用PCR方法获得编码大肠杆菌核酸内切酶Ⅳ的基因nfo,将其克隆至具有T7启动子的表达载体pET23a中,转化不同的宿主菌。经过IPTG的诱导,在宿主BL21(DE3)LysE菌中获得表达,表达量占菌体总蛋白量的9.6%。通过同位素比放测定显示表达产物具有AP核酸内切酶活性  相似文献

人鼻病毒3C蛋白酶具有高特异性,且在低温条件下具有较高酶活,目前已被商品化.在大肠杆菌中利用麦芽糖结合蛋白(MBP)作为融合标签可促进人鼻病毒3C蛋白酶的可溶性表达.将人鼻病毒3C蛋白酶的DNA编码序列克隆到pRK792载体上,然后导入到大肠杆菌BL21(DE3),进行诱导表达,获得MBP-LEVLFQGP-6x His-人鼻病毒3C蛋白酶融合蛋白,随后通过Ni柱亲和纯化获得目的蛋白.用该产物切割纯化的MBP-LEVLFQGP-6x His-木聚糖酶融合蛋白,可获得木聚糖酶并用底物平板检测其活性.实验结果表明人鼻病毒3C蛋白酶能够在大肠杆菌中表达,并且能特异性地在其底物识别序列进行切割,从而移除MBP标签,获得仅带有6x His的人鼻病毒3C蛋白酶.该酶可以特异性地去除木聚糖酶融合蛋白中的MBP标签,获得有活性的木聚糖酶.利用MBP标签能够促进人鼻病毒3C蛋白酶可溶性表达,且人鼻病毒3C蛋白酶的活性不受影响,有利于一些不稳定的蛋白在低温下的标签去除和纯化.  相似文献

铜绿假单胞菌PA0057基因为Ⅳ类未知基因,通过NCBI比对,发现PA0057基因编码的蛋白与金属β-内酰胺酶同源性为96%,因此推测PA0057蛋白可能具有金属β内酰胺酶活性.提取铜绿假单胞菌PAOI基因组DNA,利用设计的引物通过PCR方法扩增得到PA0057基因,将其克隆至克隆载体pGEM-T,并转化人大肠杆菌DH5α;而后将其克隆至表达载体pET28a中,转化人大肠杆菌BL21(DE3)中,IPTG诱导表达,并通过Ni-NTA亲和柱进行纯化,纯化后蛋白超滤浓缩得高浓度蛋白;通过药敏纸片检测PA0057蛋白活性.  相似文献

目的筛选出脂肪酶活性较高菌株,通过脂肪酶基因克隆技术获得高表达的脂肪酶,根据酶活曲线确定该酶的最适温度和最适pH.方法采用透明圈法,以三丁酸甘油酯为底物筛选脂肪酶活性较高菌株;通过PCR扩增获得其脂肪酶基因Pseudomonas peli(PP)序列,构建pET-28 a表达载体,并在大肠杆菌Escherichia coli BL21(DE3)中进行异源表达;通过Ni柱将脂肪酶纯化,并根据酶活曲线确定该酶的最适温度和最适pH.结果筛选到一株脂肪酶活性较高的菌株,16S rRNA基因序列比对结果初步鉴定为Pseudomonas peli.PCR扩增得到的基因片段长度为801 bp,其序列与Pseudomonas peli中的脂肪酶基因序列相似性达到100%,PP基因在大肠杆菌中异源表达蛋白的分子量约29 KD,该蛋白在55℃、pH 7.0时活性最高.结论通过基因克隆表达后得到的脂肪酶Ni柱纯化后纯度达95%以上,且耐高温,为脂肪酶工业化应用奠定了基础.  相似文献

为获得具有催化活性的糖基转移酶纯酶,本研究以短小芽孢杆菌基因组DNA为模板,利用简并PCR技术扩增到基因(GT-A)全长序列.该序列全长1 287bp、编码423个氨基酸,分子量约为49.2KD.经序列分析,该基因属于糖基转移酶基因.根据GT-A基因开放阅读框序列设计引物,构建了原核表达重组质粒GTA-pet28a,并在大肠杆菌BL21(DE3)中成功诱导出了一个约50kD的融合蛋白.纯化后测定其糖基转移酶活性,与37℃相比,80℃的反应温度其活性能提高3.4倍左右.研究结果表明,该酶是一种具有应用潜力的嗜高温糖基转移酶.  相似文献

研究目的:克隆表达人肠激酶轻链编码基因,以期应用于融合蛋白的切割与纯化.方法:从人的全基因组中扩增出编码人肠激酶轻链的5个外显子基因片段,经过酶切、连接后得到正确编码的人肠激酶轻链完整基因序列;随后,将目的基因片段插入原核表达载体pBV220中,构建成融合型表达载体pBV—EKL,转化大肠杆菌BL21(DE3),调节温度至42℃诱导重组蛋白表达;通过镍亲和层析纯化得到重组蛋白.结果:所表达的重组蛋白经SDS—PAGE分析,相对表观分子质量约为31kDa,表达量占菌体总可溶蛋白量的46%;通过镍亲和层析纯化得到的重组蛋白经复性后显示出具有催化切割活性.结论:成功构建了能大量表达重组人肠激酶轻链的菌株,为进一步进行重组人肠激酶活性的研究及其在生产和医学方面的应用研究奠定了基础.  相似文献

为获得高分辨率的雄激素受体与其响应元件的复合物晶体结构,为前列腺癌的治疗提供药物靶点设计的理论基础.本研究构建了AR-DBD的原核表达载体,在0.5 mmol/L IPTG,16℃条件下诱导24 h表达该蛋白.通过与AR响应元件寡核酸的结合优化蛋白纯化条件,使AR-DBD蛋白易于纯化.最终,蛋白核酸复合物晶体呈现出边缘清晰,短棒状的立体结构,本研究制备的蛋白核酸复合物晶体可直接用于X射线衍射分析.  相似文献

SAMHD1蛋白是一种天然免疫限制因子,利用其d NTP水解酶活性或核酸酶活性抑制病毒的复制,具有广谱抗病毒功能.利用生物信息学分析该蛋白家族序列保守性,并借助HPLC层析色谱分析与酶活性测定和酶标仪方法,对人源、猪源和鸡源三物种SAMHD1蛋白的酶活性进行了比较.比对发现SAMHD1蛋白在活性空腔、变构位点及磷酸化位点等处序列上具有高度的保守性;而且发现猪源和鸡源SAMHD1蛋白同样具有d NTP水解酶和核酸酶活性;3种蛋白中,人源SAMHD1蛋白d NTP水解酶活性最高,猪源蛋白核酸酶活性最低.这一结果为研究SAMHD1蛋白家族的结构功能关系及为畜牧业优良品种的开发提供启示.  相似文献

Language is a means of verbal communication. People use language to communicate with each other. In the society, no two speakers are exactly alike in the way of speaking. Some differences are due to age, gender, statue and personality. Above all, gender is one of the obvious reasons. The writer of this paper tries to describe the features of women's language from these perspectives: pronunciation, intonation, diction, subjects, grammar and discourse. From the discussion of the features of women's language, more attention should be paid to language use in social context. The Williston Basin is a significant petroleum province, containing oil production zones that include the Middle Cambrian to Lower Ordovician, Upper Ordovician, Middle Devonian, Upper Devonian and Mississippian and within the Jurassic and Cretaceous. The oils of the Williston Basin exhibit a wide range of geochemical characteristics defined as "oil families", although the geochemical signature of the Cambrian Deadwood Formation and Lower Ordovician Winnipeg reservoired oils does not match any "oil family". Despite their close stratigraphic proximity, it is evident that the oils of the Lower Palaeozoic within the Williston Basin are distinct. This suggests the presence of a new "oil family" within the Williston Basin. Diagnostic geochemical signatures occur in the gasoline range chromatograms, within saturate fraction gas chromatograms and biomarker fingerprints. However, some of the established criteria and cross-plots that are currently used to segregate oils into distinct genetic families within the basin do not always meet with success, particularly when applied to the Lower Palaeozoic oils of the Deadwood and Winnipeg Formation.  相似文献

理论推导与室内实验相结合,建立了低渗透非均质砂岩油藏启动压力梯度确定方法。首先借助油藏流场与电场相似的原理,推导了非均质砂岩油藏启动压力梯度计算公式。其次基于稳定流实验方法,建立了非均质砂岩油藏启动压力梯度测试方法。结果表明:低渗透非均质砂岩油藏的启动压力梯度确定遵循两个等效原则。平面非均质油藏的启动压力梯度等于各级渗透率段的启动压力梯度关于长度的加权平均;纵向非均质油藏的启动压力梯度等于各渗透率层的启动压力梯度关于渗透率与渗流面积乘积的加权平均。研究成果可用于有效指导低渗透非均质砂岩油藏的合理井距确定,促进该类油藏的高效开发。  相似文献
