Fluorescence in situ hybridization (FISH) was applied to somatic chromosomes preparations of Oryza officinalis Wall. (CC), O. sativa L. (AA)xO. officinalis F1 hybrid (AC), backcross progenies BC1 (AAC and ACC), O. latifolia Desv. (CCDD), O. alta Swallen (CCDD) and O. punctata Kotschy (BBCC) with a labelled probe of Cot-1 DNA from O. officinalis. In O. officinalis, the homologous chromosomes showed similar signal bands probed by Cot-1 DNA and karyotype analysis was conducted based on the band patterns. Using no blocking DNA, the probe identified the chromosomes of C genome clearly, but detected few signals on chromosomes of A genome in the F1 hybrid and two backcross progenies of BC1. It is obvious that the highly and moderately repetitive DNA sequences were considerably different between C and A genomes. The chromosomes of C genome were also discriminated from the chromosomes of Dand B-genome in the tetraploid species O. latifolia, O. alta and O. punctata by Cot-1 DNA-FISH. Comparison of the fluorescence intensity on the chromosomes of B, C and D genomes in O. latifolia, O. alta, and O. punctata indicated that the differentiations between C and D genomes are less than that between C and B genomes. The relationship between C and D genomes in O. alta is closer than that of C and D genomes in O. latifolia. This would be one of the causes for the fact that both the genomes are of the same karyotype (CCDD) but belong to different species. The above results showed that the Cot-1 DNA had a high specificity of genome and species. In this paper, the origin of allotetraploid in genus Oryza is also discussed.  相似文献

The molecular phylogenetic trees of 10 species representing 6 genera of the family Rhizophoraceae have been constructed using the sequences of chloroplast genesmatK andrbcL as well as the ITS regions of nuclear ribosomal DNA. Relative-rate tests between lineages in these phylogenetic trees have been performed. On the basis of the results of the relative-rate tests and related molecular evolutionary rate data, the divergence times between the lineages are estimated as follows: (i) the first divergence time in these genera is 132.25 million years ago (mya); (ii) the average divergence time between two tribes, i.e. inland Legnotideae (exceptCarallia brachiata) and mangrove Rhizophoreae, is 64.13 mya; and (iii) the average divergence time between two inland species,C. garciniaefolia andC. pectinifolia, is 19.92 mya.  相似文献

目的 利用高效液相色谱法探究杜仲、厚朴和黄柏三种皮类药材不同贮藏期药效成分的含量变化。方法 分别采用回流、超声和闪式三种方式对同批贮存一年以及贮存五年的杜仲、厚朴和黄柏三种皮类药材进行提取,并利用高效液相色谱法对提取液中有效成分进行定量分析。结果 与贮藏期一年的药材相比,经过5年的贮存,三种皮类药材的药效物质含量均随着贮藏期延长或多或少的降低,其中杜仲皮中桃叶珊瑚苷的含量减少70%以上;厚朴皮中厚朴酚及和厚朴酚的含量减少1%以下;黄柏皮中盐酸小檗碱的含量减少10%以上。结论 三种皮类药材的药效物质在贮藏期变化总体有相似的规律,贮藏时间对杜仲皮皮中桃叶珊瑚苷的含量影响较大,对厚朴皮中厚朴酚与和厚朴酚的含量影响最小,而对黄柏皮中盐酸小檗碱的含量影响较小。研究结果为三种皮类药材的质量控制提供了科学依据。  相似文献

对中国鲿科鱼类系统分类学研究中发现,凹尾拟鲿为一不同于细体拟鲿(P. pratti)的有效种,仅分布于长江上游。凹尾拟鲿胸刺前缘光滑无锯齿,颌须较短,后伸不超过胸鳍起点,本属中同时具有这两个特征的还包括18个有效种。在这18个物种中,乌苏里拟鲿(P. ussuriensis)、长脂拟鲿(P. adiposalis)、短尾拟鲿(P. brevicaudatus)和越南拟鲿(P. kyphus)与凹尾拟鲿一样,具有中等分叉的尾鳍。凹尾拟鲿与乌苏里拟鲿和长脂拟鲿的区别在于游离脊椎骨数目和背刺长短等性状上,与短尾拟鲿和越南拟鲿在背刺后缘程度、枕骨棘与项背骨间的间距上存在差异。凹尾拟鲿与细体拟鲿在形态上最为相似,但在体高、背鳍前长、脂鳍高及尾柄高等性状上存在差别。同时,在颅骨、犁骨、项背骨及腰带等骨骼特征上也存在较大差异。  相似文献

The ATP in roots and xylem sap of two woody plant species, Platycladus orientalis and Acacia auriculiformis, subjected to rapid and slow soil drying has been determined employing firefly luciferase ATP assay method (sensitivity is at 10-12 mol ATP L-1). The ATP levels in the two species were 1.6 nmol·gDW-1 and 0.6 nmol·gDW-1 in roots, and 5.6 μmol·m-3 and 8 μmol ·m-3 in xylem sap, respectively. When plants of P. orientalis and A. auriculiformis were subjected to rapid soil drying, respectively, as soil water content (SWC) decreased from the normal level ( 0. 25 g ·gDW-1) to 0. 02 and 0. 06 g ·gDW-1, separately, plant water potential ( ) dropped to -4 and -3.2 MPa, differently, the ATP in roots decreased 99.7% and 42%, respectively . When the rapidly dried soil was watered for up to 6 d, SWC and were found to recover to their normal levels, but ATP content in roots of P. orientalis and A. auriculiformis recovered by 10% and 23%, respectively. When plants of P. orientalis and A. auriculiformis were subjected to slow soil drying, respectively, as SWC was found to decrease from the normal level to 0.1 and 0.13 g·gDW-1, separately, dropped to - 2.5 and - 2 MPa, differently, the ATP content in roots decreased 98% and 38%, respectively. When the slowly dried soil was watered for up to 8 d, ATP levels in roots of P. orientalis and A, auriculiformis recovered by 30% and 24%, respectively. However, ATP concentrations in xylem sap were not directly influenced by either SWC or . The plot dots of xylem sap ATP concentration versus root ATP content were found to be distributed diagonally. It is concluded that ATP in roots is more sensitive in response to SWC changes than that to and root-sourced ATP is a source of that in xylem sap. When plants are subjected to slow soil drying, the ATP levels in both root and xylem sap are higher than that to rapid soil drying.  相似文献

Partial sequences of mitochondrial 16S rRNA gene were obtained by PCR amplification for comparisons among nine species of glyptosternoid fishes and six species of non-glyptosternoids representing 10 sisorid genera. There are compositional biases in the A-rich unpaired regions and G-rich paired regions. A-G transitions are primarily responsible for the Ts/Tv bias in unpaired regions. The overall substitution rate in unpaired regions is almost two times higher than that in the paired regions. Saturation plots at comparable levels of sequence divergence demonstrate no saturation effects. Phylogenetic analyses using both maximum likelihood and Bayesian methods support the monophyly of Sisoridae. Chinese sisorid catfishes are composed of two majorlineages, one represented by (Gagata (Bagarius, Glyptothorax)) and the other by “glyptosternoids Pseudecheneis“. The glyptosternoids may not be a monophyletic group. A previous hypothesis referring to Pseudecheneis as the sister group of monophyletic glyptosternoids, based on morphological evidence, is not supported by the molecular data. Pseudecheneis is shown to be a sister taxon of Glaridoglanis. Pareuchiloglanis might be paraphyletic with Pseudexostoma and Euchiloglanis. Our results also support the hypothesis that Pareuchiloglanis anteanalis might be considered as the synonyms of Pareuchiloglanis sinensis, and genus Euchiloglanis might have only one valid species, Euchiloglanis davidi.  相似文献

为了解气候变化情景下广东省红豆属(Ormosia)潜在适生区的变化,探究影响红豆属植物潜在适生区的主导环境因子,本研究基于MaxEnt模型和ArcGIS 10.8软件,利用17种红豆属植物的分布数据及22个环境变量数据,结合未来共享社会经济路径(SSP126、SSP585)的气候情景,分别预测红豆属植物在当前和未来气候场景下的潜在适生区分布情况。结果表明:(1)适生区预测模型的评价效果良好,受试者工作特征曲线(Receiver Operating Characteristic curve, ROC曲线)下面积(AUC平均值)均为0.8-1.0。(2)在当前气候情景下,大面积(占广东省面积的40%-100%)适生的红豆属植物有茸荚红豆(Ormosia pachycarpa)等3种;中等面积(占20%-40%)适生的有薄毛茸荚红豆(O.pachycarpa var.tenuis)等6种;小面积(占5%-20%)适生的有韧荚红豆(O.indurata)等4种;极小面积(占0%-5%)适生的有凹叶红豆(O.emarginata)等4种。其中面积占比最大的是茸荚红豆,总适生面积为15.228 8...  相似文献

The substructure of pollen exine inCedrus deodara (Roxb.) Loud. andMetasequoia glyptostroboides Hu et Cheng has been examined with an atomic force microscope (AFM). The results indicate that the exine substructure units containing sporopollenin in two species are similar in shape, which are granular, but slightly different in size. InCedrus the substructure unit of pollen exine appears to be 56–99 nm long and 42–74 nm wide, while inMetasequoia it appears to be 81–118 nm long and 43–98 nm wide. It has been observed that the subunits of pollen exine inCedrus arranged tightly to form short-rod-like or spheroidal pollen exine units, several or more than ten of which formed an island-like structure. There are various spaces among these island-like structures which are interconnected to occupy the entire pollen exine. InMetasequoia, the subunits of pollen exine also arranged tightly with a distribution tendency of cluster of 3–10, however, no obvious boundary exists among these clusters. From our results, it is concluded that there is no tendency of helical arrangement for the subunits of pollen exine inCedrus andMetasequoia, and the results support Southworth’ view that subunits of pollen exine are granular shape in lattice structure.  相似文献

Introns and exons of 7 genes (epsilon globin, gamma-1 globin, gamma-2 globin, delta globin, beta globin, Immunoglobulin andprepro-insulin) in primates have been separated out and used to infer phylogeny respectively. For each gene, results based on these two parts have been compared and showed that: (i) the topology of introns is almost consistent with that of exons in each gene, while the branch length of them varies, because of the dierent mutation rate; (ii) there is evidence that the substitution rate of exons would decrease inhominoids, but that of introns would not; (iii) divergence time of orangutan deduced from different genes based on exons is various, while that based on introns is much similar, and consistent with fossil records; (iv) there is a relationship between the G + C content and the substitution rate. When the substitution rate of introns is higher than exons in a gene, the G + C content of introns is less. The above results suggest that introns could provide useful evolutionary information among closety related species.  相似文献

目的研究厚朴叶的毒性及与厚朴皮、厚朴花的毒性比较,为厚朴叶的开发利用提供实验依据。方法通过急性毒性实验,小鼠骨髓微核实验,小鼠精子畸形实验,大鼠30 d喂养实验,来研究厚朴叶毒性和进行毒性比较。结果在急性毒性实验中,各给药组小鼠无死亡;小鼠骨髓微核实验中和精子畸形实验中,各给药组与正常组无统计学差异。大鼠30 d喂养实验中,厚朴叶对大鼠的食物利用率、血常规、肾功能都有所影响,也影响肝、卵巢、睾丸的脏体系数;在对大鼠各脏器病理组织学检查中未见明显异常;厚朴叶与厚朴皮、厚朴花的慢性毒性差异为后两者不影响血常规。结论厚朴叶无急性毒性,无致骨髓突变毒性,无致精子畸形毒性;厚朴叶有慢性毒性作用,表现在影响食物利用率、血常规、肾功能、器官发育;厚朴叶与厚朴皮、厚朴花的毒性差异表现在慢性毒性中影响血常规,其它无差异。  相似文献

小野1A是由小粒野生稻(Oryza minuta Presl.)中天然不育株与Y58S/内香B杂种后代优秀单株经多代回交转育而成。由于该雄性不育细胞质来自小粒野生稻,称为XY型雄性不育细胞质。恢保关系鉴定结果表明,XY型细胞质雄性不育系与野败型(WA)、印尼水田谷型(ID)、K型和红莲型(HL)细胞质雄性不育系的恢保关系不同,WA型、ID型、K型和HL型的保持系和恢复系中部分可以作为XY型雄性不育细胞质的保持系,部分可以作为XY型雄性不育细胞质的恢复系。XY型雄性不育细胞质突破了WA型、ID型、K型和HL型等雄性不育细胞质的恢保关系,扩大了保持系育种范围,可以用品质优良的栽培品种培育出高标准优质米不育系。由此可见,XY型雄性不育细胞质的发掘,不仅丰富了杂交稻细胞质遗传多样性,而且为培育高标准优质米不育系、进而实质性提高杂交稻稻米品质开辟了一条崭新的育种途径。  相似文献

Isolation of plant resistance genes is greatly helpful to crop resistance breeding and the insight of resistance mechanism. The cloned plant resistance genes are classified into four classes according to their putative structural domain, of which the majority possesses nucleotide-binding site (NBS) domain that consists of P-loop, kinase2a and kinase3a. The conservation of this domain affords the potential possibility of cloning the plant resistance genes, which is homology-based cloning technique. In the present study, the degenerate oligonucleotide primers were designed according to the tobaccoN andArabidopsis RPS2, and 358 clones were isolated from the genomic DNA of resistance soybean cultivar Kefengl, resistant to soybean mosaic virus, and 4 open-reading NBS analogs were finally characterized and designated asKNBS1, KNBS2, KNBS3 andKNBS4. Southern hybridization suggested that they were present with multicopy in the soybean genome;KNBS4 was mapped to F linkage group andKNBS2 co-located J linkage group with the SCAR marker ofRsa resistant to soybean mosaic virus by RFLP analysis. Northern analysis suggested thatKNBS2- related sequence was low and constitutively expressed in the root, stem and leaves of soybean. The detailed characterization of NBS analogs is very helpful to ultimately cloning the soybean resistance gene.  相似文献

Having reviewed the major classification systems proposed by various scholars across the world, it is found thatindica andjaponica underO. sativa L. are two major directions thoroughly differentiated from the Asian cultivated rice, forming the framework of the classification structure. A system withindica andjaponica as the only two subspecies is therefore reiterated. Orientational domains at room temperature in orthorhombic perovskite La0.33Ca0.67MnO3 were studied by group-theory and observed systematically using transmission electron microscopy. There are six orientational variants (A, A′, B, B′, C andC′) in orthorhombic perovskite La0.33Ca0.67MnO3. Their orthorhombicb o directions are parallel to thea p,b p andc p directions of the cubic prototypic perovskite, respectively. Using the primers which specially amplify the conservative motif of Human SRY gene, we studied the PCR amplification of Sox genes in genomic DNA of two species of mud loach:Misgurnus anguillicaudatus andParamisgurnus dabryanus. Four bands with the length of 200,550,940 and 1000 bp respectively, were presented in the PCR products ofMisgurnus anguillicaudatus. Three bands with the length of 200,550 and 900 bp were presented in that ofParamisgurnus dabryanus. Southern blotting results indicated that the 200 and 550 bp bands are specially positive. Epinephelus moara and E. bruneus are closely related species in the genus Epinephelus (Perciformes, Serranidae). Their morphological similarity, changing color pattern at different stages and living conditions make them difficult to be differentiated. To identify these two species, an improved nest-tetra-primer-specific PCR assay was developed. Three specific molecular markers, the control region NC1 (394 bp), species-specific internal region ND2-M (268 bp) and ND2-B (122 bp), were identified in the mitochondrial ND2 gene from these two grouper species. Five markers were also discovered in the ITS1 regions of their nuclear ribosomal DNA, which were the control regions NC2 (588 bp) and NC3 (563 bp), and species-specific internal regions rDNA-M (426 bp), ITS1-M (488 bp) and ITS1-B (304 bp). This method provided a highly specific, precise, reliable and rapid molecular marker technique to discriminate between the two grouper species, as well as a new way of DNA identification to differentiate closely related species in fishes.  相似文献

Origin of summer monsoon rainfall identified by δ18O in precipitation   总被引:2,自引:0,他引:2  
A negative correlation between δ^18O in monsoon precipitation and f, the ratio of precipitable water in monsoon region to that in water source area, is hypothesized. Using the Rayleigh model, a new method for identifying origin of summer monsoon rainfall is developed based on the hypothesis. In order to validate the method, the isotopic data at New Delhi, a typical station in the southwest monsoon region, and Hong Kong, a typical station in the southeast monsoon region, were collected and analyzed for case studies. The case studies indicate that the water source areas of the monsoon rairdall at the two stations identified by the method are accordant with the general atmosphere circulation patterns. The method developed in this paper is significantly important for tracing the origin of summer monsoon precipitation.  相似文献

The seed-specific promoter and transit peptide were amplified and fused to the three genesphbA, phbB andphbC encoding PHB synthetic enzymes, respectively. Seed-specific expression vectors pSCB containingphbC andphbB, and pSCAB containingphbC, phbB andphbA, were constructed by introducing the genes with promoter and peptide into the binary vector pBI101. 目的探讨厚朴皮、叶、花对小鼠胃排空及肠推进运动的影响。方法采用胃排空、小肠推进运动实验法,观察高、低剂量厚朴皮、叶、花水提物对小鼠胃排空及小肠推进运动的影响。结果厚朴干皮、根皮、枝皮、叶水提物高、低剂量均能促进胃排空和肠推进运动,与蒸馏水对照组比较差异显著(P0.01,P0.05);厚朴根皮、枝皮、叶水提物组与相对应剂量厚朴干皮水提物组比较,差异无统计学意义(P0.05)。结论厚朴干皮、根皮、枝皮、叶水提物均具有促进胃排空、肠推进运动的作用,且厚朴根皮、枝皮、叶可代替厚朴干皮用于胃肠促动。  相似文献
