 24 hours with theophylline 10-3 M). The enzymatic activity of these cells is inhibited by theophylline in vitro (80% inhibition in the presence of theophylline 10-3 M). The inhibition seems to be non competitive, since the apparent Km of the enzyme is not modified. This stimulator and inhibitor effect of theophylline is absent in Hamster fibroblasts transformed by SV 40 virus.
