Einige neue Trennungsmethoden und ihre Anwendung auf biochemische und organisch-chemische Probleme

Summary A survey is given of some recent progress, particularly in the author's own laboratory, of methods for the separation of macromolecules and particles of biological origin, as viruses, bacteriophage, microsomes, cell fragments, bacteria and whole cells. Special attention is given to recent developments in zone electrophoresis, protein chromatography, gel filtration and the new partition methods for separation of particles and macromolecules (Albertsson). A number of examples are given to illustrate the various applications. Special emphasis is laid upon the new possibilities for biochemical-genetic studies by application of high-resolution separation methods, and the significance of particle separation methods as applied to fragments of biological structures in providing a tool for structure investigations on a macromolecular level—a field which also should be of considerable interest from a chemical point of view.

Summary Histochemical investigations on enzyme activities in the transitional epithelium showed positive reactions with succinic dehydrogenase, esterase, alkaline phosphatase, acid phosphatase and amylophosphorylase (incl. branching factor). Negative results were drawn from cholinesterase, 5-nucleotidase, glucose-6-phosphatase, and carbonic anhydrase.

Zusammenfassung Das Wachstum der umbrellaren Gewebe bei der HydromedusePhialidium hemisphaericum wurde durch lokale Anfärbung mit dem Vitalfarbstoff Bismarckbraun untersucht. Die Wachstumsregion, welche ex- und subumbrellare Gewebe bildet, liegt am Rande der Medusenglocke in der Nähe des Ringkanales. Gewebewanderungen konnten keine festgestellt werden. Die möglichen Folgen der unterschiedlichen Wachstumsvorgänge bei Polyp und Meduse werden diskutiert.

Summary It appears that in thymectomized CF 1 mice the number of hemolytic plaque-forming cells is very diminished, whereas the number of precursor cells is normal. This finding suggests that in thymectomized mice precursor cells the mitosis number is lower.

Summary Comparative investigations were carried out in rats to determine the effect of butylbiguanide and of insulin on the oxidation of uniform¹⁴C-labelled glucose. The administration of equally hypoglycaemic active doses of butylbiguanide (250 mg per kg orally) or of insulin (2 IU s.c.), together with 0.1 µC¹⁴C-glucose, caused similar increases in the elimination rate of¹⁴CO₂.

Summary A quantitative and qualitative analysis of the carboxylic esterases of normal granulocytes, normal lymphocytes and leukemic cells was performed. In the myeloid leukemic cells and in the lymphatic leukemic cells, the esterase activity was significantly increased. By means of electrophoretic fractionation, four zones with esterase activity were detected in normal granulocytes and lymphocytes; the electrophoretic pattern of the leukemic cells was similar to that of the normal cells.

Summary Many dark-coloured species ofAphididae contain a colouring matter (protoaphin) of unusual structure in the haemolymph. After death of the insect this undergoes a remarkable series of changes by enzymic action yielding successively the pigments xanthoaphin, chrysoaphin and erythroaphin. Recent investigations leading to the complete structural elucidation of all these pigments is described.

Les cellules mammotropes et somatotropes de l'antehypophyse chez la souris C3H porteuse de tumeur mammaire spontanée. Etude quantitative au microscope électronique

Summary Pituitaries of mammary tumor-bearing mice (C3H) were examined by quantitative electron microscopy. The results indicate that considerable modifications occur in mammotropic and somatotropic cells. Both types show an increase of the surface of the endoplasmic reticulum and a decrease of the volume of secretory granules (in percent of cytoplasmic volume), suggesting a heightened secretory activity of these cells during mammary carcinogenesis.
