生长抑素RNA探针的制备和初步应用

利用含有外源片段生长抑素(SS)-cDNA的质粒pGEM-3进行扩增,采用HindⅢ使质粒线性化,在T7RNA聚合酶的作用下,以尿苷5'[α-32P]三磷酸盐和其它核苷酸合成了同位素标记的ssRNA探针,而以地高辛(DIG)标记的各种核苷酸的混和物合成了DIG标记的ssRNA探针,并分别与从大鼠脑组织中提取的总RNA进行点杂交,以检测探针的有效性,结果这两种探针均有较强的杂交信号,且与总RNA的量呈一定正相关关系,均可用于检测有关样品中SSmRNA的相对含量.     

蓝舌病病毒内蒙古分离株VP2基因5′端序列分析及探针制备

从内蒙古巴盟地区分离的蓝舌病病毒株(BTV—NM)提取总RNA，经反转录PCR扩增VP2基因5′端片段，并构建至pGEM—T载体中。序列测定后，将这一序列与蓝舌病毒澳大利亚株VP2基因5′端进行比较分析：同源性为40％。通过PCR法标记克隆的cDNA片段，制备地高辛标记探针，与粗提的蓝舌病发病羊病毒RNA进行Northernblot杂交，并作敏感性试验，结果表明此探针对蓝舌病毒内蒙古分离株具有特异性，可检测出50pg的病毒RNA。     

轮状病毒VP7基因的克隆及核苷酸序列分析

用反转录聚合酶链反应（RT－PCR）技术，从轮状病毒感染的细胞中扩增了916bp的VP7片段中抗原表位区。通过T4DNA连接酶将其直接连接于克隆载体质粒pGEM-T上，转化至受体菌DH5a中。提取质粒经PCR扩增、酶切鉴定，证明重组质粒pT-V7中含有轮状病毒的VP7基因片段。经核苷酸序列分析，表明正确克服了轮状病毒主要保护性抗VP7基因中抗原表位区。     

大鼠20αHSD基因调控序列的克隆与检测

酶切包含20αHSD基因及其调控序列的X噬菌体13NA,获得4．0kb的DNA大片段,将此片段与载体质粒连接得到重组质粒,酶切重组质粒后电泳,再以地高辛标记的20αHSD基因的cDNA为探针进行Southern杂交,实验获得了大片段的重组质粒。     

质粒Yp7构建的探讨

以大肠杆菌酵母穿梭质粒YRp7 DNA为材料,经体外重组构建了质粒Yp7。该质粒具有Amp~r、Tet~r两个抗药性标记和一个来自酵母细胞的Trpl基因。分子量约为5.25kb。该质粒可用于筛选水稻等真核生物DNA中的自主复制序列。     

大肠杆菌caiE基因的克隆与高效表达

用聚合酶链反应（PCR）从大肠杆菌K12s中扩增大肠杆菌肉碱代谢相关酶基因cai E,将其克隆到克隆载体pBluescripy SK中，测序表明：该基因有612bp，与文献报道相比，有10个核苷酸不同，相应的翻译氨基酸有6个相异，将该基因重组到ColE1为复制子，T7为启动子控制下的分泌型表达载体pET-22b( )中，构建表达质粒pETCaiE，重组到载体pACYC184中构建以p15A为复制子，Lac为启动子的表达质粒pACYC-CaiE；重组到载体pWSK129中，构建以pSC101为复制子，T7为启动子的表达质粒pSC-CaiE。上述表达质粒转化大肠杆菌BL21（DE3），经1mmol/L异丙基硫代－β－D－半乳糖苷（IPTG）诱导后，均可表达，SDS－PAGE分析表明表达蛋白分子质量约26ku，表达量占菌体总蛋白质的比例分别为pETCaiE30%,pACYC-CaiE8%,pSC-CaiE5%。     

定点突变内皮抑素Zn2+的结合位点及突变基因的克隆表达

从人胚肝组织中提取总RNA, 以逆转录聚合酶链式反应（RT-PCR）法获得人内皮抑素编码序列, 采用定点突变技术将His2和His4双突变为Leu2和Val4. 将突变基因cDNA插入含有T7启动子的质粒pET-28b中构建表达质粒pMendo, 转化大肠杆菌BL21(DE3), 筛选表达菌株BL21-Mute, 表达菌株经IPTG诱导后以包涵体方式产生大量内皮抑素突变蛋白. SDS-PAGE分析表明, 表达的重组蛋白占菌株可溶性蛋白质的30%. 复性、 纯化的内皮抑素突变蛋白纯度达到98%, 失去抑制人脐静脉内皮细胞增殖的活性.     

羊传染性脓疱基因工程苗的基础研究概述

该研究初步构建了羊传染性脓疱病毒HCE株基因组DNA的三种限制性内切酶物理图谱 ,建立了特异性很强的光敏生物素和地高辛标记探针检测方法 ,克隆了部分酶切片断 构建了重组表达质粒和基因工程菌 ,该工程菌能高水平地表达良好的病毒抗原 ,动物试验表明具有较好的保护效果     

Pichia酵母宿主细胞DNA残留量检测方法的建立

为检测重组人血白蛋白中外源性DNA残留量,以宿主酵母工程菌基因组DNA为模板,使用地高辛标记探针进行点杂交.结果证明:该方法检测灵敏度较好,特异性较强,操作安全简便,可用于重组人血白蛋白的检测.     

小鼠酪氨酸酶的克隆及在大肠杆菌中的表达

采用逆转录-聚合酶链式反应(RT -PCR)方法,从小鼠黑色素瘤总RNA中扩增得到小鼠酪氨酸酶的cDNA .该基因重组到T7启动子控制下的表达载体pET - 2 2b(+)中,构建表达质粒pET -TYR并转化大肠杆菌Rosetta (DE3) .SDS -PAGE及蛋白质序列测定表明,经0 .8mmol/L异丙基硫代- β-D -半乳糖苷(IPTG)诱导后,可高效表达重组的小鼠酪氨酸酶,表达量约占菌体总蛋白质的2 5 % ,表达产物为包涵体     

Markedness and its Interpretation in Language Use

Language markedness is a common phenomenon in languages, and is reflected from hearing, vision and sense, i.e. the variation in the three aspects such as phonology, morphology and semantics. This paper focuses on the interpretation of markedness in language use following the three perspectives, i.e. pragmatic interpretation, psychological interpretation and cognitive interpretation, with an aim to define the function of markedness.     

Features of Women's language

Language is a means of verbal communication. People use language to communicate with each other. In the society, no two speakers are exactly alike in the way of speaking. Some differences are due to age, gender, statue and personality. Above all, gender is one of the obvious reasons. The writer of this paper tries to describe the features of women＇s language from these perspectives： pronunciation, intonation, diction, subjects, grammar and discourse. From the discussion of the features of women＇s language, more attention should be paid to language use in social context. What＇s more, the linguistic phenomena in a speaking community can be understood more thoroughly.     

低渗透非均质砂岩油藏启动压力梯度研究

理论推导与室内实验相结合,建立了低渗透非均质砂岩油藏启动压力梯度确定方法。首先借助油藏流场与电场相似的原理,推导了非均质砂岩油藏启动压力梯度计算公式。其次基于稳定流实验方法,建立了非均质砂岩油藏启动压力梯度测试方法。结果表明:低渗透非均质砂岩油藏的启动压力梯度确定遵循两个等效原则。平面非均质油藏的启动压力梯度等于各级渗透率段的启动压力梯度关于长度的加权平均;纵向非均质油藏的启动压力梯度等于各渗透率层的启动压力梯度关于渗透率与渗流面积乘积的加权平均。研究成果可用于有效指导低渗透非均质砂岩油藏的合理井距确定,促进该类油藏的高效开发。
