It goes without saying that we are stuck with the Universe we have. Nevertheless, we would like to go beyond simply describing our observed Universe, and try to understand why it is that way rather than some other way. When considering both the state in which we find our current Universe, and the laws of physics it obeys, we discover features that seem remarkably unnatural to us. Although feast and famine cycles illustrate that remodelling of adipose tissue in response to fluctuations in nutrient availability is essential for maintaining metabolic homeostasis, the underlying mechanisms remain poorly understood. Here we identify fibroblast growth factor 1 (FGF1) as a critical transducer in this process in mice, and link its regulation to the nuclear receptor PPARγ (peroxisome proliferator activated receptor γ), which is the adipocyte master regulator and the target of the thiazolidinedione class of insulin sensitizing drugs. FGF1 is the prototype of the 22-member FGF family of proteins and has been implicated in a range of physiological processes, including development, wound healing and cardiovascular changes. Surprisingly, FGF1 knockout mice display no significant phenotype under standard laboratory conditions. We show that FGF1 is highly induced in adipose tissue in response to a high-fat diet and that mice lacking FGF1 develop an aggressive diabetic phenotype coupled to aberrant adipose expansion when challenged with a high-fat diet. Further analysis of adipose depots in FGF1-deficient mice revealed multiple histopathologies in the vasculature network, an accentuated inflammatory response, aberrant adipocyte size distribution and ectopic expression of pancreatic lipases. On withdrawal of the high-fat diet, this inflamed adipose tissue fails to properly resolve, resulting in extensive fat necrosis. In terms of mechanisms, we show that adipose induction of FGF1 in the fed state is regulated by PPARγ acting through an evolutionarily conserved promoter proximal PPAR response element within the FGF1 gene. The discovery of a phenotype for the FGF1 knockout mouse establishes the PPARγ–FGF1 axis as critical for maintaining metabolic homeostasis and insulin sensitization.  相似文献

The main-channel of the lower Yellow River is the main path of water and sediment transportation. In this paper, we put forth a suggestion that the bank-full discharge of the main-channel in the lower Yellow River should be no less than 4000 m^3/s, which is based on the demand from effective sediment transportation (or riverbed scouring), the demand of flood control on morphology of the cross section of the main-channel, and the possible riverbed-forming force of future flood. The forming of the main-channel is the process for the flood force to overcome the resistance from the riverbed. The forming of the main-channel configuration comes from the competition between riverbed-forming force of flood and resistance from the riverbed. Regarding the flood with medium or less sediment content and without floodplain inundated, the main-channel forming mechanism can be simplified as the relationship between the change of cross section area and W^αQ^β. For forming same main-channel, the needed flood force W^0.32Q^0.37 is definite, i.e., when the discharge is large, the flood volume can be smaller; if some flood is able to scour the main-channel, its force W^0.32Q^0.37 should be larger than 41. Not all floods that inundate floodplain have the function of silting in floodplain and scouring main-channel. Regarding flood with medium or less sediment content, only when the flood discharge is over 1.5 times of the relevant bankfull discharge, obvious scouring effect can occur in the main-channel.  相似文献

The anomaly of the recent increase in Atlantic major hurricane activity (MHA) is controversial. From a reconstruction of past MHA, Nyberg et al. conclude that the present activity is not unusual by comparison with that of the past 270 years. However, here I estimate the uncertainty of average MHA in the hurricane record before 1945 and show that the reconstruction of Nyberg et al. differs strongly from that record, and probably overestimates past MHA. Owing to this and further reasons, I question whether their reconstruction provides an accurate basis for conclusions about past MHA.  相似文献
