A cell line called HCxPy was obtained in vitro by transformation of dissociated hamster brain cell cultures by polyoma virus. The first foci of transformed cells became evident 90 to 120 days after viral infection. This cell line is now at the 46th passage. The cells appear tumorigenic for hamsters after subcutaneous and intracerebral injection. They carry the polyoma virus T and cell surface antigens. Good evidence for astrocytic differentiation can be found by morphological examination of the tumours and of the cultured cells.  相似文献

Using primary cultures originating from 14 day old fetal Hamster brain, we have obtained a cell line with glial morphology. These cell remain non transplantable during the first year of in vitro culture, but undergo spontaneous transformation during the second year. Following prolonged contact of the glial-like cells with 25 to 50 microgram/ml of methylnitrosourea (MNU), a malignant transformation is observed 5 months after the treatment. The lowest concentrations of (MNU) do not cause malignant transformation, but seem to inhibit (or postpone) the spontaneous transformation. After MNU treatment, cells retain their glial nature.  相似文献

Normal Hamster lung cells from an established line were treated with 1-500 microgram . ml-1 2-chlorobutadiene. Those treated with 1 microgram . ml-1 of the compound showed malignant transformations 14 weeks after treatment. The treatment with higher concentrations did not accelerate the transformation process.  相似文献

Tumors have been induced in hydrocortisone treated Mice innoculated with wild-type polyoma transformed Hamster cells. The filtrate from these tumors infects Mouse embryo cells and induces the production of polyoma virus. The polyoma virus has been characterised in the infected cells with anti-polyoma capsid serum.  相似文献

The transformation of Rous-Sarcoma-Virus-infected synchronized Chicken cells is prevented when 5-bromodeoxyuridine is added for 3 hrs. at time of the peak of the first S phase following infection. Virus progeny is normally released. The inhibitory effect of BrdU is reversed by simultaneous addition of thymidine, but not by that of deoxyuridine. Hence, the inhibition of the transformation seems to result from the incorporation of the analogue into the nuclear DNA.  相似文献

Résumé On présente une étude comparée sur la production d'interféron par les cellules non-transformées et tumorales de hamster ainsi que par les cellules de singe, après leur traîtement par le virus Sendai inactivé. Les cellules tumorales ne produisent pas l'interféron sous l'action du virus sendai. Les cellules non-transformées cessent de produire ou de libérer l'interféron, dans leur milieu de culture, en moins de 24 h après l'enlèvement du virus Sendai de ce milieu.

Morphological transformation occurred on addition of, 4NQO, but not in control cultures. The hamster cells transformed by the Rous Sarcoma Virus (V.S.R.) evidenced surface antigenic alterations that were detected by a method of cellular mediated immunocytotoxicity. Immune hamster lymphocytes were added to tritiated proline prelabeled target cells. These lymphocytes were able to lyse specifically the V.S.R. transformed cells.  相似文献

The action of alloxan was studied in vitro, on different categories of normal or cancerous cells. At concentrations of 250 and 350 gamma/ml, alloxan does not significantly inhibit the growth of normal cells, whereas it does inhibit the growth of cancerous cells. Furthermore, alloxan inhibits infection and transformation of chicken-embryo fibroblasts infected by the Rous virus.  相似文献
