研究常氧下微血管内皮细胞(MVEC)与肺动脉平滑肌细胞(PASMC)增殖的相互调节。先滤膜培养大鼠肺微血管内皮细胞(LMVEC)后,再与PASMC复合培养,采用流式细胞仪检测PASMC周期的变化。复合培养后,正常情况下使PASMC的G1期细胞数减少,S+G2/M期细胞数增多,即促进细胞生长;使LMVEC田C的G1期细胞数增多,S+G2/M期细胞数减少,即抑制细胞生长。可见,MVEC与PASMC复合培养后,常氧下促进PASMC细胞生长,抑制LMVEC的增生,存在相互作用。  相似文献

着重探讨低氧对食管癌ECa9706细胞生长及顺铂敏感性的影响.ECa9706细胞分别培养于顺铂干预的常氧和低氧(1%O2)环境下,倒置显微镜观察细胞形态,MTT法检测细胞活力,流式细胞法检测细胞凋亡和细胞周期.结果显示:与常氧环境下培养相比,低氧下细胞形态变长、变大,细胞自第7d后生长缓慢.常氧下顺铂作用细胞48h的IC50为(1.180±0.056)μM,低氧下为(2.675±0.063)μM.培养48h后,常氧对照组及常氧高浓度顺铂组(1.18μM)阻滞细胞周期于G1期,低氧下各组细胞周期阻滞于S期.低氧导致顺铂诱导的细胞凋亡率较常氧下降低.表明:低氧环境影响了细胞的形态、增殖及细胞周期,低氧下细胞对顺铂的细胞毒作用敏感性降低.  相似文献

以癌细胞诱导分化物维甲酸为平行对照.应用中药有效成份熊去氧胆酸处理人成骨肉瘤Mp63细胞.观察比较熊去氧胆酸及其与维甲酸的组合对Mp63细胞形态、细胞周期及分化的影响.实验结果显示.经熊去氧胆酸、维甲酸及其组合联合处理之后.MG63细胞体积增大.核质比例减少.形态趋于规则.细胞周期发生G0/G1期阻滞.G0/G1期细胞比例显增加.而S期和G2/M期细胞比例则明显下降.细胞终末分化标志物Ⅰ型胶原表达明显增强,骨结节形成数目增多.且结节结构更为典型.其中,熊去氧胆酸和维甲酸联合组对Mp63细胞周期的阻滞及细胞终末分化指标Ⅰ型胶原的表达、骨结节形成的诱导均强于单独处理组.结果表明.熊去氧胆酸具有与维甲酸相近似的诱导人成骨肉瘤Mp63细胞分化的作用.并且两组合具有协同诱导成骨肉瘤细胞分化的效果.  相似文献

对人工培养的虫草菌菌丝体提取物进行了体外抗肿瘤活性及其作用机理的研究.依次用石油醚、乙酸乙酯、乙醇和水提取人工培养的虫草菌菌丝体,分别得到石油醚、乙酸乙酯、乙醇和水提取物.采用MTT比色法检测发现乙酸乙酯提取物显著抑制类前骨髓白血病细胞HL-60的增殖活性,半数抑制质量浓度(IC50)约为25μg/mL.并采用流式细胞术和Western blotting等方法进一步研究虫草菌乙酸乙酯提取物抑制HL-60细胞生长的作用机理.用流式细胞仪检测HL-60细胞DNA含量发现,25μg/mL虫草菌乙酸乙酯提取物作用12~2Ah后,S期细胞减少,而G2/M期细胞增多,表明细胞周期中G2/M检验点被阻滞.Western blotting分析结果表明,在虫草菌乙酸乙酯提取物作用后,HL-60细胞中G2/M检验点相关周期蛋白p34^cdc2表达量降低.  相似文献

观察番茄红素对体外培养的雌激素受体阳性(ER+)细胞MCF-7的存活率、细胞周期及凋亡的影响。采用MTT法和H3-TdR掺入法观察番茄红素对MCF-7细胞增殖的影响;流式细胞仪观察同步化的细胞经番茄红素作用后细胞周期及凋亡的变化。番茄红素抑制MCF-7细胞的增殖和DNA合成,具有剂量效应关系,随着时间延长,抑制作用增强,最大抑制率为52.6%。流式细胞仪结果显示,番茄红素作用24h后,MCF-7细胞周期各相发生变化,G0/G1期细胞增多,而S期和G2/M期细胞减少,但未诱发其凋亡。番茄红素通过阻滞MCF-7细胞于G1期而抑制该细胞的增殖。  相似文献

分别用Sub-Gl法和PA法对人类早幼粒细胞急性白血病细胞株(HL-60)的自发凋亡进行检测,发现HL-60细胞自发凋亡时,在DNA直谆图上,G1峰前出现亚G1峰(Sub-Gl峰),G1期细胞增加,S期和G2/M期细胞减少。同时G1期细胞发生自发凋亡,而S期,G1期细胞少有凋亡发生。这对研究肿瘤细胞发生凋亡的死亡机制及临床治疗有重要意义。  相似文献

为了探讨牛膝多糖(Achyranthes bidentata polysaccharides,ABPS)结合耐力训练对部分肝切除(Partial hepatectomy,PH)大鼠骨髓增殖的影响,对经过28d游泳训练(Swimming training,ST)、灌胃ABPS的大鼠行2/3部分肝切除手术,观察术后大鼠的运动能力和骨髓细胞的形态和增殖情况.结果表明,游泳训练能够明显延长正常大鼠的游泳运动致力竭时间(P0.01),降低骨髓细胞PCNA(Proliferating cell nuclear antigen)的表达.ABPS结合游泳训练能够明显延长PH大鼠的游泳致力竭时间(和PH+ST对比,P0.01),使骨髓增生程度更活跃,有核细胞数增多,红细胞数量减少,促进了骨髓细胞PCNA的表达(和PH相比,P0.01),使G2/M期和S期细胞均明显减少,G0/G1期增多(P0.05).提示牛膝多糖结合耐力训练增强了大鼠在肝脏部分切除手术后的运动能力,逆转了耐力训练对骨髓细胞增殖的抑制作用.  相似文献

研究了文蛤多肽对体外培养的人肝癌SMMC-7721细胞的抑制作用,结果表明,经5.0μg/mL文蛤多肽处理的体外培养的人肝癌细胞(SMMC-7721)生长缓慢,倍增时间延长,细胞生长抑制率达89.4%;处理后的癌细胞形态发生明显改变,处于G0/G1期的细胞明显增多,而S期和M期细胞减少,出现明显的凋亡峰,凋亡率为22.3%.实验结果表明,文蛤多肽能有效地抑制体外培养肝癌细胞的增殖活动,可通过改变肝癌细胞的形态及细胞周期而明显抑制细胞的增殖.  相似文献

通过TdR(胸腺嘧啶核苷)调控人肝癌细胞株HepG2的细胞周期,运用双向电泳-图像分析-质谱技术研究肝癌细胞株HepG2的G1期、G2/M期全细胞蛋白质组表达的差异,探索参与肝癌细胞株HepG2的G1期、G2/M期调控相关的蛋白。以TdR诱导肝癌细胞株HepG2,分别得到同步化的G1期、G2/M期细胞,流式细胞术检测。采用比较蛋白质组学的研究技术(双向电泳、质谱分析)筛选G1期、G2/M期差异表达的蛋白质。结果流式细胞术检测显示TdR诱导后,分别获得(63.62±2.82)%的G2/M期同步化细胞,(75.24±0.17)%的G1期同步化细胞,通过双向电泳-图像分析-质谱技术得到21个差异表达的蛋白。其中G2/M期表达,G1期未见表达有10种蛋白;存在三倍以上的差异点11个:2种在G2期表达上调,9种在G1期表达上调。说明TdR阻断法能够获得很好的同步化细胞。质谱鉴定得到的这些差异表达的蛋白质涉及到细胞周期调控、DNA结合、转录调控、mRNA剪接、肽链合成起始、折叠以及细胞代谢等方面。  相似文献

Language is a means of verbal communication. People use language to communicate with each other. In the society, no two speakers are exactly alike in the way of speaking. Some differences are due to age, gender, statue and personality. Above all, gender is one of the obvious reasons. The writer of this paper tries to describe the features of women's language from these perspectives: pronunciation, intonation, diction, subjects, grammar and discourse. From the discussion of the features of women's language, more attention should be paid to language use in social context. 理论推导与室内实验相结合,建立了低渗透非均质砂岩油藏启动压力梯度确定方法。首先借助油藏流场与电场相似的原理,推导了非均质砂岩油藏启动压力梯度计算公式。其次基于稳定流实验方法,建立了非均质砂岩油藏启动压力梯度测试方法。结果表明:低渗透非均质砂岩油藏的启动压力梯度确定遵循两个等效原则。平面非均质油藏的启动压力梯度等于各级渗透率段的启动压力梯度关于长度的加权平均;纵向非均质油藏的启动压力梯度等于各渗透率层的启动压力梯度关于渗透率与渗流面积乘积的加权平均。研究成果可用于有效指导低渗透非均质砂岩油藏的合理井距确定,促进该类油藏的高效开发。  相似文献
